Porous titanium alloy is currently widely used in clinical treatment of orthopaedic diseases for its lower elastic modulus and ability to integrate with bone *** the micro-level,cells can respond to different geometries,and at the macro-level,the geometric design of implants will also affect the biological function of *** this study,three kinds of porous scaffolds with square,triangular and circle rod shapes were designed and 3D *** study observed the proliferation and differentiation of MC3T3-E1 cells during surface culture of the three types of *** also evaluated the characteristics of the three scaffolds by means of compression tests and scanning electron microscopy to provide a reference for the design of porous titanium alloy implants for clinical *** trends of cell proliferation and gene expression between the three types of scaffolds were observed after treatment with two *** results show that the square rod porous scaffolds have the best proliferative and osteogenic activities,and these findings may be due to differences in piezo-type mechanosensitive ion channel component 1(Piezo1)and Yes-associated protein(YAP)expression caused by the macro-geometric topography.

The treatment of PML/RARA+acute promyelocytic leukemia(APL)with all-trans-retinoic acid and arsenic trioxide(ATRA/ATO)has been recognized as a model for translational medicine *** an altered microenvironment is a general cancer hallmark,how APL blasts shape their plasma composition is poorly ***,we reported a cross-sectional correlation network to interpret multilayered datasets on clinical parameters,proteomes,and metabolomes of paired plasma samples from patients with APL before or after ATRA/ATO induction *** study revealed the two prominent features of the APL plasma,suggesting a possible involvement of APL blasts in modulating plasma *** was characterized by altered secretory protein and metabolite profiles correlating with heightened proliferation and energy consumption in APL blasts,and the other featured APL plasma-enriched proteins or enzymes catalyzing plasma-altered metabolites that were potential trans-regulatory targets of PML/***,results indicated heightened interferon-gamma signaling characterizing a tumor-suppressing function of the immune system at the first hematological complete remission stage,which likely resulted from therapy-induced cell death or senescence and ensuing supraphysiological levels of intracellular ***,our work sheds new light on the pathophysiology and treatment of APL and provides an information-rich reference data cohort for the exploratory and translational study of leukemia microenvironment.

Dear editor,Patients with clear-cell renal cell carcinoma(ccRCC)potentially have a high recurrence rate of more than 40%[1].Patients receive radical or partial nephrectomy,sometimes assisted by targeted therapies or/and immune checkpoint inhibitors(ICIs),to improve overall survival(OS)[2].Large-cohort studies and large-scale human tissue sequencing have provided a series of molecular subtyping methods for precision medicine-based ccRCC risk stratification and therapeutic regimens,such as ClearCode34,the prognostic risk predictor[3],and seven subsets for ICI and angiogenesis blockade outcomes[4].However,these subtyping results are all based on human genomes without including the host microbiota,which may be non-negligible genome components.

The current therapeutic drugs for Alzheimer's disease only improve symptoms,they do not delay disease *** re,there is an urgent need for new effective *** underlying pathogenic factors of Alzheimer's disease are not clear,but neuroinflammation can link various hypotheses of Alzheimer's disease;hence,targeting neuroinflammation may be a new hope for Alzheimer's disease *** inflammation can restore neuronal function,promote neuro regeneration,reduce the pathological burden of Alzheimer's disease,and improve or even reverse symptoms of Alzheimer's *** review focuses on the relationship between inflammation and various pathological hypotheses of Alzheimer's disease;reports the mechanisms and characteristics of small-molecule drugs(e.g.,nonsteroidal anti-inflammatory drugs,neurosteroids,and plant extracts);macromolecule drugs(e.g.,peptides,proteins,and gene therapeutics);and nanocarriers(e.g.,lipid-based nanoparticles,polymeric nanoparticles,nanoemulsions,and inorganic nanoparticles)in the treatment of Alzheimer's *** review also makes recommendations for the prospective development of anti-inflammatory strategies based on nanocarriers for the treatment of Alzheimer's disease.

Electrochemically converting CO_(2)into value-added chemicals is a promising approach to mitigate anthropogenic carbon emissions,yet largely limited to short-chained C_(1)–C_(3)***,we demonstrate a tandem artificial synthesis of biodegradable polyhydroxybutyrate(PHB)plastic from CO_(2)building *** synthesis of defects-enriched Bi catalyst is firstly demonstrated by plasma bombardment and following in situ electrochemical reduction,which delivers a HCOOH Faradaic efficiency above 80%at tunable concentration from 2 to 250 mM,an energy efficiency up to 41%,and a single-pass carbon conversion efficiency up to 60%.Annular dark field and second electron microscopic analysis,density functional theory(DFT)calcualtions,coupled with H-type and solid-state electrolyzer assessments,point out the vital role of defective and/or stepped Bi surface sites in promoting CO_(2)-to-HCOOH ***,as-synthesized high-purity HCOOH is used as the sole carbon source for C-chain growth within microbial fermentation reactor with Ralstonia eutropha,where activated formate dehydrogenase and increased metabolites related to Calvin–Benson–Bassham cycle are found to be responsible for the enhanced polyester accumulation.

Aim:Tumor budding(TB)has excellent prognostic value in many solid tumors,but there is little research on it in hepatocellular carcinoma(HCC).This study assessed the prognostic value of TB in patients with HCC who received ***:This retrospective study included 210 patients with HCC who received curative hepatectomy at the First affiliated hospital of xi'an jiaotong university,between 2016 and *** was evaluated on hematoxylin-and eosin-stained slides according to the criteria established by the 2016 International Tumor Budding Consensus Conference.t-tests,Chi-squared tests,and rank-sum tests were used to correlate the extent of TB with clinicopathological *** analysis was performed using Cox regression models and the Kaplan–Meier ***:The positive detection rate of TB was 45.2%(95/210)in 210 patients with *** positive for TB always exhibit lower tumor differentiation,higher hepatitis B virus DNA levels,and more severe liver *** Cox analysis identified TB(hazard ratio[HR]:2.232,95%confidence interval[CI]:1.479–3.368,p<0.001)as an independent prognostic factor for patients'recurrence-free survival(RFS),similar to tumor size(HR:1.070,95%CI:1.070–1.142,p=0.042)and satellite nodule(HR:2.266,95%CI:1.298–3.956,p=0.004).Kaplan–Meier analysis also demonstrated that TB-positive patients had a significantly worse ***,subgroup analysis revealed that among HCC patients with negative microvascular invasion(MVI),TB was also strongly associated with RFS(HR:3.206,95%CI:1.667–6.168,p<0.001).These findings suggest that TB may serve as a supplemental prognostic biomarker for HCC-negative ***:TB is an adverse prognostic biomarker for HCC,particularly in patients negative for *** evaluation should be considered in the postoperative pathological examination of HCC in clinical practice.
