Objective: To investigate the inhibitory effects of paeoniflorin on migration- and invasionpromoting capacities of gastric cancer associated fibroblasts (GCAFs) and to explore the molecular mechanism underlying the effects. Methods: Paired gastric normal fibroblast (GNF) and GCAF cultures were established from resected tissues. GCAFs were treated with control medium, or 2.5, 5 or 10 μg/mL paeoniflorin. Conditioned media were prepared from GNFs, GCAFs, control-treated GCAFs and paeoniflorin-treated GCAFs, and used to culture AGs human gastric cancer cells. The migration and invasion capacities of AGs cells were determined with wound healing test and transwell invasion assay, respectively. The interleukin 6 (IL-6) mRNA and microRNA-149 expression in GCAFs were detected by reverse transcription-quantitative polymerase chain reaction. The IL-6 protein expression and secretion by GCAFs were measured with Western blot and enzymelinked immunosorbent assay analysis, respectively. The protein levels of phosphorylated signal transducer and activator of transcription 3 (sTAT3), matrix metalloproteinase (MMP) and MMP9 in AGs cells were examined by Western blot. Results: GCAFs displayed enhanced capacities to induce AGs cell migration and invasion as compared with GNFs. Paeoniflorin treatment significantly inhibited the migration- and invasion-promoting capacities of GCAFs (Ps produced and secreted more IL-6 into the conditioned medium than GNFs, leading to over-activation of sTAT3-MMP signaling in AGs cells. Paeoniflorin suppressed IL-6 production and secretion by up-regulating microRNA149 expression in GCAFs, and subsequently prevented GCAFs from activating IL-6-sTAT3-MMP signaling of AGs cells. Conclusions: Paeoniflorin inhibits the migration- and invasion-promoting capacities of GCAFs by targeting microRNA-149 and IL-6. Paeoniflorin is potentially a novel therapeutic agent against cancer microenvironment.

AIM: To evaluate whether the level of thrombospondin-1(TsP-1) in aqueous humor can predict the prognosis of trabeculectomy in patients with primary angle-closure glaucoma(PACG). METHODs: This case-control study involved 26 patients with PACG who experienced a failed trabeculectomy(case group) and 78 age-and sex-matched patients with PACG who underwent successful trabeculectomy(control group). Aqueous humor was collected at the time of trabeculectomy and tested for TsP-1 and TGF-β2 levels with an enzyme-linked immunosorbent assay method. Logistic regression modeling was used to assess the risk factors for failed trabeculectomy. REsULTs: The mean TsP-1 aqueous concentrations were significantly higher in the case group(20.67±9.79 ng/mL) than the control group(5.17±2.29 ng/mL)(Psforming growth factor-β2(TGF-β2) aqueous concentrations were significantly different between the case and control group, at 3633.25 and 1090.24 pg/mL, respectively(Pstic regression analysis revealed TsP-1 level as an independent risk factor for a failed trabeculectomy(OR=3.540; 95%CI=1.092-11.482). CONCLUsION: The aqueous humor TsP-1 and TGF-β2 levels are higher in PACG eyes with failed trabeculectomy than with successful trabeculectomy at one year. The aqueous humor TsP-1 level is an independent risk factor associated with failed trabeculectomy.

AIM To investigate the inhibitory effect of astragaloside IV on the pathological functions of cancer-associated fibroblasts,and to explore the underlying *** Paired gastric normal fibroblast(GNF) and gastric cancer-associated fibroblast(GCAF) cultures were established from resected tissues. Chinese men should have a higher prostate-specific antigen （PsA）＂gray zone＂ than the traditional value of 2.5-10.0 ng ml-1 since the incidence of prostate cancer （PCa） in Chinese men is relative low. We hypothesized that PsA density （PsAD） could improve the rate of PCa detection in Chinese men with a PsA higher than the traditional PsA ＂gray zone.＂ A total of 461 men with a PsA between 2.5 and 20.0 ng ml-1, who had undergone prostatic biopsy at two Chinese centers were included in the analysis. The men were then further divided into groups with a PsA between 2.5-10.0 ng ml-1 and 10.1-20.0 ng ml-1. Receiver operating characteristic （ROC） curve was used to evaluate the efficacy of PsA and PsAD for the diagnosis of PCa. In men with a PsA of 2.5-10.0 ng ml-1 or 10.1-20.0 ng ml-z, the areas under the ROC curve were higher for PsAD than for PsA. This was consistent across both centers and the cohort overall. When the entire cohort was considered, the optimal PsAD cut-off for predicting PCa in men with a PsA of 2.5-10.0 ng m1-1 was 0.15 ng ml-2 ml-2, with a sensitivity of 64.4% and specificity of 64.6%. The optimal cut-off for PsAD in men with a PsA of 10.1-20.0 ng m1-1 was 0.33 ng ml-1 ml-1, with a sensitivity of 60.3% and specificity of 82.7%. PsAD can improve the effectiveness for PCa detection in Chinese men with a PsA of 2.5-10.0 ng ml-1 （traditional Western PsA ＂gray zone＂） and 10.1-20.0 ng ml-2 （Chinese PsA ＂gray zone＂）.
