Different mechanisms underlie similar species-area relationships in two tropical archipelagoes

Despite much research in the field of island biogeography,mechanisms regulating insular diversity remain ***,we aim to explore mechanisms underlying plant species-area relationships in two tropical archipelagoes in the South China *** found positive plant species-area relationships for both coral and continental ***,our results showed that different mechanisms contributed to similar plant species-area relationships between the two *** coral islands,soil nutrients and spatial distance among communities played major roles in shaping plant community structure and species *** contrast,the direct effect of island area,and to a lesser extent,soil nutrients determined plant species richness on continental ***,increasing soil nutrients availability(N,P,K)had opposite effects on plant diversity between the two *** summary,the habitat quality effect and dispersal limitation are important for regulating plant diversity on coral islands,whereas the passive sampling effect,and to a lesser extent,the habitat quality effect are important for regulating plant diversity on continental *** generally,our findings indicate that island plant species-area relationships are outcomes of the interplay of both niche and neutral processes,but the driving mechanisms behind these relationships depends on the type of islands.

CD8^(+)T cell immune responses are regulated by multi-layer networks,while the post-translational regulation remains largely *** ectodomain shedding is an important post-translational process orchestrating receptor expression and signal transduction through proteolytic cleavage of membrane ***,by targeting the sheddase A Disintegrin and Metalloprotease(ADAM)17,we defined a post-translational regulatory mechanism mediated by the ectodomain shedding in CD8^(+)T *** and proteomic analysis revealed the involvement of post-translational regulation in CD8^(+)T cells.T cellspecific deletion of ADAM17 led to a dramatic increase in effector CD8^(+)T cell differentiation and enhanced cytolytic effects to eliminate pathogens and ***,ADAM17 regulated CD8^(+)T cells through cleavage of membrane ***17 inhibition led to elevated CD122 expression and enhanced response to IL-2 and IL-15 stimulation in both mouse and human CD8^(+)T ***,inhibition of ADAM17 in CD8^(+)T cells improved the efficacy of chimeric antigen receptor(CAR)T cells in solid *** findings reveal a critical post-translational regulation in CD8^(+)T cells,providing a potential therapeutic strategy of targeting ADAM17 for effective anti-tumor immunity.

Insights into the Impact of Bifidobacterium bifidum， B. breve， and B. longum on the Immune Response Against Salmonella Infection in Neonatal Rats

Infants exhibit diminished susceptibility to external infections， a phenomenon closely linked to the pervasive presence of the bifidobacterial community within their intestinal tract. Nonetheless， a significant knowledge gap remains concerning the distinct species of bifidobacteria initiating anti-infection immune responses during the early developmental stages. In this study， the impact of early intervention with Bifidobacterium bifidum (B. bifidum)， B. breve， and B. longum on neonatal rats infected with Salmonella enterica serovar Typhimurium (S. typhimurium) SL1344 were compared. The study encompasses various immunity levels， including gut immunity， central immunity (thymus)， peripheral immunity (spleen)， and brain immunity. Following Salmonella infection， significant alterations in neonatal rats were observed in growth and developmental levels， immune markers， cytokine levels， balance of T lymphocyte subpopulations， intestinal barrier function， and blood-brain barrier integrity. Compared with B. breve and B. longum， B. bifidum demonstrated more pronounced efficacy in regulating these physiological processes. By conducting multi-level analyses of gut microbiota， bifidobacterial community， colonic content metabolomics， and serum metabolomics， the significance of B. bifidum's role is underscored， and the immune-enhancing function of messenger metabolites is unveiled. Among these metabolites， γ-L-Glutamyl-L-glutamic acid and Orotic acid were found to be shared by all three species， while Hippuric acid and 1a，1b-dihomo Prostaglandin F2α were unique to B. bifidum， and DL-Arginine was specific to B. longum. Overall， this study has provided novel insights into the intervention and immunomodulation by Bifidobacterium in early-life infections， emphasizing the significant role of B. bifidum.

Daily consumption of Siraitia grosvenorii alleviated lipid accumulation and colon barrier injury via modulation of gut microbiota in high-fat diet C57BL/6 mice

Siraitia grosvenorii (SG)， a traditional edible medicine in China， has shown potential application in health foods due to multiple bioactivities. However， few comparative studies have investigated the effect of daily intake of SG products by traditional drinking way on alleviating metabolic disorders caused by long-term high fat and high sugar diet (HF-HSD). This study investigated the effects of two commercial SG products (SG juice containing comprehensive compositions and mogrosides extract containing high content mogroside V) supplement on fat content， lipid accumulation， colon barrier and gut microbiota in a 38-week HF-HSD mice trial. Results showed that the SG intervention could decrease the weight gain and body fat content， alleviated lipid accumulation， low-grade inflammation and colon barrier injury. SG juice intake was found to increase the antioxidant enzyme activities of serum glutathione peroxidase (GSH-Px) and liver superoxide dismutase (SOD). 16S rDNA analysis found that SG could restore the gut microbiota with downregulation of harmful bacteria increased by HF-HSD and enhancement of the abundances of beneficial genus. In particular， SG juice significantly enriched taxonomic family Prevotellaceae and genus Alloprevotella. These results indicated that SG might be a functional additive to prevent disease risk caused by excess fat and sugar diet， and active ingredients resulting to the discrepancy of performance especially on gut microbiota are worthy of in-depth investigation.

极端干旱和食草昆虫可以共同影响植物群落,并且它们的交互作用可能受到干旱模式的影响。然而,目前尚缺乏相关的观测证据。本研究以半干旱草原为研究对象,通过控制实验探讨了不同极端干旱模式与食草昆虫的交互作用对植物群落的影响。实验设置了3种干旱处理,分别为对照(自然降雨量)、慢性干旱(生长季5–8月减少66%降雨量)和急性干旱(6–7月完全无降雨)。在各小区设置了两种食草昆虫处理,分别是有食草昆虫和无食草昆虫。结果表明,食草昆虫和极端干旱及二者的交互作用对物种丰富度无显著影响。食草昆虫缓解了慢性干旱对总盖度的负面影响,而这种缓解作用在急性干旱下较低。食草昆虫对慢性干旱带来的负面效应具有更强的缓解作用,这主要是因为食草昆虫导致慢性干旱下适口性较高的杂类草(具有较低叶片碳含量、叶片干物质含量,以及较低叶片淀粉、钙、镁、锰含量)的大幅降低,进而降低了禾草的负面效应。上述结果表明,干旱和食草昆虫的交互作用受干旱模式的影响。通过调节植物的适口性来控制食草昆虫的采食行为,可以减轻极端干旱对草原生态系统的影响。

In the latest issue of Nature,Zhang et *** a novel ladder-like structure of FOXP3-DNA interaction involving FOXP3 multimerization and remote DNA bridging through a combination of biochemistry,structural biology,cell biology,and bioinformatics analyses.1 In this commentary,we highlight their key findings and provide our insights into the research paradigm for further exploration of a novel transcriptional regulation mode as well as a therapeutic strategy from the structural aspects of the FOXP3 complex(Figure 1).

Transforming growth factor-beta 1 enhances discharge activity of cortical neurons

Transforming growth factor-beta 1(TGF-β1)has been extensively studied for its pleiotropic effects on central nervous system *** neuroprotective or neurotoxic effects of TGF-β1 in specific brain areas may depend on the pathological process and cell types ***-gated sodium channels(VGSCs)are essential ion channels for the generation of action potentials in neurons,and are involved in various neuroexcitation-related ***,the effects of TGF-β1 on the functional properties of VGSCs and firing properties in cortical neurons remain *** this study,we investigated the effects of TGF-β1 on VGSC function and firing properties in primary cortical neurons from *** found that TGF-β1 increased VGSC current density in a dose-and time-dependent manner,which was attributable to the upregulation of Nav1.3 *** VGSC current density and Nav1.3 expression were significantly abolished by preincubation with inhibitors of mitogen-activated protein kinase kinase(PD98059),p38 mitogen-activated protein kinase(SB203580),and Jun NH2-terminal kinase 1/2 inhibitor(SP600125).Interestingly,TGF-β1 significantly increased the firing threshold of action potentials but did not change their firing rate in cortical *** findings suggest that TGF-β1 can increase Nav1.3 expression through activation of the ERK1/2-JNK-MAPK pathway,which leads to a decrease in the firing threshold of action potentials in cortical neurons under pathological ***,this contributes to the occurrence and progression of neuroexcitatory-related diseases of the central nervous system.

Carbon neutralization has been introduced as a long-term policy to control global warming and climate *** plant photosynthesis produces the most abundant lignocellulosic biomass on Earth,its conversion to biofuels and bioproducts is considered a promising solution for reducing the net carbon ***,natural lignocellulose recalcitrance crucially results in a costly biomass process along with secondary waste *** updating recent advances in plant biotechnology,biomass engineering,and carbon nanotechnology,this study proposes a novel strategy that integrates the genetic engineering of bioenergy crops with green-like biomass processing for cost-effective biofuel conversion and high-value *** selecting key genes and appropriate genetic manipulation approaches for precise lignocellulose modification,this study highlights the desirable genetic site mutants and transgenic lines that are raised in amorphous regions and inner broken chains account for high-density/length-reduced cellulose nanofiber assembly in *** the amorphous regions and inner-broken chains of lignocellulose substrates are defined as the initial breakpoints for enhancing biochemical,chemical,and thermochemical conversions,desirable cellulose nanofibers can be employed to achieve nearcomplete biomass enzymatic saccharification for maximizing biofuels or high-quality biomaterials,even under cost-effective and green-like biomass processes in *** study emphasizes the optimal thermal conversion for generating high-performance nanocarbons by combining appropriate nanomaterials generated from diverse lignocellulose ***,this study provides a perspective on the potential of green carbon productivity as a part of the fourth industrial revolution.
