In an attempt to isolate and identify the target genes relevant to salt tolerance in a mangrove plant （Sesuvium portulacastrum L.）, a subtracted cDNA library was constructed via suppressive subtractive hybridization （SSH）, in which the poly（A）＋RNA isolated from salt-tolerant S. portulacastrum leaves was used as a tester, whereas the driver was poly（A）＋RNA, derived from salt-sensitive S. portulacastrum leaves. Screening of this subtracted cDNA library revealed five clones, of which the expression levels in the salt-tolerant plant were markedly higher than those observed in the salt-sensitive plant, indicating that these candidate clones may be involved in salt-tolerance pathways. Among the clones isolated, P66, P175, and P233 are novel because no significant similarity was obtained upon alignment with the GenBank database. Clone P89 demonstrated high homology with NADPH of Arabidopsis thaliana, whereas clone P152 was highly homologous with the gene encoding late embryogenesis abundant （LEA） protein of A. thaliana. The full-length gene of clone P152, with a predicated 344 amino acid residues, was shown to bear LEA-2 domains, a signature motif for proteins that have been enriched under salty and drought conditions. It is thus implied that clone P152 would be a salt-tolerance gene of S. portulacastrum. In addition, we have also developed a strategy for the extraction of total RNA from mangrove plants.

Mammalian and plant Rabl and Rab2 are small GTPases that regulate vesicle trafficking in the endoplasmic reticulum （ER） to Golgi compartments. Little is known about their functional diversification or potential interaction. We cloned sugarcane （Saccharum officinarum L.） Rab1A and Rab2A genes and studied their functional differences by expression and complementation experiments. We found differential expression of the two genes during sugarcane leaf development： SoRab2A expression declined from the dividing base to the maturing tip of the growing leaves, whereas SoRab1A was constitutively expressed, suggesting that SoRab2A is required for cell division and expansion and SoRablA is required for cells at all developmental stages. We used a yeast temperature sensitive ypt1-A 136D mutant strain to further investigate these shared and unique functions. Yptl is a small GTPase that regulates vesicle transport in the same cellular location as Rabl and Rab2. Neither SoRab1A nor SoRab2A alone could restore the growth of the mutant at restrictive temperatures when SoRab1A and SoRab2A were transformed separately. However, SoRab1A transformants maintained normal morphology and viability at non-permissive temperature, and resumed growth when returned to permissive temperature, whereas SoRab2A transformants died at non-permissive temperature, suggesting that SoRablA function is required for a cell＇s viability. Mutant growth was fully restored when SoRab1A and SoRab2A were co-transformed, indicating that SoRablA and SoRab2A complement each other and they both are needed to restore the function of ypt1-A136D. These results demonstrate that SoRab1A and SoRab2A serve distinct but overlapping functions, mostly by regulating the transportation of different sets of proteins.

Twenty-four compounds including eight steroids （1-8）, nine triterpenoids （9-16, 24）, three flavonoids （20-22）, and four benzenecarboxylic derivatives （17-19, 23） were isolated and identified from stems and twigs of medicinal mangrove plant Sonneratia caseolaris. The structures of the isolated compounds were determined by extensive analysis of their spectroscopic data. Among these rnetabolites, compounds 1, 4-20 and 22-24 were isolated and identified for the first time from S. caseolaris. In the in vitro cytotoxic assay against SMMC-7721 human hepatoma cells, compound 21 （3＇,4＇,5,7-tetrahydroxyflavone） exhibited significant activity with IC50 2.8 μg/mL, while oleanolic acid （14）, 3,3＇-di-O-methyl ether ellagic acid （18）, and 3,3＇,4-O-tri-O-methyl ether ellagic acid （19） showed weak activity. None of these compounds displayed significant antibacterial activites.

Two new tremulane sesquiterpenes coriolopsin A（1） and coriolopsin B（2）,together with two known ones conocenol C（3） and ceriponol E（4）,were isolated from the fermentation extract of endophytic fungus Coriolopsis sp.J5 from Ceriops *** structures and absolute configuration of compounds 1 and 2 were determined based on HR-ESI-MS and spectroscopic data（IR,UV,1D,2D NMR and X-ray crystallographic analysis）.

Soil Microbiological and Biochemical Properties as Affected by Different Long-Term Banana-Based Rotations in the Tropics

Soil microbiological and biochemical properties under various field crop rotations such as grains, pastures and vegetables have been studied intensively under short-term period. However, there is limited information about the influence of banana-based rotations on soil organic C, total N(TN), microbial biomasses and enzyme activities under long-term crop rotations. A field experiment arranged in a randomized complete block design with three replicates was carried out at the Wanzhong Farm in Ledong(18?37′–18?38′N, 108?46′–108?48′E), Hainan Province, China, to compare the responses of these soil parameters to long-term(10-year) banana(Musa paradisiaca)-pineapple(Ananas) rotation(AB), banana-papaya(Carica) rotation(BB) and banana monoculture(CK) in a conventional tillage system in the Hainan Island. Soil p H, total organic C(TOC), dissolved organic C(DOC), TN, total P(TP) and available P(AP) were found to be significantly higher(P < 0.01) in AB and BB than CK at 0–30 cm soil depth. Microbial biomass C(MBC) and N(MBN) were observed 18.0%–35.2% higher in AB and 8.6%–40.5% higher in BB than CK at 0–30 cm. The activities of urease(UA), invertase(IA), dehydrogenase(DA) and acid phosphatase(APA) showed a mean of 21.5%–59.6% increase in AB and 26.7%–66.1% increase in BB compared with CK at 0–30 cm. Higher p H, TOC and DOC at 0–10 and 10–20 cm than at 20–30 cm were obtained despite of the rotations. Soil MBC and MBN and activities of UA, IA and DA decreased markedly(P < 0.01) with increasing soil depth in the different rotation soils as well as the monoculture soil. In general, soil microbial biomass and enzymatic activities were more sensitive to changes in banana-based rotations than soil chemical properties, and consequently they were well-established as early indicators of changes due to crop rotations in the tropics.

Responses of Soil Nematode Abundance and Diversity to Long-Term Crop Rotations in tropical China

A field experiment was carried out from 2003 to 2013 in the Wanzhong Farm of the Hainan Island, China, to determine the effects of two long-term banana rotations on the abundance and trophic groups of soil nematode communities in the island. The experiment was set out as a randomized complete block design with three replications of three treatments： banana-pineapple rotation （AB）, banana-papaya rotation （BB） and banana monoculture （CK） in a conventional tillage system. Soil samples were taken at depths of 0-10, 10-20 and 20-30 cm, and nematodes were extracted by a modified cotton-wool filter method and identified to the genus level. Nematode ecological indices of Shannon-Weaver diversity （Ht）, dominance index （A）, maturity index （MI）, plant parasite index （PPI）, structure index （SI）, enrichment index （EI）, and channel index （CI） were calculated. A total of 28 nematode genera with relative abundance over 0.1% were identified, among which Tylenchus and Paratylenchus in the AB, Thonus in the BB, Tylenchus and Helicotylenchus in the CK were the dominant genera. The rotation soils favored bacterivores, fungivores and omnivores-predators with high colonizer-persister （c-p） values. Soil food web in the rotation systems was highly structured, mature and enriched as indicated by SI, MI and EI values, respectively. Higher abundance of bacterivores and lower values of CI suggested that the soil food web was dominated by a bacterial decomposition pathway in rotation soils. Nematode diversity was much higher after a decade of rotation. Soil depth had significant effects on the abundance of soil nematodes, but only on two nematode ecological indices （λ and MI）.

Hevein has been found to be an essential element in coagulation of rubber particles in latex of rubber trees. In a previous study, we cloned a 1 241-bp fragment of a 5＇ upstream region of the hevein gene by genome walking. This fragment was analyzed by a 5＇ end nested deletion method in the present study, fused with a uidA （gus） gene to produce a series of tested constructs, which were transferred into C-serum of latex and the Gus activities were detected. Results showed that the fragment from -749 to -292 was sufficient for expression of gus gene in latex, and the fragment from -292 to -168 was crucial in response to abscisic acid inducement. In a transient transgenic test of rubber leaf with particle bombardment, construct Hev749 conferred gus-specific expression in veins, in which the latex tubes mainly distributed. This implies that the fragment from -749 to -292 was laticiferous-specific.

Qualitative and Quantitative Analysis of Flidersiachromones in Three Agarwood Samples by HPLC-MS/MS

HPLC/ESI/MS2 was used in the qualitative and quantitative analysis of flidersiachromones（FTPECs） in three agarwood samples ofAquilaria sinensis, including an artificial holing agarwood, a wild agarwood, and a high quality agarwood named ＂Qi Nan＂. From these three agarwood samples, totally forty-six FTPECs were identified, and twenty-five of which were tentatively identified by the analysis of MS fragmentation ions, the others were indentiffed via comparing the retention time and MS fragmentation ions of them with those of the reference compounds. Besides, quantitative determination of five characteristic FTPECs（F14, F16 and F18-F20） in the three agarwoods was carried out, and it was found that the absolute content of each compound of these five FTPECs showed positive correlation with the quality of agarwood samples. Due to the wide distribution of four of these FTPECs（F16 and F18-F20） in the chemical constituents of agarwood, it is suggested that the total absolute content of these four FTPECs may be set as one of basis for agarwood quality evaluation.

Objective:To investigate whether eytochalasin D can induce antitumor activities in a tumor ***:Murine CT26 colorectal carcinoma cells were cultured hi vitro and cytochalasin D was used as a cytotoxic agent to detect its capabilities of inhibiting CT26 cell proliferation and inducing cell apoptosis by MTT and a TUNEL-based apoptosis *** CT26 tumor model was established to observe the tumor growth and survival *** tissues were used to detect the mierovessel density by *** addition,alginate encapsulated tumor cell assay was used to quantify the tumor angiogenesis in ***:Cytochalasin D inhibited CT26 tumor cell proliferation in lime and dose dependent manner and induced signiflcanl CT26 cell apoptosis,which almost reached the level induced by the positive control *** optimum effective dose of cytochalasin D for in vivo therapy was about 50 mg/*** D in vivo treatment significandy inhibited tumor growth and prolonged the survival times in CT26 tumor-bearing *** results of immunohistochemistry analysis and alginate encapsulation assay indicated that the cytochalasin D could effectively inhibited tumor angiogenesis. Conclusions:Cytochalasin D inhibits CT26 tumor growth potentially through inhibition of cell proliferation,induction of cell apoptosis and suppression of tumor angiogenesis.
