Alternative splicing of the PECTINESTERASE gene encoding a cell wall-degrading enzyme affects postharvest softening in grape

The firmness of table grape berries is a crucial quality parameter. Despite extensive research on postharvest fruit softening, its precise molecular mechanisms remain elusive. To enhance our comprehension of the underlying molecular factors, we initially identified differentially expressed genes(DEGs) by comparing the transcriptomes of folic acid(FA)-treated and water-treated(CK) berries at different time points. We then analyzed the sequences to detect alternatively spliced(AS) genes associated with postharvest softening. A total of 2,559 DEGs were identified and categorized into four subclusters based on their expression patterns, with subcluster-4 genes exhibiting higher expression in the CK group compared with the FA treatment group. There were 1,045 AS-associated genes specific to FA-treated berries and 1,042 in the CK-treated berries, respectively. Gene Ontology(GO) annotation indicated that the AS-associated genes in CK-treated berries were predominantly enriched in cell wall metabolic processes,particularly cell wall degradation processes. Through a comparison between treatment-associated AS genes and subcluster-4 DEGs, we identified eight genes, including Pectinesterase 2(VvPE2, Vitvi15g00704), which encodes a cell wall-degrading enzyme and was predicted to undergo an A3SS event. The reverse transcription polymerase chain reaction further confirmed the presence of a truncated transcript variant of VvPE2 in the FA-treated *** study provides a comprehensive analysis of AS events in postharvest grape berries using transcriptome sequencing and underscores the pivotal role of VvPE2 during the postharvest storage of grape berries.

Genomic selection (GS) has the potential to improve selection efficiency and shorten the breeding cycle in fruit tree breeding. In this study,we evaluated the effect of prediction methods, marker density and the training population (TP) size on pear GS for improving its performance and reducing cost. We evaluated GS under two scenarios:(1) five-fold cross-validation in an interspecific pear family;(2) independent validation. Based on the cross-validation scheme, the prediction accuracy (PA) of eight fruit traits varied between 0.33 (fruit core vertical diameter)and 0.65 (stone cell content). Except for single fruit weight, a slightly better prediction accuracy (PA) was observed for the five parametrical methods compared with the two non-parametrical methods. In our TP of 310 individuals, 2 000 single nucleotide polymorphism (SNP) markers were sufficient to make reasonably accurate predictions. PAs for different traits increased by 18.21%-46.98%when the TP size increased from 50to 100, but the increment was smaller (-4.13%-33.91%) when the TP size increased from 200 to 250. For independent validation, the PAs ranged from 0.11 to 0.45 using rrBLUP method. In summary, our results showed that the TP size and SNP numbers had a greater impact on the PA than prediction methods. Furthermore, relatedness among the training and validation sets, and the complexity of traits should be considered when designing a TP to predict the test panel.

Expression levels of aminopeptidase-N genes in the lightbrown apple moth, Epiphyas postvittana

Five aminopeptidase-N genes （EpAPN1-5） of the tortricid moth Epiphyas postvittana have been isolated from a midgut expressed sequence tag （EST） library. Relative RNA expression of these genes was measured by quantitative reverse transcription polymerase chain reaction using actin as a reference gene. Measurements were made on various tissues of fifth instar larvae, over all stages of the life cycle and under differing dietary conditions： different protein sources and levels, and in presence of trypsin and metalloprotease inhibitors. Gene expression for all five genes was the greatest in midgut tissue, but was also detected in the hindgut, fat body and Malpighian tubules. EpAPN4 was consistently the highest expressed, with EpAPN3 at about half that level; EpAPN5 was the least expressed. During larval stages expression was high, generally increasing over the instars, after an early peak as neonates or first instars. Expression in other life stages was much lower. Males and females showed differing expression in the pupal and adult stages： female expression was higher in the pupa, this reversed in the adult. Gene expression levels and ratios both changed with diet. A natural apple leaf diet depressed levels. Diets with the most impaired amino acid balance induced the greatest change; generally EpAPN1 increased by the greatest proportion. The addition of proteinase inhibitor also increased gene expression, and it was noteworthy that the trypsin inhibitor addition, which has been shown to double aminopeptidase activity, also doubled levels of gene expression.

Relationship Between Stimulated Ethylene Production and Alternative Respiration Pathway in "Royal Gala" Apple Fruit

Endogenous ethylene production and alternative oxidase (AOX) protein expression in 'Royal Gala' apple fruits were investigated after treatments with cold ( 0℃ for 1 week) and heat ( 38℃ for 1 h). A monoclonal antibody to the terminal oxidase of the alternative pathway from Sauromatum guttatum was used to identify the AOX protein in apple fruits. The molecular mass of AOX in 'Royal Gala' apple fruits is approximately 38 kDa, similar to those reported in tobacco and tomato. The cold treatment depressed the release of endogenous ethylene production before the climacteric ethylene production and obviously induced the expression of AOX protein expression. The heat treatment had the opposite effects on the ethylene production and AOX protein expression. In addition, the climax of endogenous ethylene production preceded the maximum AOX expression after the cold temperature treatment. It is therefore proposed that in climacteric fruits the production of induced ethylene is not coordinated with the level of AOX protein.

Responses of 9 lepidopteran species to Bacillus thuringiensis： How useful is phylogenetic relatedness for selecting surrogate species for nontarget arthropod risk assessment？

To evaluate phylogenetic relatedness as a proxy for susceptibility to Bacillus thuringiensis （Bt） when selecting species to act as surrogates for others in prerelease test- ing, we examined the responses of 11 laboratory-reared lepidopteran colonies, comprising members of 2 families, 5 genera, and 9 species, to a commercial Bt preparation. Sur- vival, pupal mass, and timing of pupation and adult emergence of 2 noctuids （Spodoptera litura and Helicoverpa armigera） and 7 tortricids （Cnephasia jactatana, Ctenopseustis obliquana, Ctenopseustis herana, Planotortrix octo, Planotortrix notophaea, Planotor- trix excessana [2 different laboratory colonies], and Epiphyas postvittana [2 colonies]） were examined after feeding first instar larvae with artificial diet containing 5 μL/100 mL Dipel ES （Bt subsp, kurstaki）. Bt caused significant larval mortality in all species except S. litura, in which only pupation was delayed compared with untreated controls. Neither of the noctuid species tested would act as a suitable surrogate for the other in tests of Bt impacts on survival. With the exception of the 2 colonies orE. postvittana, which differed from each other not only in their responses to Bt but also in their development times when not treated with Bt, species within each tortricid genus had similar responses to Bt and thus could act as surrogates for each other. Members of different genera within this family could represent each other only if relatively coarse measurement endpoints （e.g., toxic or not） were considered adequate for assessing risks to nontarget species in the field.

The allelic diversity of primitive germplasm of fruit crops provides a useful resource for introgressing novel genes to meet consumer preferences and environmental ***-breeding facilitates the identification of novel genetic variation in the primitive germplasm and expedite its utilisation in cultivar breeding *** generations of pre-breeding could be required to minimise linkage drag from the donor parent and to maximise the genomic content of the recipient *** this study we investigated the potential of genomic selection(GS)as a tool for rapid background selection of parents for the successive generation.A diverse set of 274 accessions was genotyped using random-tag genotyping-by-sequencing,and phenotyped for eight fruit quality *** relationship between‘own phenotypes’of 274 accessions and their general combining ability(GCA)was also *** heritability influenced the strength of correspondence between own phenotype and the *** average(across eight traits)accuracy of predicting own phenotype was 0.70,and the correlations between genomic-predicted own phenotype and GCA were similar to the observed *** results suggest that genome-assisted parental selection(GAPS)is a credible alternative to phenotypic parental selection,so could help reduce the generation interval to allow faster accumulation of favourable alleles from donor and recipient parents.
