Association of insulin resistance with serum ferritin and aminotransferases-iron hypothesis

AIM: To investigate the relationship of iron indices with diabetes mellitus(DM) in those without ***: This cross-sectional study examined data collected during the Third National Health and Nutrition Examination Survey(NHANES III). Only those who fasted properly and were not anemic with transferrin saturation cluded(n = 6849). Insulin sensitivity and beta cell function were calculated from fasting glucose and insulin concentrations. Indices of iron metabolism were examined in the presence or absence of DM. We examined the relationship of insulin sensitivity and beta cell function with serum ferritin concentration. The influence of c-reactive protein and liver enzymes was also ***: Serum ferritin concentration was significantly higher in diabetic subjects(P = 0.0001 toce remained significant after adjustment for age, body mass index, alcohol consumption, and mineral/iron supplement(P = 0.03 toconcentration was negatively associated with insulin sensitivity(P = 0.05 to 0.00001), but not with beta cell function. The alanine aminotransferase was correlated with serum ferritin concentration(P = 0.02 tociated insulin ***: As most of diabetes is type 2 diabetes and insulin resistance is a cardinal feature of type 2diabetes, disordered iron metabolism could play a role in the pathogenesis of insulin resistance and type 2diabetes through its effect on liver function.

AIM: To determine whether inducible nitric oxide syn-thase (iNOS) stimulation of human atrial fragments can be diminished by the naturally occurring signal molecules, such as morphine, anandamide, and estrogen . The use of iNOS as an indicator is justified since it has been associated with initiation of various types of cellular damage either directly or indirectly. METHODS: Western blots were performed on control and drug-exposed atrial tissue before and after lipopolysaccharide (LPS) and interferon-γ (IFN-γ) exposure. RESULTS: Preincubation of the tissue with morphine, anandamide or estrogen prior to, but not after, the addition of LPS + IFN-γ, blocked iNOS expression. The nitric oxide donor SNAP also blocked iNOS induction while preincubation of atrial fragments with an inhibitor of NOS, L-NAME, prior to morphine or anandamide exposure, restored LPS + IFN-γ induction of iNOS. cONcLUSION : These data suggest a direct regulatory link at the transcriptional level between constitutive (c) NOS and iNO

Amyloid-b-protein(Ab),the key component of senile plaques in Alzheimer’s disease(AD)brain,is produced from amyloid precursor protein(APP)by cleavage of b-secretase and then *** adaptor proteins with phosphotyrosine-binding(PTB)domains,including Dab(gene:DAB)and Numb(gene:NUMB),can bind to and interact with the conserved YENPTY-motif in the APP *** we describe,for the first time,the effects of RNAi knock-down of Dab and Numb expression on APP processing and Ab *** knock-down of Dab and Numb in H4 human neuroglioma cells stably transfected to express either FL-APP(H4-FL-APP cells)or APP-c99(H4-APP-c99 cells)increased levels of APP-c-terminal fragments(APP-cTFs)and lowered Ab levels in both cell lines by inhibiting g-secretase cleavage of ***,RNAi knock-down of APP also reduced levels of Numb in H4-APP *** findings suggest that pharmacologically blocking interaction of APP with Dab and Numb may provide novel therapeutic strategies of *** notion of attenuating g-secretase cleavage of APP via the APP adaptor proteins,Dab and Numb,is particularly attractive with regard to therapeutic potential,given that side effects of gsecretase inhibition owing to impaired proteolysis of other g-secretase substrates,***,might be avoided.

Telomerase -specific replication -selective virotherapy for oral squamous cell carcinoma cell lines

Purpose: Replication-selective tumor-specific viruses present a novel approach for treating neoplastic disease。These vectors are designed to induce virus-mediated lysis of tumor cells after selective viral propagation within the tumor。Telomerase activation is considered to be a critical step in carcinogenesis, and its activity is closely correlated with human telomerase reverse transcriptase (hTERT) expression。We investigated the antitumor effect of the hTERT-specific replication-competent adenovirus on oral squamous cell carcinoma (OScc) cells。Methods: An adenovirus 5 vector [tumor- or telomerase-specific replication-competent adenovirus (OBP-301)] was constructed, in which the hTERT promoter element drives expression of E 1A and EIB genes linked with an internal ribosome entry site, and we examined the selective replication and antitumor effect in human OScc cells in vitro and in vivo orthotopic graft model。Furthermore, we describe a new approach for visualizing tumors whose fluorescence can be detected using telomerase-specific replication-competent adenovirus expressing green fluorescent protein (GFP) (OBP-401)。OBP-401 contains the replication cassette, as same as OBP-301, and the GFP gene for monitoring viral replication。Results: OBP-301 replicated efficiently and induced more over 50% cell killing in a panel of human OScc cell lines, but not in normal human fibroblasts, which were lacking telomerase activity。In orthotopic graft model, intratumoral injection of OBP-301 resulted in a significant inhibition of tumor growth and prolongation of survival。Our data clearly indicate that OBP-301 displays an acceptable toxicity profile as well as a therapeutic oncolytic activity for OScc cells in vitro and in vivo。OBP-401 was intratumorally injected into OScc tumors orthotopically implanted into the tongue in BALB/c nu/nu mice。cervical lymph node metastasis could be visualized under a three-chip color cooled charged-coupled device camera。

目的通过网络meta分析比较不同类型和方式的心理治疗在青少年焦虑障碍患者中的效果。方法检索PubMed, cochrane cENTRAL, Embase, PsycINFO, Web of Science,cINAHL, ProQuest Dissertations, LILAcS, international trial registers以及FDA报道的从建库至2016年6月31日的文献。筛选出比较两种结构化心理治疗或结构化心理治疗与对照组在儿童青少年焦虑障碍患者中效果的随机对照试验。4位研究者分为两组独立地进行数据提取与质量评价。使用随机效应模型的成组meta分析和贝叶斯网络meta分析来整合数据。评价治疗结束和随访结束时的疗效（焦虑症状的改变）,耐受性（治疗结束时的失访）,以及生活质量和功能改善。短程和长程的随访的疗效同样也被测量。通过GRADE框架评估证据质量对每个网络的贡献。评价治疗结束和随访结束时的疗效（焦虑症状的改变）,耐受性（治疗结束时的失访）,以及生活质量和功能改善,同时评价短程和长程的随访的疗效。通过GRADE框架评估证据质量对每个网络的贡献。结果共纳入96个临床试验包括6256名参与者,比较了11种心理治疗与4种不同的对照措施。基于GRADE框架,大多数证据被评为低风险或极低风险。疗效结果显示,大多数心理治疗在治疗结束时（SMDs:-1.44到-0.65）和最长随访时（SMDs:-1.84到-1.64）显著优于等待治疗组。然而,只有团体cBT治疗比某些心理治疗和所有对照措施在治疗结束时和短期随访时有显著疗效。耐受性结果显示,阅读疗法比大多数心理疗法和对照措施具有更多的全因失访（ORs:0.06到0.37）。生活质量和功能改善结果显示,只有不同方式的cBT治疗比心理安慰剂治疗和等待治疗组显着有益（SMDs:0.65到2.01）。结论团体cBT是儿童青少年焦虑障碍患者的首选心理治疗。其他类型和方式的心理治疗可作为替代治疗选择。需进一步研究特定的焦虑障碍,特定障碍的心理治疗方法,以及影响疗效的因素。

Effect of calcium Phosphate Glass on Dentinal Tubule Sealing after Irradiation with the carbon Dioxide Laser

The aim of this study was to evaluate the dentinal tubule sealing and acid resistance of dentin specimens following the application of calcium phosphate glass powder prior to irradiation with a cO2(carbon dioxide)*** models simulating open dentinal tubules were divided into two groups:experimental(calcium phosphate glass slurry applied to the dentin surface)and control(no slurry applied to the surface).All specimens in the experimental group and five specimens in the control group were irradiated with a cO2 *** defocused laser beams(0.5 and 1 W)were applied(spot size,5 mm in diameter)from a distance of 20 mm for 10 *** surfaces and cross-sectional areas of the specimens were examined using an SEM(scanning electron microscope).In addition,the resistance to acid was evaluated in these *** open dentinal tubules in the control groups were sealed following irradiation with the cO2 laser at 0.5 W and 1.0 ***,sealing of open dentinal tubules was observed in the experimental group after cO2 laser *** acid resistance of the dentin surface was improved after cO2 laser irradiation;specimens in the experimental group presented with significantly lower amounts of ca ion release compared to those in the control *** findings indicate that cO2 laser irradiation alone or after the application of calcium phosphate glass powder can effectively seal the dentinal tubules and alleviate dentin hypersensitivity.
