Mapping of QTLs for Germination Characteristics under Non-stress and Drought Stress in Rice

Identification of genetic factors controlling traits associated with seed germination under drought stress conditions, leads to identification and development of drought tolerant varieties. Present study by using a population of F2：, derived from a cross between a drought tolerant variety, Gharib （indica） and a drought sensitive variety, Sepidroud （indica）, is to identify and compare QTLs associated with germination traits under drought stress and non-stress conditions. Through QTL analysis, using composite interval mapping, regarding traits such as germination rate （GR）, germination percentage （GP）, radicle length （RL）, plumule length （PL）, coleorhiza length （COL） and coleoptile length （CL）, totally 13 QTLs were detected under pole drought stress （-8 MPa poly ethylene glycol 6000） and 9 QTLs under non-stress conditions. Of the QTLs identified under non-stress conditions, QTLs associated with COL （qCOL-5） and GR （qGR-1） explained 21.28% and 19.73% of the total phenotypic variations, respectively Under drought stress conditions, QTLs associated with COL （qCOL-3） and PL （qPL-5） explained 18.34% and 18.22% of the total phenotypic variations, respectively. A few drought-tolerance-related QTLs identified in previous studies are near the QTLs detected in this study, and several QTLs in this study are novel alleles. The major QTLs like qGR-1, qGP-4, qRL-12 and qCL-4 identified in both conditions for traits GR, GP, RL and CL, respectively, should be considered as the important and stable trait-controlling QTLs in rice seed germination. Those major or minor QTLs could be used to significantly improve drought tolerance by marker-assisted selection in rice.

Gas-Water production of a Continental Tight-Sandstone Gas Reservoir under Different Fracturing Conditions

A numerical model of hydraulic fracture propagation is introduced for a representative reservoir(Yuanba continental tight sandstone gas reservoir in Northeast Sichuan).Different parameters are considered,i.e.,the interlayer stress difference,the fracturing discharge rate and the fracturing fluid *** results show that these factors affect the gas and water production by influencing the fracture *** interlayer stress difference can effectively control the fracture *** greater the stress difference,the smaller the dimensionless reconstruction volume of the reservoir,while the flowback rate and gas production are lower.A large displacement fracturing construction increases the fracture-forming efficiency and expands the fracture *** larger the displacement of fracturing construction,the larger the dimensionless reconstruction volume of the reservoir,and the higher the fracture-forming efficiency of fracturing fluid,the flowback rate,and the gas *** viscosity fracturing fluid is suitable for long fractures,while high viscosity fracturing fluid is suitable for wide *** an increase in the fracturing fluid viscosity,the dimensionless reconstruction volume and flowback rate of the reservoir display a non-monotonic behavior,however,their changes are relatively small.

Performance of Iranian Okra Ecotypes Under Various Accelerated Aging Conditions

Knowledge of seed quality and preservation of okra seeds is very limited, and the main purpose of this study was to simulate the condition of seed quality loss during storage and the physiological implications, which has led to seed vigor loss. A factorial experiment was conducted with the aim of studying the mechanism of longevity loss of the three endemic okra ecotypes of Iran(Ahwaz, Isfahan, Mashhad) in response to different accelerated aging durations(24, 48, 96, and 192 h). The accelerated aging condition was incubation of seeds to the temperature of 40 °C with 99% humidity. Germination of okra seeds was improved at 24 and 48 h compared to non-aged seeds, while at 192 h of accelerated aging, seed germination(49%) reduced to 50% of its value in 48 h(96.4%). Our results revealed that Iranian okra ecotypes were able to survive accelerated aging conditions up to 48 h. Antioxidant enzyme activity decreased with the increase in aging durations. Among investigated cultivars, seedling vigor of Mashhad ecotypes was the most sensitive to aging treatments. Our results revealed that the seed reservoir use efficiency value was 66%lower when exposed to 192 h of aging compared to the control. The cultivar Isfahan exhibited the minimum reduction of antioxidant activity,and, interestingly, this ecotype was the most aging tolerant.

Sequence boundaries and regularities in the oil-gas distribution of the low swelling slope belt in the continental rift basin

Where are the zones more enriched in sand deposits in the down slope and deep depression of the low swelling slope belt? Are there any screening conditions for oil and gas there? These are the chief geological problems to be solved during exploration of a region. Taking the Paleogene system developed along the east slope belt of Chengdao as an example the concepts of sequence stratigraphy and sedimentary sequenc are applied. A new research method likened to a way ''to get a melon by following the vine'' is proposed to determine the direction for exploring within un-drilled or less-drilled areas. This is the process: ''the characteristics of the sequence boundary ? the forming mechanism of the stratigraphic sequence ? the conditions of oil and gas accumulation ? the distribution zones of oil and gas''. The relationship between the dynamic mechanism of stratigraphic sequence and the forming conditions for oil and gas accumulation establishes that the tectonic disturbance of the slope belt has significant responses as denudation and deposition. Above the stratigraphic sequence boundary there are large scale sand bodies of the low stand system tract (LST) that have developed in the low swelling slope belt and its deep depression. Below the sequence boundary there are the remaining sand bodies of the high stand system tract (HST). On the slope there is a convergence of mudstone layers of the extended system tract (EST) with the mudstone of the underlying strata, which constitutes the screening conditions for the reservoir of the down slope and deep depression. The distribution regularities in preferred sand bodies on the surface of the sequence boundary, and in the system tract, indicate the ordering of oil-gas deposits. From the higher stand down to the depth of the slope there are, in order, areas where exploration was unfavorable, major areas of stratigraphic overlap of oil-gas reservoirs, unconformity screened oil-gas reservoirs, and, finally, sandstone lens oil-gas

Laboratory experiments have been carried out to study the fluid flow in the wellbore of a horizontal gas well during the production *** related pressure distribution has been determined considering different cases(different inflow media,different perforation opening methods and different liquid holdup).It has been found that the larger the fluid flow rate,the greater the pressure changes in the wellbore under the same hole opening *** uniformity of the perforation opening method was also an important factor affecting the magnitude of the wellbore pressure *** liquid holdup also affected the pressure distribution,especially when the gas volumetric flow rate exceeded 200 m3/*** of the outcomes of the present experimental study with the predictions of a theoretical model available in the literature has provided a relative error smaller than 20%.

Effects of N Management on Yield and N Uptake of Rice in Central China

Efficient N fertilizer management is critical for the economic production of rice and the long-term protection of environmental quality. A field experiment was designed to study the effects of N fertilizer management practices on grain yield and N uptake office. The experiment was laid out in the randomized complete block design with four replications in Central China during 2008 and 2009. Five N treatments denoted as No, NIsoA, N15oB, N24OA, and N24oB, respectively, were studied. NO represented no N application and served as a control, N15OA and N15oB indicated the total N application of 150 kg N ha-I but with two different application schedules （A and B） across the early stage of rice growth. Schedule A was applied as follows： 40% basal, 30% at 10 d after transplanting （DAT） and 30% at 36 DAT （nearly at the panicle initiation stage）, while schedule B was as follows： 30% at basal, 20% at 10 DAT, and 50% at 36 DAT. Similarly, N240A and N240B indicated the total N application of 240 kg N ha-1 with schedules A and B as described above. To quantify N uptake from fertilizer and soil, a 15N experiment was also conducted within the main experimental field, with micro-plots. Grain yields were significantly increased as N rates increased from 0 to 240 kg N ha-1. At the same rate, splitting N application as schedule B significantly increased the grain yield, spikelets per panicle, percentage of ripened grain, and 1 000-grain weight, compared with the N application according to schedule A. Mean rice recovery of N fertilizer by ~SN tracing method ranged from 25.39% at N24oA to 34.89% at NIsoB, however, N fertilizer residual rate in the soil ranged from 12.40% at N240A to 16.61% at N^s0w About 31.5 and 28.5% of total uptake of ~SN derived from basal fertilizer was absorbed at panicle initiation and heading stages, respectively.
