Panax notoginseng Saponins Protect Kidney from Diabetes by Up-regulating Silent Information Regulator 1 and Activating Antioxidant Proteins in Rats

Objective： To explore the mechanism of the protective effects of Panax notoginseng saponins（PNS） on kidney in diabetic rats. Methods： Diabetic rat model was obtained by intravenous injection of alloxan, and the rats were divided into model, PNS-100 mg/（kg·day） and PNS-200 mg/（kg·day） groups, 10 each. Another 10 rats injected with saline were served as control. Periodic acid-Schiff staining and immunological histological chemistry were used to observe histomorphology and tissue expression of bone morphogenetic protein-7（BMP-7）. Silent information regulator 1（SIRT1） was silenced in rat mesangial cells by RNA interference. The mR NA expressions of SIRT-1, monocyte chemoattractant protein-1（MCP-1）, transforming growth factor β1（TGF-β1） and plasminogen activator inhibitor-1（PAI-1） were analyzed by reverse transcription polymerase chain reaction. The protein expressions of SIRT1 and the acetylation of nuclear factor κB（NF-κB） P65 were determined by western blotting. The concentration of MCP-1, TGF-β1 and malondialdehyde（MDA） in culture supernatant were detected by enzyme-linked immuno sorbent assay. The activity of superoxide dismutase（SOD） was detected by the classical method of nitrogen and blue four. Results： In diabetic model rats, PNS could not only reduce blood glucose and lipid（P〈0.01）, but also increase protein level of BMP-7 and inhibit PAI-1 expression for suppressing fibrosis of the kidney. In rat mesangial cells, PNS could up-regulate the expression of SIRT1（P〈0.01） and in turn suppress the transcription of TGF-β1（P〈0.05） and MCP-1（P〈0.05）. PNS could also reverse the increased acetylation of NF-κB p65 by high glucose. In addition, redox regulation factor MDA was down-regulated（P〈0.05） and SOD was up-regulated（P〈0.01）, which were both induced by SIRT1 up-regulation. Drug-induced liver injury(DILI)is a common adverse drug reaction,which can even result in liver failure[1,2].The chinese medical Association issued the Guidelines for the Diagnosis and Treatment of DILI based on the Roussel Uclaf Causality Assessment Method(RUCAM)in 2015[3].A previous study reported that traditional chinese medicines(TCMs),herbal and dietary supplements,and antituberculosis drugs were the main causes of DILI in China[4].Herb-induced liver injury(HILI)refers to liver injury caused by TCMs,natural drugs,and their related preparations[5].

Stevens-Johnson syndrome/toxic epidermal necrolysis(SJS/TEN)is a rare adverse cutaneous reaction with a low incidence and high *** posing a serious threat to patients’health and lives,there is no high-quality evidence for a standard treatment *** we report the case of a 62-year-old man with stage IV pancreatic cancer who experienced immunotherapy-induced SJS/*** consensus-based regular treatments at a local hospital,his symptoms became ***,he consented to receive chinese herbal medicine(CHM)*** affected parts of the patient were treated with the CHM Pi-Yan-Ning which was applied externally for 20 min twice a *** 7 days of treatment,the dead skin began peeling away from the former lesions that had covered his hands,feet,and lips,indicating that skin had *** 12 days of treatment,the patient’s skin was completely *** this case,SJS/TEN was successfully treated with Pi-Yan-Ning,suggesting that there might be tremendous potential for the use of Pi-Yan-Ning in the treatment of severe skin reactions to drug *** basic investigations and clinical trials to explore the mechanism and efficacy are needed.

Protective effects of effective ingredients of Danshen (Radix Salviae Miltiorrhizae)and Honghua (Flos Carthami)compatibility after rat hippocampal neurons induced by hypoxia injury

OBJECTIVE: To investigate the effective ingredients of Danshen(Radix Salviae Miltiorrhizae) and Honghua(Flos Carthami)(Danhong) on protective properties towards neonatal rat hippocampal neurons under hypoxia ***: Primary culture of neonatal rat hippocampal neurons was used to model hypoxia damage on the hippocampus. Methyl thiazolyl tetrazolium(MTT) assay and preliminary experiments were conducted to identify the four effective ingredients of Danhong, along with the injection of positive drug onto hippocampal neurons at a non-toxic dosage level. The cultured cells were randomly divided into 12 groups: the normal group, model group,positive drug control group and nine compatibility groups of the four effective ingredients. Different test methods were applied to determine lactate dehydrogenasein(LDH), total superoxide dismutase(T-SOD), malondialdehyde(MDA), 6-keto prostaglandin F_(la)(6-keto-PGF_(1 a)), thromboxane B_2(TXB_2),mitochondrial membrane potential(MMP), free calcium ions concentration([Ca^(2 +)]i) and early onsetcell ***: Different compatibility groups could inhibit the content of LDH and intracellular calcium overload, increase activity in T-SOD, decrease level of MDA and TXB2, improve level of 6-keto-PGF1 aand MMP, and prevent the early onset cell ***: The compatibility of four effective ingredients of Danhong had protective effect toward hippocampi hypoxia. The mechanism might be related to inhibit oxidative stress damage and cell apoptosis, resist thrombosis, and reduce the intracellular calcium ion of overload.

Photothermal/matrix metalloproteinase-2 dual-responsive gelatin nanoparticles for breast cancer treatment

The chemotherapy combined with photothermal therapy has been a favorable approach for the treatment of breast *** present study,nanoparticles with the characteristics of photothermal/matrix metalloproteinase-2(MMP-2)dual-responsive,tumor targeting,and size-variability were designed for enhancing the antitumor efficacy and achieving"on-demand"drug release *** on the thermal sensitivity of gelatin,we designed a size-variable gelatin nanoparticle(GNP)to encapsulate indocyanine green(ICG)and doxorubicin(DOX).Under an 808 nm laser irradiation,GNP-DOX/ICG responded photothermally and swelled in size from 71.58?4.28 to 160.80?9.51 nm,which was beneficial for particle retention in the tumor sites and release of the loaded ***,GNP-DOX/ICG showed a size reduction of the particles to 33.24?4.11 nm and further improved drug release with the degradation of overexpressed MMP-2 in *** the subsequently performed in vitro experiments,it was confirmed that GNP-DOX/ICG could provide a therapeutic effect that was enhanced and ***,GNP-DOX/ICG could efficiently suppress the growth of 4 T1 tumor in *** conclusion,this study may provide a promising strategy in the rational design of drug delivery nanosystems based on gelatin for chemo-photothermal therapy to achieve synergistically enhanced therapeutic efficacy against breast cancer.
