Genetic variation is a key factor influencing cytokine production capacity,but which genetic loci regulate cytokine production before and after vaccination,particularly in African population is ***,we aimed to identify single-nucleotide polymorphisms(SNPs)controlling cytokine responses after microbial stimulation in infants of West-African ancestry,comprising of low-birth-weight neonates randomized to bacillus Calmette-Gue rin(BCG)vaccine-at-birth or to the usual delayed ***-wide cytokine cytokine quantitative trait loci(cQTL)mapping revealed 12 independent loci,of which the LINC01082-LINC00917 locus influenced more than half of the cytokine-stimulation pairs ***,nine distinct cQTLs were found among infants randomized to *** validation confirmed that several complement genes affect cytokine response after BCG *** observed a limited overlap of common cQTLs between the West-African infants and cohorts of Western European *** data reveal strong population-specific genetic effects on cytokine production and may indicate new opportunities for therapeutic intervention and vaccine development in African populations.

Room-temperature tracking of chiral recognition process at the single-molecule level

The molecular-level identification of a chiral recognition process of phthalocyanine （Pc） was studied on a Cu（100） surface by scanning tunneling microscopy （STM）. STM revealed that a chiral Pc molecule forms a series of metastable dimer configurations with other Pc molecules. Eventually, the Pc molecule recognizes another Pc molecule with the same chirality to form a stable dimer configuration. Homochiral dimers were found on the Cu surface, demonstrating the chiral specificity of Pc dimerization. The mechanism for this chiral recognition process is identified, disclosing the critical role of the particular adsorption geometry of the chiral dimers on the Cu surface.

*** strongest locus which associated with type 2 diabetes(T2D)by the common variant rs7903146 is the transcription factor 7-like 2 gene(TCF7L2).We aimed to quantify the interaction of diet/lifestyle interventions and the genetic effect of TCF7L2 rs7903146 on glycemic traits,body weight,or waist circumference in overweight or obese adults in several randomized controlled trials(RCTs).*** October 2016 to May 2018,a large collaborative analysis was performed by pooling individualparticipant data from 7 *** RCTs reported changes in glycemic control and adiposity of the variant rs7903146 after dietary/lifestyle-related interventions in overweight or obese *** treatment interaction models which used the genetic effect encoded by the allele dose and common covariates were applicable to individual participant data in all *** the joint analysis,a total of 7 eligible RCTs were included(n=4,114).Importantly,we observed a significant effect modification of diet/lifestyle-related interventions on the TCF7L2 variant rs7903146 and changes in fasting *** with the control group,diet/lifestyle interventions were related to lower fasting glucose by-3.06(95%CI,-5.77 to-0.36)mg/dL(test for heterogeneity and overall effect:I^(2)=45:1%,p<0:05;z=2:20,p=0:028)per one copy of the TCF7L2 T risk ***,regardless of genetic risk,diet/lifestyle interventions were associated with lower waist ***,there was no significant change for diet/lifestyle interventions in other glycemic control and adiposity traits per one copy of TCF7L2 risk *** findings suggest that carrying the TCF7L2 T risk allele may have a modestly greater benefit for specific diet/lifestyle interventions to improve the control of fasting glucose in overweight or obese adults.

Polymeric hydrogels are fascinating platforms as 3D scaffolds for tissue repair and delivery systems of therapeutic molecules and *** others,methacrylated gelatin(GelMA)has become a representative hydrogel formulation,finding various biomedical *** efforts on GelMA-based hydrogels have been devoted to combining them with bioactive and functional nanomaterials,aiming to provide enhanced physicochemical and biological properties to *** benefits of this approach are multiple:i)reinforcing mechanical properties,ii)modulating viscoelastic property to allow 3D printability of bio-inks,iii)rendering electrical/magnetic property to produce electro-/magneto-active hydrogels for the repair of specific tissues(e.g.,muscle,nerve),iv)providing stimuli-responsiveness to actively deliver therapeutic molecules,and v)endowing therapeutic capacity in tissue repair process(e.g.,antioxidant effects).The nanomaterial-combined GelMA systems have shown significantly enhanced and extraordinary behaviors in various tissues(bone,skin,cardiac,and nerve)that are rarely observable with *** we systematically review these recent efforts in nanomaterials-combined GelMA hydrogels that are considered as next-generation multifunctional platforms for tissue *** approaches used in GelMA can also apply to other existing polymeric hydrogel systems.

The evolution of a two-chambered heart,with an atrium and a ventricle,has improved heart function in both deuterostomes(vertebrates)and some protostomes(invertebrates).Although studies have examined the unique structure and function of these two chambers,molecular comparisons are few and limited to ***,we focus on the two-chambered protostome heart of the mollusks,offering data that may provide a better understanding of heart ***,we asked if the atrium and ventricle differ at the molecular level in the mollusk *** do so,we examined two very different species,the giant African land snail(Lissachatina fulica)and the relatively small,aquatic yesso scallop(Mizuhopecten yessoensis),with the assumption that if they exhibited commonality these similarities would likely reflect those across the *** found that,although the hearts of these two species differed histologically,their cardiac gene function enrichments were similar,as revealed by transcriptomic ***,the atrium and ventricle in each species had distinct gene function clusters,suggesting an evolutionary differentiation of cardiac chambers in ***,to explore the relationship between vertebrate and invertebrate two-chambered hearts,we compared our transcriptomic data with published data from the zebrafish,a well-studied vertebrate model with a two-chambered *** analysis indicated a functional similarity of ventricular genes between the mollusks and the zebrafish,suggesting that the ventricle was differentiated to achieve the same functions in invertebrates and *** the first such study on protostomes,our findings offered initial insights into how the two-chambered heart arose,including a possible understanding of its occurrence in both protostomes and deuterostomes.

Single center experience with intrathoracic impedance monitoring in chronic heart failure patients

Background The Medtronic InSync Sentry is the first available cardiac resynchronization therapy defibrillator （CRT-D）which can monitor fluid status by measuring intrathoracic impedance. This study was designed to observe the effectiveness of intrathoracic impedance monitoring on detecting aggravation of heart *** We retrospectively analyzed the clinical data of 14 consecutive patients. Patients were regularly followed up every 3-6 months after the implantation. At each visit, interrogation of the device was done. Patients were instructed to inform the researcher on hearing the device alert, and to take extra 40 milligrams of furosemidum if they had aggravated symptoms later. If the symptoms could not be relieved, they were asked to see a doctor. Data about heart failure hospitalization were collected from the medical *** During 18-48 months follow-up, a total of 7 patients encountered 28 alert events. On one hand, alert events appeared before all deteriorated symptoms and heart failure hospitalizations. On the other hand, there were 23 alerts followed by deterioration of heart failure symptoms, and 2 alerts related to 2 hospitalizations caused by pulmonary infection in one patient. Only 5 patients were hospitalized 10 times for deterioration of cardiac *** The function of intrathoracic impedance monitoring is reliable in predicting deterioration of heart failure.
