Selenium Regulates Antioxidant Capacities and Diterpenoid Biosynthesis in the Medicinal Plant Isodon rubescens

Dōng líng căo,the dried aboveground parts of Isodon rubescens(Hemls.)Hara.,is commonly consumed as a med-icinal decoction or tea *** beverages can be an important source of human dietary selenium(Se).However,how *** plants respond to exogenous Se remains *** this study,a pot cultivation experiment was employed to investigate the phenotypic and physiological responses of *** plants exposed to *** days after applying different concentrations of sodium selenate to the soil,the Se enrichment capa-city,growth indices,antioxidant capacities,and the content ofﬂavonoids and diterpenoids were measured in the ***,the oridonin content was quantiﬁed using the high-performance liquid chromatography method,and the expression levels of key diterpenoid synthesis genes were analyzed by quantitative real-time PCR(qRT-PCR).*** plants efﬁciently accumulated Se,with the Se content increasing proportionally to the applied dose,reaching levels of nearly 200 mg·kg^(-1) dry leaves as Se concentration *** of the three Se treat-ments signiﬁcantly altered the phenotypic indices,except a longer root length occurred in the 3μM·kg^(-1) Se *** three Se doses,6μM·kg^(-1) Se gave the highest accumulation ofﬂavonoids,diterpenoids,and oridonin,with the increase of 2.0-,1.8-,and 1.9-fold in aboveground parts,*** application boosted the activities of antioxidant enzymes and antioxidant capacities according to 2,2-Diphenyl-1-picrylhydrazyl(DPPH),ferric reducing/antioxidant power,and tea brewing color *** key synthase genes were upregulated signiﬁcantly by 6μM·kg^(-1) Se treatment,notably 1-deoxy-D-xylulose 5-phosphate reductoisomerase(IrDXR),with a 5-fold increase,and kaurene synthase-like 4(IrKSL4),with a 6-fold ***,Se application in *** cultivation may be a potential biofortiﬁcation method to supplement Se while increasingﬂavonoid and diterpenoid contents.

Metabolism is mobilized to meet the biosynthetic,bioenergetic,and regulatory demands of immune cell *** nothing is known about the metabolic basis of antibody class switch in B cells.A recent study published in Nature Communications through a joint effort by Chen Lab(metabolism biology)and Liu Lab(B cell biology)from Tsinghua University reveals that the monocarboxylate carrier MCT1(monocarboxylate transporter 1)promotes pyruvate oxidation and histone acetylation to support class switch recombination(CSR),the process by which B cells make antibodies of different isotypes with unique effector functions.

Water scarcity and pollution pose a threat to the sustainable development of cities and ***,it is crucial to analyze the hydrochemical characteristics and carbon dynamics of waterdeficient *** the Taiyuan section of Fenhe River as the research object,we systematically explored the hydrochemical characteristics of surface water and its evolutionary processes,as well as the ecological effect of algal carbonic anhydrase in carbon cycle using the hydrochemical evolution method and correlation *** ternary diagram demonstrates that the main water chemical type in Fenhe River was SO^(2-)_(4)·Cl^(-)-Na^(+).The Gibbs and end-member diagrams of each ion display that the chemical composition of surface water was mainly controlled by silicate *** chemical ions originated mainly from dissolution of some minerals,such as plagioclase,halite,dolomite,calcite,and *** diatoms had a lower CO_(2)requirement because they exhibited a higher abundance at a lower partial pressure of CO_(2)(p CO_(2)).However,high CO_(2)concentration had a positive effect on cyanobacteria,which reduced the active transport of HCO_(3)^(-),saved the energy needed for this part of active transport,and indirectly improved the overall photosynthetic efficiency of *** anhydrase(CA)activity was significantly negatively correlated with p CO_(2)and positively correlated with HCO_(3)^(-)concentration,indicating that CA in water promoted the conversion of CO_(2)to HCO_(3)^(-).The HCO_(3)^(-)generated from this process continued to participate in the erosion of silicate rocks,sequestering CO_(2)in the form of Ca CO_(3),which has a non-negligible impact on the carbon sink in the Fenhe *** consequences may have important implications for the biogeochemical cycling occurring in urban water.

Coptis chinensis Franch.,also named Chinese goldthread is a member of Ranunculaceae in the order Ranunculales and represents an important lineage of early eudicots with traditional medicinal *** our study,by using syntenic analysis combined with phylogenomic analysis of *** and four other representative genomes from basal and core eudicots,we confirmed that the WGD event in *** was shared by Aquilegia coerulea and Papaver somniferum *** quickly occurred after Ranunculales diverged from other eudicots,likely a Ranunculales common tetraploidization(RCT).The synonymous nucleotide substitutions at synonymous sites distribution of syntenic blocks across these genomes showed that the evolutionary rate of the *** genome is faster than that of the *** genome by approximately 13.7%,possibly due to Papaveraceaes having an additional special tetraploidization event(PST).After Ks correction,the RCT dated to 115—130 million years ago(MYA),which was close to the divergence of Ranunculaceaes and Papaveraceaes approximately115.45—130.51 ***,we identified homologous genes related to polyploidization and speciation and constructed multiple sequence alignments with different reference ***,the event-related subgenomes in the basal genomes all showed genomic fractionation bias,suggesting a likely allopolyploid nature of the RCT,PST and T-Alpha and T-Beta events in Tetracentron *** addition,we detected that the sixteen P450 subfamilies were markedly expanded in the genomes of Ranunculales,and most of them were related to the RCT and PST *** constructed a new platform for Early Eudicot Comparative Genomic Research(http://***/***)to store more *** summary,our findings support the WGD of *** shared by Ranunculales,which is likely an allotetraploidization *** present effort offered new insights into the evolution of key polyploidization events and the genes related to second

Dormancy represents a fascinating adaptive strategy for organisms to survive in unforgiving *** a period of dormancy,organisms often exhibit exceptional *** period is typically divided into hibernation and aestivation based on seasonal ***,the mechanisms by which organisms adapt to their environments during dormancy,as well as the potential relationships between different states of dormancy,deserve further ***,we selected Perccottus glenii and Protopterus annectens as the primary subjects to study hibernation and aestivation,*** on histological and transcriptomic analysis of multiple organs,we discovered that dormancy involved a coordinated functional response across *** analyses revealed noteworthy disparities between the two dormant species in their responses to extreme ***,similarities in gene expression patterns pertaining to energy metabolism,neural activity,and biosynthesis were noted during hibernation,suggesting a potential correlation between hibernation and *** further explore the relationship between these two phenomena,we analyzed other dormancy-capable species using data from publicly available *** comparative analysis revealed that most orthologous genes involved in metabolism,cell proliferation,and neural function exhibited consistent expression patterns during dormancy,indicating that the observed similarity between hibernation and aestivation may be attributable to convergent *** conclusion,this study enhances our comprehension of the dormancy phenomenon and offers new insights into the molecular mechanisms underpinning vertebrate dormancy.

GPCRs are dominant targets for approved drugs and the discovery of lead compound targeting them is still ***-based screening technique is a promising platform to uncover GPCR ***,the intrinsic activities of them are seldom simultaneously determined during the *** beta2-adrenoceptor(β2AR)as a probe,this work created a strategy for screening GPCR ligands with simultaneous characterization of their downstream G protein binding responses associated with *** strategy included(i)the design and expression of a protein miniature formed byβ2AR and G proteinα-subunit(Gαs)using circularly permuted HaloTag(cpHalo)as a flexible linker;(ii)immobilization of the miniature onto silica gel by a click dehalogenation reaction;(iii)systematic characterization of the immobilized miniature by fluorescent and chromatographic studies,and(iv)simulating of ligand-inducedβ2AR-Gαs signaling cascade by chromatographic assays using GTP as an *** immobilized miniature exhibited specificity toβ2AR and Gαs antibodies and *** specificity is stable at least within fifteen days with the variation less than 1%.The intrinsic activities ofβ2AR ligands were distinguished by the changes of GTP chromatographic behaviors on Gαs-cpHalo-β2AR *** strengthened the binding affinity and kinetics of GTP with Gαs,while antagonist did not give any effect on *** the intrinsic activity evaluation,we believe,it will improve the attributes of chromatographic methods for drug discovery efforts with minimizing false-positive results.

Solving Multi-Objective Vehicle Routing Problems with Time Windows: A Decomposition-Based Multiform Optimization Approach

In solving multi-objective vehicle routing problems with time windows (MOVRPTW),most existing algorithms focus on the optimization of a single problem formulation. However,little effort has been devoted to exploiting valuable knowledge from the alternate formulations of MOVRPTW for better optimization performance. Aiming at this insufficiency,this study proposes a decomposition-based multi-objective multiform evolutionary algorithm (MMFEA/D),which performs the evolutionary search on multiple alternate formulations of MOVRPTW simultaneously to complement each other. In particular,the main characteristics of MMFEA/D are three folds. First,a multiform construction (MFC) strategy is adopted to construct multiple alternate formulations,each of which is formulated by grouping several adjacent subproblems based on the decomposition of MOVRPTW. Second,a transfer reproduction (TFR) mechanism is designed to generate offspring for each formulation via transferring promising solutions from other formulations,making that the useful traits captured from different formulations can be shared and leveraged to guide the evolutionary search. Third,an adaptive local search (ALS) strategy is developed to invest search effort on different alternate formulations as per their usefulness for MOVRPTW,thus facilitating the efficient allocation of computational resources. Experimental studies have demonstrated the superior performance of MMFEA/D on the classical Solomon instances and the real-world instances.
