Introduction:All countries face the potential threat of imported polioviruses,including both wild type and circulating vaccine-derived *** response,we conducted a province-level assessment in china to evaluate the risk of importation and transmission of type 1 wild poliovirus(WPV1)and type 2 vaccine-derived poliovirus(VDPV2).Methods:Distinct risk assessment tools for WPV1 and VDPV2 were employed,incorporating three primary indicators—population immunity,poliovirus surveillance,and importation ***1 was assessed using 13 secondary indicators,whereas VDPV2 utilized 21 secondary *** used comprehensive provincial data from the preceding five ***(S-values)were derived from the secondary indicators’criteria and ratings,and used to classify the provincial-level administrative divisions(PLADs)into three risk categories:high,medium,and *** top 10%of PLADs were designated as highrisk,with the remaining provinces equally distributed into medium-and low-risk ***:In 2023,Xizang,Qinghai,and Xinjiang PLADs faced the highest risk of WPV1 importation and transmission;Xizang,Shaanxi,and Hainan PLADs were at the greatest risk for VDPV2 importation and ***:Risk assessment for VDPV2 importation and transmission has identified a distinct set of high-risk provinces compared to those identified by WPV1 risk *** and proactive response measures tailored to the specific risks should be implemented to maintain china’s polio-free status.

Introduction:This study examines the seasonal and genetic characteristics of human metapneumovirus(HMPV)in Henan from 2017 to ***:Samples from patients with acute respiratory infection(ARI)testing positive for HMPV were subjected to real-time reverse transcription polymerase chain reaction The G gene was amplified and sequenced from these samples for epidemiological and phylogenetic ***:We enrolled 2,707 ARI patients from October 2017 to March 2023,finding an HMPV positivity rate of 6.17%(167/2,707).Children under five exhibited the highest infection rate at 7.78%(138/1,774).The 2018 and 2019 HMPV outbreaks predominantly occurred in spring(March to May),with peak positivity rates of 31.11%in May 2018 and 19.57%in May 2019.A notable increase occurred in November 2020,when positivity reached a historic high of 42.11%,continuing until January *** February 2021 through March 2023,no significant seasonal peaks were observed,with rates ranging from 0%to 8.70%.Out of 81 G gene sequences analyzed,46.91%(38/81)were identified as subtype A(A2c:45.67%,37/81;A2b:1.23%,1/81)and 53.09%(43/81)as subtype B(B1:9.88%,8/81;B2:43.21%,35/81).Notably,an AAABBA switch pattern was observed in HMPV *** dominant strains were A2c111nt-dup in subtype A and B2 in subtype ***:Six years of surveillance in Henan Province has detailed the seasonal and genetic dynamics of HMPV,contributing valuable insights for the control and prevention of HMPV infections in *** findings support the development of targeted HMPV vaccines and immunization strategies.

Objective This study investigated the impact of occupational mercury(Hg) exposure on human gene transcription and expression, and its potential biological *** Differentially expressed genes related to Hg exposure were identified and validated using gene expression microarray analysis and extended validation. Hg-exposed cell models and PTEN lowexpression models were established in vitro using 293T cells. PTEN gene expression was assessed using qRT-PCR, and Western blotting was used to measure PTEN, AKT, and PI3K protein levels. IL-6 expression was determined by *** Combined findings from gene expression microarray analysis, bioinformatics, and population expansion validation indicated significant downregulation of the PTEN gene in the high-concentration Hg exposure group. In the Hg-exposed cell model(25 and 10 μmol/L), a significant decrease in PTEN expression was observed, accompanied by a significant increase in PI3K, AKT, and IL-6 ***, a low-expression cell model demonstrated that PTEN gene knockdown led to a significant decrease in PTEN protein expression and a substantial increase in PI3K, AKT, and IL-6 *** This is the first study to report that Hg exposure downregulates the PTEN gene, activates the PI3K/AKT regulatory pathway, and increases the expression of inflammatory factors, ultimately resulting in kidney inflammation.
