Two is better than one: combinatorial receptor targeting enhances hepatocellular carcinoma (HCC) therapeutic response

Finding effective therapies for hepatocellular carcinoma(HCC)is an ongoing challenge for both researchers and clinicians.Enthusiasm for new treatment options emerging from in vitro,preclinical,and clinical studies is often tempered by limited patient benefits and associated side effects,tolerated in some cases due to the lack of alternatives.The complexity of identifying effective treatment options resides in several features of HCC,notably the extraordinary heterogeneity in molecular alterations among different patients(1-3),the lack of consistent alterations associated with oncogene addictions(4),and a so-called“non-oncogene addiction”by which tumorigenicity results from the action of non-mutated genes(5,6).This complex configuration also prevents identification of unequivocal alterations used to design therapies,as for some other cancer types.Current HCC treatments are based on the Barcelona Clinic Liver Cancer(BCLC)guidelines according to the stage of lesions among other parameters.For HCC at early stages(30-40%of HCC patients),treatments include lesion resection,liver transplantation,radiofrequency ablation(RFA)and microwave ablation(MWA),achieving a median overall survival(OS)of more than 60 months.For HCC at intermediate stages(~20%of HCC patients),the advised treatment is generally transarterial chemoembolization(TACE),achieving an OS of 20 months.For HCC at advanced stages(~40%of HCC patients),targeted therapies are the most frequent treatments,leading to an OS of 11 months with most targeted monotherapies.For HCC at terminal stages,only palliative care is provided(~10%of HCC patients),with less than 3 months OS(7,8).

Influence of toxic substances on dynamical behavior of a delayed diffusive predator-prey model

In this paper,we propose and investigate a delayed diffusive predator-prey model affected by toxic substances.We first study the boundedness and persistence property of the model.By analyzing the associated characteristic equation,we obtain the conditions for the existence of steady state bifurcation,Hopf bifurcation and turing bifurcation.Furthermore,we also study the Hopf bifurcation induced by the delay.Finally,our theoretical results are verified by numerical simulation.The numerical observation results are in good agreement with the theoretically predicted results.Theoretical and numerical simulations indicate that toxic substances have a great impact on the dynamics of the system.

Evaluating ChatGPT’s Consciousness and Its Capability to Pass the turing Test: A Comprehensive Analysis

This study explores the capabilities of ChatGPT, specifically in relation to consciousness and its performance in the turing Test. The article begins by examining the diverse perspectives among both the cognitive and AI researchers regarding ChatGPT’s ability to pass the turing Test. It introduces a hierarchical categorization of the test versions, suggesting that ChatGPT approaches success in the test, albeit primarily with na?ve users. Expert users, conversely, can easily identify its limitations. The paper presents various theories of consciousness, with a particular focus on the Integrated Information Theory proposed by Tononi. This theory serves as the framework for assessing ChatGPT’s level of consciousness. Through an evaluation based on the five axioms and theorems of IIT, the study finds that ChatGPT surpasses previous AI systems in certain aspects;however, ChatGPT significantly falls short of achieving a level of consciousness, particularly when compared to biological sentient beings. The paper concludes by emphasizing the importance of recognizing ChatGPT and similar generative AI models as highly advanced and intelligent tools, yet distinctly lacking the consciousness attributes found in advanced living organisms.

We study the problem of the unsupervised learning of graphical models in mixed discrete-continuous domains.The problem of unsupervised learning of such models in discrete domains alone is notoriously challenging,compounded by the fact that inference is computationally demanding.The situation is generally believed to be significantly worse in discrete-continuous domains:estimating the unknown probability distribution of given samples is often limited in practice to a handful of parametric forms,and in addition to that,computing conditional queries need to carefully handle low-probability regions in safety-critical applications.In recent years,the regime of tractable learning has emerged,which attempts to learn a graphical model that permits efficient inference.Most of the results in this regime are based on arithmetic circuits,for which inference is linear in the size of the obtained circuit.In this work,we show how,with minimal modifications,such regimes can be generalized by leveraging efficient density estimation schemes based on piecewise polynomial approximations.Our framework is realized on a recent computational abstraction that permits efficient inference for a range of queries in the underlying language.Our empirical results show that our approach is effective,and allows a study of the trade-off between the granularity of the learned model and its predictive power.

Acute myeloid leukaemia(AML)patients harbouring certain chromosome abnormalities have particularly adverse prognosis.For these patients,targeted therapies have not yet made a significant clinical impact.To understand the molecular landscape of poor prognosis AML we profiled 74 patients from two different centres(in UK and Finland)at the proteomic,phosphoproteomic and drug response phenotypic levels.These data were complemented with transcriptomics analysis for 39 cases.Data integration highlighted a phosphoproteomics signature that define two biologically distinct groups of KMT2A rearranged leukaemia,which we term MLLGA and MLLGB.MLLGA presented increased DOT1L phosphorylation,HOXA gene expression,CDK1 activity and phosphorylation of proteins involved in RNA metabolism,replication and DNA damage when compared to MLLGB and no KMT2A rearranged samples.MLLGA was particularly sensitive to 15 compounds including genotoxic drugs and inhibitors of mitotic kinases and inosine-5-monosphosphate dehydrogenase(IMPDH)relative to other cases.Intermediate-risk KMT2A-MLLT3 cases were mainly represented in a third group closer to MLLGA than to MLLGB.The expression of IMPDH2 and multiple nucleolar proteins was higher in MLLGA and correlated with the response to IMPDH inhibition in KMT2A rearranged leukaemia,suggesting a role of the nucleolar activity in sensitivity to treatment.In summary,our multilayer molecular profiling of AML with poor prognosis and KMT2A-MLLT3 karyotypes identified a phosphoproteomics signature that defines two biologically and phenotypically distinct groups of KMT2A rearranged leukaemia.These data provide a rationale for the potential development of specific therapies for AML patients characterised by the MLLGA phosphoproteomics signature identified in this study.

Mechanism analysis of regulating turing instability and Hopf bifurcation of malware propagation in mobile wireless sensor networks

A dynamical model is constructed to depict the spatial-temporal evolution of malware in mobile wireless sensor networks(MWSNs). Based on such a model, we design a hybrid control scheme combining parameter perturbation and state feedback to effectively manipulate the spatiotemporal dynamics of malware propagation. The hybrid control can not only suppress the turing instability caused by diffusion factor but can also adjust the occurrence of Hopf bifurcation induced by time delay. Numerical simulation results show that the hybrid control strategy can efficiently manipulate the transmission dynamics to achieve our expected desired properties, thus reducing the harm of malware propagation to MWSNs.

Battery state-of-charge estimation using machine learning analysis of ultrasonic signatures

The potential of acoustic signatures to be used for State-of-Charge(SoC)estimation is demonstrated using artificial neural network regression models.This approach represents a streamlined method of processing the entire acoustic waveform instead of performing manual,and often arbitrary,waveform peak selection.For applications where computational economy is prioritised,simple metrics of statistical significance are used to formally identify the most informative waveform features.These alone can be exploited for SoC inference.It is further shown that signal portions representing both early and late interfacial reflections can correlate highly with the SoC and be of predictive value,challenging the more common peak selection methods which focus on the latter.Although later echoes represent greater through-thickness coverage,and are intuitively more information-rich,their presence is not guaranteed.Holistic waveform treatment offers a more robust approach to correlating acoustic signatures to electrochemical states.It is further demonstrated that transformation into the frequency domain can reduce the dimensionality of the problem significantly,while also improving the estimation accuracy.Most importantly,it is shown that acoustic signatures can be used as sole model inputs to produce highly accurate SoC estimates,without any complementary voltage information.This makes the method suitable for applications where redundancy and diversification of SoC estimation approaches is needed.Data is obtained experimentally from a 210 mAh LiCoO2/graphite pouch cell.Mean estimation errors as low as 0.75%are achieved on a SoC scale of 0-100%.

The release of the generative pre-trained transformer(GPT)series has brought artificial general intelligence(AGI)to the forefront of the artificial intelligence(AI)field once again.However,the questions of how to define and evaluate AGI remain unclear.This perspective article proposes that the evaluation of AGI should be rooted in dynamic embodied physical and social interactions(DEPSI).More specifically,we propose five critical characteristics to be considered as AGI benchmarks and suggest the Tong test as an AGI evaluation system.The Tong test describes a value-and ability-oriented testing system that delineates five levels of AGI milestones through a virtual environment with DEPSI,allowing for infinite task generation.We contrast the Tong test with classical AI testing systems in terms of various aspects and propose a systematic evaluation system to promote standardized,quantitative,and objective benchmarks and evaluation of AGI.

Light perception at dawn plays a key role in coordinating multiple molecular processes and in entraining the plant circadian clock.The Arabidopsis mutant lacking the main photoreceptors,however,still shows clock entrainment,indicating that the integration of light into the morning transcriptome is not well understood.In this study,we performed a high-resolution RNA-sequencing time-series experiment,sampling every 2 min beginning at dawn.In parallel experiments,we perturbed temperature,the circadian clock,photoreceptor signaling,and chloroplast-derived light signaling.We used these data to infer a gene network that describes the gene expression dynamics after light stimulus in the morning,and then validated key edges.By sampling time points at high density,we are able to identify three light-and temperature-sensitive bursts of transcription factor activity,one of which lasts for only about 8 min.Phytochrome and cryptochrome mutants cause a delay in the transcriptional bursts at dawn,and completely remove a burst of expression in key photomorphogenesis genes(HY5 and BBX family).Our complete network is available online(http://www-users.***.ac.uk/∼de656/dawnBurst/dawnBurst.html).Taken together,our results show that phytochrome and cryptochrome signaling is required for fine-tuning the dawn transcriptional response to light,but separate pathways can robustly activate much of the program in their absence.

Alignment between galaxies and large-scale structure

Based on the Sloan Digital Sky Survey DR6 （SDSS） and the ＇Millennium Simulation （MS）, we investigate the alignment between galaxies and large-scale structure. For this purpose, we develop two new statistical tools, namely the alignment correlation function and the cos（20）-statistic. The former is a two-dimensional extension of the traditional two-point correlation function and the latter is related to the ellipticity correlation function used for cosmic shear measurements. Both are based on the cross correlation between a sample of galaxies with orientations and a reference sample which represents the large-scale structure. We apply the new statistics to the SDSS galaxy catalog. The alignment correlation function reveals an overabundance of reference galaxies along the major axes of red, luminous （L 〉～L＊） galaxies out to projected separations of 60 h-lMpc. The signal increases with central galaxy luminosity. No alignment signal is detected for blue galaxies. The cos（2θ）-statistic yields very similar results. Starting from a MS semi-analytic galaxy catalog, we assign an orientation to each red, luminous and central galaxy, based on that of the central region of the host halo （with size similar to that of the stellar galaxy）. As an alternative, we use the orientation of the host halo itself. We find a mean projected misalignment between a halo and its central region of -25°. The misalignment decreases slightly with increasing luminosity of the central galaxy. Using the orientations and luminosities of the semi-analytic galaxies, we repeat our alignment analysis on mock surveys of the MS. Agreement with the SDSS results is good if the central orientations are used. Predictions using the halo orientations as proxies for cen- tral galaxy orientations overestimate the observed alignment by more than a factor of 2. Finally, the large volume of the MS allows us to generate a two-dimensional map of the alignment correlation function, which shows the reference galaxy distribution to be flat- tened parallel to the orientat
