Glycogen synthase kinase-3β positively regulates the proliferation of human ovarian cancer cells

Although glycogen synthase kinase-3(GSK-3)might act as a tumor suppressor since its inhibition is expected tomimic the activation of Wnt-signaling pathway,GSK-3β may contribute to NF-κ3 activation in cancer cells leading toincreased cancer cell proliferation and *** we report that GSK-3β activity was involved in the proliferationof human ovarian cancer cell both in vitro and in *** of GSK-3 activity by pharmacological inhibitors sup-pressed proliferation of the ovarian cancer *** constitutively active form of GSK-3β induced entryinto the S phase,increased cyclin D1 expression and facilitated the proliferation of ovarian cancer ***,GSK-3 inhibition prevented the formation of the tumor in nude mice generated by the inoculation of human ovariancancer *** findings thus suggest that GSK-3β activity is important for the proliferation of ovarian cancer cells,implicating this kinase as a potential therapeutic target in ovarian cancer.

Objective： To make a prognostic effect analysis of molecular subtype on young breast cancer ***： Totally 187 cases of young breast cancer patients less than 40 years old treated in obstetrics and gynecology hospital of fudan university between June 2005 and June 2011 were included in our study. We described their clinical-pathological characteristics, disease-free survival（DFS） rate, and overall survival（OS） rate after a median follow-up period of 61 months. The factors associated with prognosis were also evaluated by univariate and multivariate ***： All patients were premenopausal, with an average age of 35.36±3.88 years old. The mean tumor size was 2.43±1.53 cm. Eighty-one cases had lymph node metastasis（43.3%）, 126 cases had lymphovascular invasion（67.4%）, and 125 cases had histological grade III（66.8%） disease. Twenty-seven cases（14.4%） were Luminal A subtype, 99 cases（52.9%） were Luminal B subtype, 29 cases（15.5%） were human epidermal growth factor receptor 2（HER-2） overexpression subtype, while 32 cases（17.1%） were triple negative breast cancer（TNBC） subtype according to 2013 St Gallen expert consensus. One hundred and thirty-five cases underwent mastectomy whereas 52 cases had breast-conserving surgery. One hundred and seventy-eight cases underwent adjuvant or neoadjuvant chemotherapy. Recurrence or metastasis occurred in 29 cases, 13 of which died. The 5-year DFS and OS rates were 84% and 92%. Multivariate analysis showed that nodal status（P=0.041） and molecular subtype（P=0.037） were both independent prognostic factors of DFS, while nodal status（P=0.037） and TNBC subtype（P=0.048） were both independent prognostic factors of OS. Conclusions： Molecular subtype is an independent prognostic factor of young breast cancer patients. TNBC has a high risk of relapse and death.

For infertility treatment,the selection of in vitro fertilization(IVF)or intracytoplasmic sperm injection(ICSI)is decided by multiplying indicators(including fallopian tube factors,semen count,and semen motility),except for sperm *** this study,we conducted a retrospective analysis,from implantation to birth,over a period of 5 years.A total of 1873 couples with primary or secondary fallopian tube factors and an increased defective sperm morphology rate(DSMR)were divided into different groups to receive IVF or ICSI *** comparing the outcomes,we found that the F1 group(DSMR98%,IVF group 3;P98%,ICSI group 3)had higher cleavage rate(P<0.001),biochemical pregnancy rate(P<0.05),clinical pregnancy rate(P<0.05)and live birth rate(P<0.05)than the F3 ***,the ICSI subgroup had a lower two pronuclei fertilization rate than the IVF subgroup(P<0.05).Our data suggest that the sperm morphology should also be considered when selecting IVF or ICSI combined with other semen parameters before the first assisted reproductive technologies(ART)cycle,especially for males with severe sperm defects.

Objective To assess the fibrinogen levels in polycystic ovary syndrome （PCOS） patients and to fgure out the potential of fibrinogen to be an early phase laboratory biomarker for PCOS diagnosis. Methods A systemic literature research was performed from databases of Medline via OVID, PubMed, Web of Knowledge and the Cochrane library databases, etc. Fifteen studies including 694 PCOS patients and 548 normal controls were selected after quality assessment. Data of fibrinogen levels, age and body weight were extracted and analyzed systemically. Results Fibrinogen increases in PCOS patients with regard to data poolings of overall studies （mean difference=21.51, 95%CI=3.78-39.25, P=0.02）, National Institutes of Health （NIH）-based studies （mean difference=8. 50, 95%CI=0.91-16. 09, P= 0. 03） and Rotterdam-based studies （mean difference=45.61, 95%CI=12.18-79.03, P=0.007） respectively. PCOS patients are younger （mean difference= -2.15, 95%CI= -3.65 - -0. 64, P=0. 005） and with heavier body weight （mean difference=0.92, 95%CI=0.59- 1.25, P〈0.001） than normal controls. Conclusions Increasing in PCOS patients, fibrinogen might be used as a diagnosis biomarker for PCOS, especially in the population with younger age and heavier body weight, which indicates that fibrinogen might be used as an early phase laboratory screening biomarker in clinical practice.

Objective: This study aimed to investigate the relationship between previous contraceptive methods and postmenopausal health in older ***: A questionnaire was administered to value the health status of women aged at least 65 years. Factors associated with general health and reproductive health were ***: A total of 2746 older women were enrolled, and hypertension was the most common non-neoplastic disease (37.62%, 1033/2746) among the group. A total of 123 patients were diagnosed with postmenopausal neoplastic disease. Breast cancer was the most common malignancy among older women (37/123, 31%). Women in the ligation group had a lower incidence of hypertension (239/1033, 23.14%), and women in the intrauterine devices (IUDs) group had a higher incidence of postmenopausal human papillomavirus (HPV) infection (140/200, 70%). The incidence of breast cancer significantly decreased after tubal ligation (2/37, 5.4%), whereas women who used condoms had a significantly lower incidence of cervical cancer after menopause (2/26, 7.69%).Conclusions: Hypertension is a common condition among women aged 65 and older, and breast cancer is the most prevalent malignancy. Among women aged 65 and older, the incidence of hypertension and breast cancer was lower after tubal ligation. Cervical cancer is the most common gynecological malignancy in postmenopausal women. Women who used IUDs had a higher risk for HPV infection. The incidence of cervical cancer is significantly lower in women who used condoms compared to those who did not.
