Objective： Plasma phospholipid transfer protein （PLTP） is a key determinant of lipoprotein metabolism, and both animal and human studies converge to indicate that PLTP promotes atherogenesis and its thromboembolic complications. Moreover, it has recently been reported that PLTP modulates inflammation and immune responses. Although earlier studies from our group demonstrated that PLTP can modify macrophage activation, the implication of PLTP in the modulation of T-cell-mediated immune responses has never been investigated and was therefore addressed in the present study. Approach and results： In the present study, we demonstrated that PLTP deficiency in mice has a profound effect on CD4＋ thO cell polarization, with a shift towards the anti-inflammatory th2 phenotype under both normal and pathological conditions. In a model of contact hypersensitivity, a significantly impaired response to skin sensitization with the hapten-2,4-dinitrofluorobenzene （DNFB） was observed in PLTP-deficient mice compared to wild-type （WT） mice. Interestingly, PLTP deficiency in mice exerted no effect on the counts of total white blood cells, lymphocytes, granulocytes, or monocytes in the peripheral blood. Moreover, PLTP deficiency did not modify the amounts of CD4＋ and CD8＋ T lymphocyte subsets. However, PLTP-deficiency, associated with upregulation of the th2 phenotype, was accompanied by a significant decrease in the production of the pro-thl cytokine interleukin 18 by accessory cells. Conclusions： For the first time, this work reports a physiological role for PLTP in the polarization of CD4＋ T cells toward the pro-inflammatory th I phenotype.

the wavelength modulation indices are measured based on harmonic amplitude ratio of 4famp/6famp (4famp and 6famp are the 4- and 6-th harmonic central peak amplitudes correspondingly) with the Doppler-free selective reflection modulation spectroscopy. the experiments for the 6S1/2(F = 4) → 6P3/2(F' = 5) transition of cesium D2 line with 30-MHz linewidth were carried out. the 4f- and 6f-harmonic signals were detected with two digital lock-in amplifiers separately. the maximum error for modulation indices measurement was ±0.1 within the range of m from 3 to 6. the non-linear modulation behaviour of an external cavity diode laser induced by voltage tuning was studied with this method. the method for modulation indices measurement does not require a solid etalon as usual for measuring the wavelength modulation depth and the absorption linewidth correspondingly.

Influence of hypoxia on retinal progenitor and ganglion cells in human induced pluripotent stem cell-derived retinal organoids

AIM:To observe the effect of low oxygen concentration on the neural retina in human induced pluripotent stem cell(hiPSC)-derived retinal organoids(ROs).MEthODS:the hiPSC and a three-dimensional culture method were used for the *** embryoid bodies(EBs)were randomly and equally divided into hypoxic and normoxic *** of the EBs were taken on days 38,45,and 52,and the corresponding volume of EBs was ***,samples were collected at these three timepoints,followed by fixation,sectioning,and ***:the proportion of Ki67-positive proliferating cells increased steadily on day 38;this proliferationpromoting effect tended to increase tissue density rather than tissue *** days 45 and 52,the two groups had relatively similar ratios of Ki67-positive *** immunofluorescence analysis showed that the ratio of SOX2-positive cells significantly increased within the neural retina on day 52(Pthe percentage of PAX6-and chX10-positive cells significantly decreased following hypoxia treatment at all three timepoints(PchX10 at day 45(P>0.05).Moreover,the proportion of PAX6-/TUJ1+cells within the neural retinas increased considerably(Pthat hypoxic conditions can enlarge the retinal progenitor cell pool in hiPSC-derived ROs.

the progress of research in nucleic acid immunization promises new avenues to many infectious disease prophylaxis and therapeutics in human and animals. the commercialization and licensing of DNA vaccines are subjected to similar if not more stringent regulatory and market conditions as in many conventional vaccines in terms of administration regimen and dose presentation. the efficacy of a rabies DNA vaccine containing a plasmid encoding for the rabies virus glycoprotein gene was evaluated in dogs in a single-dose regimen. the extended duration of immunity of the vaccine was demonstrated by challenge with a wild type virus 17 months post vaccination. Similarly, conventional equine West Nile Virus vaccines are available in the United States as a monovalent as well as multivalent combination vaccines containing a number of other viral antigens.

Phosphatase and Tensin Homologue Genetic Polymorphisms and their Interactions with Viral Mutations on the Risk of Hepatocellular Carcinoma

Background： chronic hepatitis B virus （HBV） infection is the major cause of hepatocellular carcinoma （HCC）. Some HBV mutants and dysregulation of phosphatase and tensin homolog （PTEN） may promote the development of HCC synergistically. We aimed to test the effects of PTEN genetic polymorphisms and their interactions with important HBV mutations on the development of HCC in HBV-infected subjects. Methods： Quantitative po[ymerase chain reaction was applied to genotype PTEN polymorphisms （rs1234220, rs2299939, rsl234213） in 1012 healthy controls, 302 natural clearance subjects, and 2011 chronic H BV-infected subjects including 1021 HCC patients. HBV mutations were determined by sequencing. the associations of PTEN polymorphisms and their interactions with HBV mutations with HCC risk were assessed using multivariate logistic regression analysis. Results： Rs1234220 C allele was significantly associated with HCC risk compared to healthy controls （adjusted odds ratio [AOR] = 1.35, 95% confidence interval [CI] = 1,07-1.69） and HCC-free HBV-infected subjects （AOR = 1.27, 95% CI = 1.01-1 .57）. rsl234220 C allele was significantly associated with increased frequencies of HCC-risk A 1652G, C 1673T, and C 1730G mutations in genotype B H BV-in fected subjects. Rs2299939 GT genotype was inversely associated with HCC risk in HBV-infected patients （AOR 0.75, 95% CI 0.62-0.92）. the interaction of rs2299939 variant genotypes （GT＋TT） with A3054T mutation significantly increased HCC risk （AOR = 2.41, 95% CI = 1.08-5.35）; whereas its interaction with C3116T mutation significantly reduced HCC risk （AOR = 0.34, 95% CI 0.18-0.66）. these significant effects were only evident in males alter stratification. Conclusions： PTEN polymorphisms and their interactions with HBV mutations may contribute to hepatocarcinogenesis in males. the host-virus interactions are important in identifying HBV-infected subjects who are more likely to develop HCC.

Prognosis Value of Two Gravity Scores in Patients Admited to the Intensive Care Unit at University Hospital of Parakou in Benin

Introduction: the gravity of the clinical state of the patient of intensive care is linked to the existence of one or several lesions and visceral defaults putting its forecast into play. Objective: To determine the prognosis value of two general graveness’ scores of patients admitted to intensive care at University Hospital of Parakou in Benin. Patients and Methods: Descriptive and analytical observational study data were collected from March 1st to June 30th, 2017. the SAPS II and APAchE II were calculated during first 24 hours of hospitalization to assess the clinical graveness and predict patient’s mortality. Results: We enrolled 185 patients representing 89.37% of admissions, majority were men (63.78%). Mean age was 38.89 ± 17.92 years (16 to 99), mean of hospitalization duration was 4.36 ± 2.2 days. Neurological failure was the most common disorder observed (58.37%). Mean SAPS II and APAchE II were 29.54 ± 19.04 and 14.24 ± 10.49 respectively. Mean predicted mortality of SAPS II and APAchE II was 19.12 ± 5.05 and 25.69 ± 5.00 respectively. the mortality rate was 25.95% and increased with severity scores. the sensibility of APAchE II and SAPS II score was 72.90% and 66.70% respectively. SAPS II score was found to be more specific (85.40%) than APAchE II (80.03%). Conclusion: those severity patient scores accurately predicted the prognosis of patients in intensive care unit and should be integrated into our practice.
