Molecular phylogeny of Osmanthus(Oleaceae) based on non-coding chloroplast and nuclear ribosomal internal transcribed spacer regions

The phylogenetic relationships of Osmanthus *** investigated using the nuclear ribosomal internal transcribed spacer(ITS) regions and non-coding chloroplast regions(psbA-trnH,trnL-F).The two datasets support the conclusion that Osmanthus is polyphyletic,with some species of the subtribe Oleinae nested within *** didymopetalus *** is nested within the clade formed by species of section Osmanthus in two *** attenuatus ***,*** ***,and *** *** of traditional section Osmanthus are clearly divergent from other accessions,and do not form a monophyletic group with other Osmanthus *** marginatus *** embedded in the clade formed by species of section Osmanthus in the ITS *** cpDNA trees all species of section Osmanthus are placed in the large clade and all species of section Leiolea formed a *** taxonomic incongruence among trees for ITS and cpDNA indicate hybridization,as introgression may have occurred among some species of sections Osmanthus and *** of Osmanthus is discussed in light of molecular and morphological data,and a revised infrageneric classification with three sections(Leiolea,Siphosmanthu,and Osmanthus) is *** section Linocieroides is abandoned and united with section Osmanthus.

In this study,we aimed to study the phylogeographic pattern of Juniperus sabina,a shrub species commonly occurring in the northern,northwestern and western China. We sequenced three chloroplast DNA fragments (trnL-trnF,trnS-trnG,and trnD-trnT) for 137 individuals from 16 populations of this species. Five chloroplast DNA chlorotypes (A,B,C,D,and E) were identified and they showed no overlapping distribution. The population subdivision is very high (GST=0.926,NST=0.980),suggesting a distinct phylogeographic structure (NST>GST,Pallopatric differentiations between regional populations and independent glacial refugia for postglacial recolonization. The deserts that developed during the late Quaternary might have acted as effective barriers to promote genetic differentiation among these regions. However,the low diversity dominated by the single chlorotype within each fragmented region suggested that all current populations were derived from a common regional range expansion.

The Tibetan antel.pe(chiru,Pantholops hodgsoni),a heavily poached species and symbol of the QinghaiTibetan Plateau(QTP),is noted worldwide for its special calving *** species originated in the early Quaternary and it is interesting to know how the following climatic oscillations affected its demographic dynamics in the climate-sensitive *** this study,we analyzed the mitochondrial D-loop region from 312 individuals sampled in all of the six major *** found high rates of gene flow and little genetic differentiation between populations,suggesting that the calving migration may have homogenized the genetic pool of this *** mismatch distribution analyses and coalescent simulations suggested that this species experienced a demographic expansion approximatel. 600-200 Kyr following the retreat of the large glaciers developed in the QTP at 800-600 Kyr,rather than at the end of the last glacial age,as previously suggested,based on a limited sample *** addition,we found evidence of a chiru population decrease probably related to the human settings at the QTP during the middle Holocene.

Poly(ethylene oxide)-graft-methotrexate Macromolecular Drugs Conjugating via Aminopteridine Ring Exhibit Potent Anticancer Activity

Water soluble poly(ethylene oxide)-graft-methotrexate (PEO-g-MTX) conjugates with a robust amide linkage via the amine or carboxylic acid groups of MTX were designed,prepared and investigated for in vitro anti-tumor *** was conjugated to multi-functional PEO containing multiple pendant carboxylic acid (PEO-g-COOH)or amine groups (PEO-g-NH2) via the carbodiimide chemistry,which afforded PEO-g-MTX conjugates with an amide bond to the aminopteridine ring or carboxylic acid groups of MTX (denoted as PEO-g-MTX(COOH) and PEO-g-MTX(NH2),respectively).Dynamic light scattering (DLS) revealed that all PEO-g-MTX conjugates,with MTX contents varying from 4.8 to 19.6 wt％,existed as unimers in phosphate buffer (PB,pH 7.4,20 mmol·L-1).Interestingly,MTT assays showed that PEO-g-MTX(COOH) exhibited potent anti-tumor activity in HeLa,A549,KB and NIH3T3 cells with cytotoxicity profiles comparable to that of free *** contrast,PEO-g-MTX(NH2)revealed diminishing cytostatic effect with IC50 (half maximal inhibitory concentration) ten to hundred times higher than that of PEO-g-MTX(COOH).Moreover,PEO-g-MTX(COOH) conjugates allowed facile conjugation with targeting ***,folate-decorated PEO-g-MTX(COOH) macromolecular drugs showed apparent targetability to folate receptor-overexpressing KB cells with an IC50 over 12-fold lower than non-targeting PEO-g-MTX-(COOH) control and about 2-fold lower than free MTX under otherwise the same conditions.
