Comparative analysis of real-world data of frequent treatment sequences in metastatic prostate cancer

Background:The incidence of prostate cancer is increasing worldwide.A significant proportion of patients developmetastatic disease and are initially prescribed androgen deprivation therapy(ADT).However,subsequent sequences of treatments in real-world settings that may improve overall survival remain an area of active *** and methods:Data were collected from 384 patients presenting with de novo metastatic prostate cancer from 2011 to 2015 at a tertiary cancer *** were categorized into surviving(n=232)and deceased(n=152)groups at the end of 3 *** sequence pattern mining techniques(Generalized Sequential Pattern Mining and Sequential Pattern Discovery using Equivalence Classes)were applied to determine the exact order of the most frequent sets of treatments in each ***:Degarelix,as the initial form of ADT,was uniquely in the surviving *** sequence of ADT followed by abiraterone and docetaxel was uniquely associated with a higher 3-year overall *** followed by fosfestrol was found to have a unique niche among surviving patients with a long duration of response to the initial *** who received chemotherapy followed by radiotherapy and those who received radiotherapy followed by chemotherapy were found more frequently in the deceased ***:We identified unique treatment sequences among surviving and deceased patients at the end of 3 *** should be the preferred formof *** who received ADT followed by abiraterone and chemotherapy showed better *** requiring palliative radiation and chemotherapy in any sequence were significantlymore frequent in the deceased group,identifying the need to offer such patients the most efficacious agents and to target them in clinical trial design.

Modulating Tankyrases(TNKS),interactions with USP25 to promote TNKS degradation,rather than inhibiting their enzymatic activities,is emerging as an alternative/specific approach to inhibit the Wnt/β-catenin ***,we identified UAT-B,a novel neoantimycin analog isolated from Streptomyces conglobatus,as a small-molecule inhibitor of TNKS-USP25 protein-protein interaction(PPI)to overcome multi-drug resistance in colorectal cancer(CRC).The disruption of TNKS-USP25 complex formation by UAT-B led to a significant decrease in TNKS levels,triggering cell apoptosis through modulation of the Wnt/β-catenin ***,UAT-B successfully inhibited the CRC cells growth that harbored high TNKS levels,as demonstrated in various in vitro and in vivo studies utilizing cell line-based and patient-derived xenografts,as well as APC^(min/+)spontaneous CRC ***,these findings suggest that targeting the TNKS-USP25 PPI using a small-molecule inhibitor represents a compelling therapeutic strategy for CRC treatment,and UAT-B emerges as a promising candidate for further preclinical and clinical investigations.

Analysis of photovoltaic power plant(PVPP)operations is important to both investors and users and to producers of equipment,researchers and designers as *** paper presents the analysis of the year-long operations of the group PVPPs in Serbia connected to the distribution network which was carried out through specific indicators such as electric power generation,power,capacity factors,specific yield,losses,period of cost effectiveness of the investment,*** attention has been paid to the influence of the azimuth and tilt of the photovoltaic modules on the performances of *** obtained through the year-long extensive measurements were compared to expected yields of PVPPs on the basis of the Climate SAF PVGIS *** results of the analysis indicate that available software applications cannot always provide a good enough forecast of expected electric power production since they cannot always assess well enough the effects of the surrounding facilities on operations of a *** addition,it has been found that the correlation between losses of annual achieved specific yield of a PVPP and the azimuth is represented by a polynomial of the second degree,whereas the correlation between losses of the annual achieved specific yield of a PVPP and the tilt is represented by an exponential function.

Collecting silver artefacts has traditionally been a very popular *** is addictive,therefore the number of potential collectors and investors appears to grow each ***,increases in the interest and buying potentials resulted in a number of forgeries manufactured and introduced to the open antique *** items such as early silver candlesticks dictate a very high price,for many high quality fakes show very good appearances and matching similarities with *** copies are traditionally manufactured by casting using the original items as *** details and variances in design features,position and shape of hallmarks,including the final surface quality are usual features to distinguish the fakes from the *** paper presents results of a study conducted on several silver candlesticks,including two artefacts bearing features of those produced in the mid 18th century,one original Italian candelabrum from Fascist era,and small candlesticks made in the early 20th ***,the paper presents some interesting contemporary coins-replicas of many those produced in different *** coins were offered for sale by unscrupulous dealers via auctions and *** the main results and findings from this study are discussed from a manufacturing point of view,such as fabrication technology,surface quality and hallmarks,which will help the collectors,dealers and investors to detect and avoid forgeries.

CAPTURING COMPLEX,NON-LINEAR TEAM BEHAVIOURS DURING COMPETITIVE FOOTBALL PERFORMANCE

This study investigated changes in the complexity （magnitude and structure of variability） of the collective behaviours of association football teams during competitive performance. Raw positional data from an entire competitive match between two professional teams were obtained with the ProZone tracking system. Five compound positional variables were used to investigate the collective patterns of performance of each team including： surface and geometrical centre. Analyses involve the coefficient （ApEn）, as well as the linear association between both area, stretch index, team length, team width, of variation （%CV） and approximate entropy parameters. Collective measures successfully captured the idiosyncratic behaviours of each team and their variations across the six time periods of the match. Key events such as goals scored and game breaks （such as half time and full time） seemed to influence the collective patterns of performance. While ApEn values significantly decreased during each half, the %CV increased. Teams seem to become more regular and predictable, but with increased magnitudes of variation in their organisational shape over the natural course of a match.

Rice bran polysaccharide ameliorates ovalbumin-induced allergy and improves gut microbiota composition

This study is the first investigation of the effects of polysaccharide extracted from rice bran (RBP) on ovalbumin (OVA)-induced allergy and the gut microbiota， which hasn't been reported previously. RBP in the medium-dose was found to significantly reduce the symptoms of food allergy (FA) in mice， lower the levels of histamine and allergen-specific (IgE)， and attenuate intestinal inflammation. Further studies demonstrated that RBP decreased allergy-associated responses in intestinal epithelial cells and Th2 cells while regulating Th1/Th2 differentiation to reach a dynamic equilibrium and downregulating Th2 polarization caused by allergies. Short-chain fatty acid (SCFAs) content and 16S rRNA sequencing revealed that RBP enhanced the abundance and diversity of the gut microflora and raised the content of SCFAs. These results suggest that RBP may mitigate FA by modulating the gut microbiota. This study revealed the effective use of rice bran and served as a basis for the development of innovative functional foods with anti-allergic properties.

Effective engineering asset management(EAM) is critical to economic development and improving livability in society, but its complexity often impedes optimal asset functionalities. Digital twins(DTs) could revolutionize the EAM paradigm by bidirectionally linking the physical and digital worlds in real *** is great industrial and academic interest in DTs for EAM. However, previous review studies have predominately focused on technical aspects using limited life-cycle perspectives, failing to holistically synthesize DTs for EAM from the managerial point of view. Based on a systematic literature review,we introduce an analytical framework for describing DTs for EAM, which encompasses three levels: DT1.0 for technical EAM, DT 2.0 for technical-human EAM, and DT 3.0 for technical-environmental EAM. Using this framework, we identify what is known, what is unknown, and future directions at each level. DT 1.0 addresses issues of asset quality, progress, and cost management, generating technical *** lacks multi-objective self-adaptive EAM, however, and suffers from high application cost. It is imperative to enable closed-loop EAM in order to provide various functional services with affordable DT 1.0. DT2.0 accommodates issues of human-machine symbiosis, safety, and flexibility management, generating managerial value beyond the technical performance improvement of engineering assets. However, DT 2.0currently lacks the automation and security of human-machine interactions and the managerial value related to humans is not prominent enough. Future research needs to align technical and managerial value with highly automated and secure DT 2.0. DT 3.0 covers issues of participatory governance, organization management, sustainable development, and resilience enhancement, generating macro social value.
