Roles of Aqueous Extract of Marigold on Arsenic-Induced Oxidative Damage in Pancreatic Islet β-Cells

Roles of Marigold extracts (ME) on arsenic trioxide (ATO)-induced oxidative damage to pancreatic β-cells need to be further elucidated. In this study, NIT-1 cells were treated with different concentrations of and/or ATO, following by the cell viability was detected by CCK8 assay. Then, intracellular reactive oxygen species (ROS) levels, lipid peroxide (MDA) contents and superoxide dismutase (SOD) activity were measured with a fluorescence probe method and colorimetric assay, respectively. The apoptosis rate and morphology was detected and observed with hoechst 33,258 staining assay. The mRNA levels and protein expressions of nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) were measured by real-time fluorescence quantitative polymerase chain reaction and protein immunoblotting assay, respectively. Our results indicated that Co-treatment with ME and ATO exacerbated the cell viability decreasing reduced by ATO, while the addition of ME after ATO treatment effectively promote the recovery of ATO reduced survival rates. The ATO group increased apoptosis (P P β-cells by modulating the activation of the Nrf2 signaling pathway.

Clinical studies in COVID-related olfactory disorders:Review of an institutional experience

Objective:To share a single institutional experience with clinical research on COVID-related olfactory dysfunction(OD).Data Source/Method:Narrative review of published original data and ongoing clinical trials on COVID-related OD at Washington University from 2020 to 2023.Results:There were three new diagnostic-/patient-reported outcome measures developed and tested.We report five clinical trials of interventions for COVID-related olfactory disorders:combined Visual-Olfactory training(VOLT)with patient-preferred scents versus standard olfactory training(VOLT trial),oral gabapentin versus placebo(Gabapentin for the Relief of Acquired Chemosensory Experience trial),nasal theophylline irrigations versus placebo(Smell Changes and Efficacy of Nasal Theophylline trial),stellate ganglion block(single-arm),and mindfulness-based stress reduction(MBSR)versus lifestyle intervention(MBSR trial).Conclusions:Initial intervention trials for COVID-related OD have shown potential for improving subjective and objective olfactory outcomes.However,there remains no gold standard treatment that definitively outperforms placebo in controlled trials.Therefore,continued investigation of novel therapeutic strategies for COVID-related OD is necessary to maximize olfactory outcomes for affected patients.

Background:Lung Adenocarcinoma(LUAD)is the leading cause of death from lung cancer.Cuproptosis is the latest discovered way of programmed cell death,and Cuproptosis-Related Gene(CRG)is associated with the risk of LUAD.At present,there are few research of LUAD and Cuproptosis focuses on Long non-coding RNA(LncRNA).As genomics advances,LncRNA emerges as a potential target for understanding tumor progression and prognosis,offering prospects for biological targeted therapy.Therefore,this study provides new biomarkers and therapeutic targets for LUAD from the perspective of LncRNA.Methods:Gene expression,clinical outcome and gene mutation data of LUAD patients were downloaded from TCGA database.Spearman correlation was used to analyze the correlation between LncRNA and CRG.Univariate Cox,multivariate Cox and LASSO Cox regression analysis were used to construct a prognostic model of Cuproptosis-LncRNAs.GO and KEGG enrichment and immune function analysis were performed on differentially expressed genes between different risk groups.Then,immune escape analysis was performed on LUAD patients with different TIDE score.Finally,drug sensitivity analysis was performed on these differentially expressed genes.Results:A total of 2244 Cuproptosis-LncRNAs were found.Through the application of univariate Cox regression analysis,multivariate Cox regression analysis,and LASSO Cox regression analysis,a prognostic model was developed,integrating 15 Cuproptosis-LncRNAs to assess the risk of mortality.Following that,the model underwent assessment through risk score analysis,Kaplan-Meier survival analysis,risk distribution,and evaluation of survival outcomes.The results revealed an AUC value of 0.755 for the model,surpassing the AUC of other clinical pathological features.The results of KEGG analysis showed that the differentially expressed genes in different model groups were mainly involved in Amoebiasis,Fat digestion and absorption,and other signaling pathways.The results of TMB showed that the prognostic model of TMB combined with ris

Introduction: Sleep disorders among professional drivers are a major road safety problem. They cause fatigue and drowsiness at the wheel, which can lead to road traffic accidents and even accidents at work. The aim of this study was to assess sleep disorders among professional drivers of oil tankers in Ouagadougou, Burkina Faso. Methods: This was a descriptive and analytical cross-sectional study carried out from 11 April 2020 to 11 September 2020 in the national hydrocarbon company’s fuel depot. All the drivers present at the time were included and accomplished a questionnaire incorporating the commonly scales using to assess sleep disorders, sleep quality and sleep apnoea. Data analysis using R 3.6.1 software enabled to perform univariate and multivariate analyses to identify associated factors. Adjusted odd ratios were used to measure the strength of association. The significance level chosen was p ≤ 0.05. Results: All the 339 respondents were male. A proportion of 33.6% suffered from insomnia, including 12.7% with moderate to severe insomnia, and 18.58% were at risk of sleep apnoea. On the Epworth scale, 26.6% of drivers showed sleep debt and 4.1% excessive daytime sleepiness. The risk factors associated with insomnia were the use of psychostimulants. A history of diabetes and insomnia were risk factors associated with sleep apnoea syndrome. Conclusion: The prevalence of sleep disturbance and insomnia among the respondents were 52.2% and 33.6% respectively. Use of psychostimulants was the identified risk factor. The prevalence of SAS was 4.72%, with diabetes and insomnia as associated factors.

Carbonaceous aerosols affect air quality adversely,affect global warming,and human health.However,our understanding of the impact of ultrafine(PM_(0.1))carbonaceous particulate matter is incomplete,particularly the effects during haze episodes.This study monitored diurnal variations in PM_(0.1) in Chiang Mai,Thailand,from March to April 2020.We investigated carbonaceous PM_(0.1) collected by an ambient nano-sampler and evaluated their effect by using a carbon analyzer(IMPROVE_TOR).The results showed that burning large open areas in the dry season was crucial for increasing the particle mass concentration because of the large open burnings that occurred in this area.The majority of biomass fires near the sampling site occurred during the night,which would allow more particles to be released thus resulting in higher concentrations of PM_(0.1).Hence,the release of PM_(0.1) during the night would obviously result in higher concentrations than that during the day.In the eight carbon profiles,organic carbon 3(OC3)was predicted to be a marker of biomass fires.The carbon indices displayed that PM_(0.1) was influenced by biomass burning both daytime and nighttime.The findings reported herein should be of great impor-tance in terms of establishing biomass burning control policies for mitigating heavy haze pollution in Thailand and elsewhere.

Low carbon development of blast furnaces is one of the key technological directions in the current development of ironmaking.Owing to the differences in the physical and chemical properties of hydrogen and carbon,hydrogen-rich media entering a blast furnace will change the heat distribution,thus affecting the stability of production.Accordingly,a heat distribution model was proposed to study the temperature distribution in a blast furnace,simultaneously considering gas-solid heat exchange,slag and iron melting,and chemical reactions.The model was used to analyze the temperature distribution of a 2300 m^(3) blast furnace and was verified via comparison with actual production data.Subsequently,the effects of the injection rate of hydrogen-rich media,H2 concentration,and oxygen enrichment rate of the blast on the temperature distribution were investigated.Results indicated that the increase in the injection rate of the hydrogen-rich media decreased the amount of direct reduction and led to an increase in the furnace temperature.Furthermore,an increase in the oxygen enrichment rate led to a decrease in the furnace temperature,but could reduce the solid fuel ratio,while the change in H2 concentration had less effect on the temperature distribution.The combination of hydrogen-rich media injection and the increase in the oxygen enrichment rate would help to adjust the temperature distribution to the same level as the conventional blast furnace conditions.

Capsicum is a nutritious vegetable and its cultivation in farms is getting popular in Bangladesh. Although many efforts have lain to explore better yielding and nutritionally rich cultivars with suitable modern cultivation techniques but still have to find the desired outcome. Thus, it’s necessary to conduct further research to identify the high-yielding and nutritious capsicum cultivars in Bangladesh. An experiment was conducted from July 2021 to June 2022 at the Bangladesh Institute of Research and training on Applied Nutrition (BIRTAN) research field with three cultivars of capsicum: B0 = California Wonder, B1 = BARI Misti Morich-1 and B2 = BARI Misti Morich-2 and three mulching: T0 = No mulching, T1 = Water hyacinth, T2 = Poly Mulching in randomized complete block design with three replications to identify better quality capsicum cultivar and suitable mulching material. Among cultivars the BARI Misti Morich-2 (B2) showed increased agronomic parameters like number of branches and effective branches per plant, leaves length and width, consequently yield and yield contributing traits were also enhanced like fruits per plant, fruit length, fruit diameter and yield per plant (25.97%, 4.54%, 3.64% and 21.43%, respectively). Poly Mulching (T2) increased agronomic traits, yield traits and yield (0.61 kg) than BARI Misti Morich-1 (T1). The combined effect of B2T2 increased the number of branches per plant, effective branches per plant, leaves length and breadth by 40%, 90%, 15.57% and 26.22%, respectively, hence resulting in an increased yield of 20%. BARI Misti Morich-2 cultivar showed an increase in Fe, Zn and Vitamin-C content of 26.24% and 23.10%, 8.82% and 5.14%, and 6.03% and 5.74% than B0 and B1 cultivars, respectively. Therefore, BARI Misti Morich-2 exhibited the improved agronomic, yield and nutritional traits of capsicum under poly mulching among other cultivars in Bangladesh.

Assessment of the Spatio-Temporal Trends of Annual Extreme Temperature Indices over Tanzania during the Period of 1982-2022

Extreme weather and climatic phenomena, such as heatwaves, cold waves, floods and droughts, are expected to become more common and have a significant impact on ecosystems, biodiversity, and society. Devastating disasters are mostly caused by record-breaking extreme events, which are becoming more frequent throughout the world, including Tanzania. A clear global signal of an increase in warm days and nights and a decrease in cold days and nights has been observed. The present study assessed the trends of annual extreme temperature indices during the period of 1982 to 2022 from 29 meteorological stations in which the daily minimum and maximum data were obtained from NASA/POWER. The Mann-Kendall and Sen slope estimator were employed for trend analysis calculation over the study area. The analyzed data have indicated for the most parts, the country has an increase in warm days and nights, extreme warm days and nights and a decrease in cold days and nights, extreme cold days and nights. It has been disclosed that the number of warm nights and days is on the rise, with the number of warm nights trending significantly faster than the number of warm days. The percentile-based extreme temperature indices exhibited more noticeable changes than the absolute extreme temperature indices. Specifically, 66% and 97% of stations demonstrated positive increasing trends in warm days (TX90p) and nights (TN90p), respectively. Conversely, the cold indices demonstrated 41% and 97% negative decreasing trends in TX10p and TN10p, respectively. The results are seemingly consistent with the observed temperature extreme trends in various parts of the world as indicated in IPCC reports.

Peking Union Medical College(PUMC)launched the"4+4"Medical Doctor(MD)pilot program in 2018,admitting students with non-medical backgrounds from top universities,aligning with national medical talent training policies to foster diverse and eager learners in medicine.On the occasion of the graduation of the first class of the"4+4"MD pilot class at PUMC in 2023,we reviewed the teaching reform in the pilot program and carried out a systematic survey and interviews with students,faculties,and management staff of the pilot class.This article reports on the measures taken by the pilot class at PUMC in enrollment and curriculum setting,and demonstrates the achievements of the pilot class in terms of student academic background structure,knowledge acquisition and skill learning,scientific research ability,and course evaluation.The results indicated that the pilot class had met the national demand for the"Medicine+X"talent training model.More specifically,with a diverse academic backgrounds,the pilot class graduates had academic levels comparable to the eight-year medical education graduates,and their scientific research abilities were satisfactory.The pilot program at PUMC will optimize the curriculum setting,strengthen the construction of faculty,learning resources,and teaching facilities,and reform the academic evaluation methods,thus deepening the reform of medical education and improving the"4+4"MD program as a novel medical education model.
