Evaluation of in vitro cell and blood compatibility and in vivo analgesic activity of plant-derived dietary supplements

Objective: In vitro cell and blood compatibility of three dietary supplements, comprised of multiple plant extracts, Pneumo Go(PG), Green active(GA) and Equistasi(Eq), and their main component, the phytocomplex Matrix U.B.ò(Union Bio s.r.l.)(M), were evaluated. Moreover, preliminary in vivo tests were performed on GA in order to assess its ability to reduce pain in an animal ***: Cell compatibility was determined using fibroblasts(NIH3T3) and primary adult human microvascular endothelial cells(HMVECad) and the neutral red uptake test. Blood compatibility was evaluated by analyzing blood parameters after incubation of the products with sodium citrate anticoagulated whole blood. Thrombin time was determined by adding thrombin to aliquots of human plasma containing the samples. Clotting time was revealed by an automatic coagulometer. The in vivo analgesic effect of GA was evaluated in Wistar rats using the formalin ***: M and PG reduced the percentage of viable NIH3T3 cells, indicating their interference in the cell cycle. GA and Eq stimulated fibroblast proliferation and neutralized the toxic effect of M. M and PG reduced HMVECad cell viability. GA and Eq did not affect cell viability as well as negative control. The hemocompatibility tests indicated that all the samples did not interfere with fibrinogen. The in vivo test carried out in male rats showed a significant analgesic effect of GA in all formalin-induced pain ***: No hemotoxicity and good cell compatibility were found for all the tested samples. GA and Eq were the best candidates for further biocompatibility testing. Moreover, GA reduced pain in the animal model.

Probability distributions of arsenic in soil from brownfield sites in Beijing （China）： statistical characterization of the background populations and implications for site assessment studies

A probabilistic analysis was performed on soil arsenic concentration data from 4 brownfield sites at Beijing （Chaoyang and Haidian Districts）, involved in environmental assessment studies. The available data sets were processed to provide a statistical characterization of the background populations and differentiate ＂anomalous data＂ from the natural range of variation of arsenic concentrations in soil. The site-specific background distributions and the existing wide-scale background values defined for the Beijing area were compared, discussing related implications for the definition of metal contamination soil screening levels （ssls） in site assess- ment studies. The statistical analysis of As data sets discriminated site-specific background populations, encompassing 88% to 94% of the sample data, from oufliers values, associated with either subsoil natural enrichments or possible anthropogenic releases. Upper Baseline Concentration （UBC） limits （＋ 2σ level）, including most of the site-specific metal background variability, were derived based on the statistical characterization of the background populations. sites in the Chaoyang south District area had UBC values in the range 10.4-12.6mg·kg^-1. These ranges provide meaningful ssl values to be adopted for As in local site assessment studies. Using the wide-scale background value for the Beijing area would have erroneously classified most of the areas in the subject sites as potentially contaminated.

A Novel Gaussian Extrapolation Approach for 2D Gel Electrophoresis saturated Protein spots

Analysis of images obtained from two-dimensional gel electrophoresis （2D-GE） is a topic of utmost importance in bioinformatics research, since commercial and academic software available currently has proven to be neither completely effective nor fully automatic, often requiring manual revision and refinement of computer generated matches. In this work, we present an effective technique for the detection and the reconstruction of over-saturated protein spots. Firstly, the algorithm reveals overexposed areas, where spots may be truncated, and plateau regions caused by smeared and overlapping spots. Next, it reconstructs the correct distribution of pixel values in these overexposed areas and plateau regions, using a two-dimensional least-squares fitting based on a generalized Gaussian distribution. Pixel correction in saturated and smeared spots allows more accurate quantification, providing more reliable image analysis results. The method is validated for *** exposed 2D-GE images, comparing reconstructed spots with the corresponding non-saturated image, demonstrating that the algorithm enables correct spot quantification.

The Climate Impact of large-scale Distributors:Acting on the supply Chain

This work aims to analyse the actions that companies working in large-scale distribution carry along their value chains to minimise impacts on climate *** operating in this field are aware that it is less effective to act directly on the core processes and need to involve the upstream value chain in their carbon reduction *** businesses,in fact,need to focus on the indirect GHG(Greenhouse Gases)emissions and depend on how suppliers manage their *** this sector,virtuous companies collaborate with their suppliers to get involved in a common path of quantifying and cutting said impacts *** aspect is particularly relevant in the case of large-scale ***,the process is not immediate since the supply chain is usually very dense and diverse,for instance,adopting various approaches that do not always *** any case,the key aspect is mapping these suppliers:one of the tools mostly used for this purpose is the survey,as a quick instrument able to reach hundreds of suppliers at the same time,receiving a fast and standardized response,which can easily be processed to form a comprehensive and harmonized mapping of the results as the first step for the subsequent implementation of mitigation strategies.

Mitigation measures of debris flow and landslide risk carried out in two mountain areas of North-Eastern Italy

The design of remediation works for the mitigation and prevention of the associated risk is needed where these geological hazards affect anthropized areas. remedial measures for landslides commonly include slope reshaping, plumbing, drainage, retaining structures and internal slope reinforcement, while debris flow control works consist in open or closed control structures. The effectiveness of the remedial works implemented must be assessed by evaluating the reduction of the risk over time. The choice of the most appropriate and cost-effective intervention must consider the type of hazard and environmental issues, and selects, wherever possible, naturalistic engineering operations that are consequently implemented according to the environmental regulations or the design and specification standards imposed by the competent public administrations. The mitigation procedures consist of five basic steps:(a) acquisition of the knowledge of the hazard process;(b) risk assessment with identification of possible disaster scenarios;(c) planning and designing of specific remedial measures to reduce and/or eliminate the potential risk;(d) slope monitoring after application of remedial measures,(e) transfer of knowledge to the stakeholders. This paper presents two case studies describing the practice for the design of the mitigation measures adopted for debris flow and active landslide sites in North-Eastern Italy. The first case study is a debris flow site, for which, based on observation of past events and numerical simulations using the software FlOW-2D, the most suitable mitigation measures were found to be the construction of a debris basin, barriers and breakers. The second case study deals with an active landslide threatening a village. Based on the landslide kinematics and the results of numerical simulations performed with the code FlAC, hard engineering remedial works were planned to reduce the driving forces with benching and by increasing the available resisting forc

Nowadays,the interest in hybrid vehicles is constantly increasing,not only in the automotive sector,but also in other transportation systems,to reduce pollution and emissions and to improve the overall efficiency of the *** railway vehicles are typically the most eco-friendly transportation system,since commonly their primary energy source is electricity,they can still gain benefits from hybrid technologies,as many lines worldwide are not *** fact,hybrid solutions allow ICEpowered(internal combustion engine)railway vehicles,such as diesel multiple units(DMUs),to operate in fullelectric mode even when the track lacks *** possibility to switch to full electric mode is of paramount importance when the vehicle runs on urban or underground track sections,where low or zero emission levels are *** conduct the feasibility study of hybridization of an existing DMU vehicle,designed by Blue Engineering s.r.l.,running on the Aosta–Torino Italian railway line,which includes a non-electrified urban track section and an electrified underground *** hybridization is obtained by replacing one of the diesel generators installed on the original vehicle with a battery pack,which ensures the vehicle to operate in full-electric mode to complete its mission *** hybridization is also exploited to implement a regenerative braking strategy,which allows an increase in the energetical efficiency of the vehicle up to 18%.This work shows the sizing of the battery pack based on dynamic simulations performed on the Turin underground track section,and the results demonstrate the feasibility of the hybridization process.
