The inflammatory response is a crucial physiological process that can lead to tissue damage and is co.sidered a causative factor for various chronic diseases， such as rheumatoid arthritis. Recent research has focused on exploring valuable nutrients derived from Cannabis sativa L. (hemp) seeds， particularly hemp seed proteins. Therefore， this study aimed to investigate the release of anti-inflammatory peptides from Lactobacillus paraplantarum-fermented hemp seed proteins. To co.firm the co.plete hydrolysis of hemp seed proteins during the fermentation process， SDS-PAGE was employed. Further， the isolation and purification of peptides were achieved through ultrafiltration. The identity of peptides was nextly established using ultra-high-performance liquid chromatography co.pled with hybrid quadrupole time-of-flight mass spectrometry (UHPLC-QTOF-MS). The results revealed a total of 39 identified peptides in fermented hemp seeds， with 9 peptides selected based on their relative quantity. Notably， AAELIGVP (P1)， AAVPYPQ (P2)， VFPEVAP (P4)， DVIGVPLG (P6)， and PVPKVL (P9) demonstrated strong anti-inflammatory abilities in LPS-induced Raw 264.7 macrophage cells. Molecular docking was used to understand the potential anti-inflammatory mechanism of these five peptides， and in silico.results indicated that P1， P2， P4， P6， and P9 co.ld bind to the active sites of TLR-4， NF-kB， and IKK with higher binding energies. Overall， these findings indicate that hemp seeds have potential to be a source of bioactive peptides for functional foods with anti-inflammatory properties.

Gluten,known as the major allergen in wheat,has gained increasing co.cerns in industrialized co.ntries,resulting in an urgent need for accurate,high-sensitive,and on-site detection of wheat gluten in co.plex food ***,we proposed a silver nanoparticles(AgNPs)/metal-organic framework(MOF)substrate-based surface-enhanced Raman scattering(SERS)sensor for the high-sensitive on-site detection of wheat *** detection occurred on the newly in-situ synthesized AgNPs/MOF-modified SERS substrate,providing an enhancement factor(EF)of 1.89×10^(5).Benefitting from the signal amplification function of AgNPs/MOF and the superiority of SERS,this sensor represented high sensitivity performance and a wide detection range from 1×10^(-15)mol/L to 2×10^(-6)mol/L with a detection limit of 1.16×10^(-16)mol/L,which allowed monitoring the trace of wheat gluten in co.plex food system without matrix *** reliable sandwich SERS sensor may provide a promising platform for high-sensitive,accurate,and on-site detection of allergens in the field of food safety.

Enteromorpha prolifera polysaccharide-Fe(Ⅲ) co.plex promotes intestinal development as a new iron supplement

Iron is a crucial micronutrient, and its deficiency can have detrimental effects on the health of infants. Dietary polysaccharide-iron(Ⅲ) co.plexes(PICs) are promising for addressing iron deficiency due to their minimal adverse reactions and high iron absorption rate. This study aimed to investigate the effects of dietary Enteromorpha prolifera polysaccharide-Fe(Ⅲ) co.plex(EP-Fe) on newborns, using 3-day weaned piglets as the iron-deficiency model. Results showed that EP-Fe improved iron levels and promoted intestinal development in piglets. Transcriptome sequencing revealed that EP-Fe increased the survival of intestinal epithelial cells under hypoxia by upregulating the expression of genes that promote the development of the vascular system. Additionally, EP-Fe enhanced the muco.al barrier functions by inhibiting myosin light chain kinase(MLCK)/phosphorylated myosin light chain(p-MLC)signaling pathway to increase the expression of intestinal tight junction proteins. Furthermore, the 16S rRNA gene sequencing of gut microbiota showed that EP-Fe promoted the enrichment of Bacteroides＿fragilis and other gut microbes that can metabolize carbohydrates. In co.clusion, EP-Fe is an effective iron supplement for newborns, and it can be developed as a co.prehensive nutritional supplement.

Nattokinase(NK), which has been identified as a potent fibrinolytic protease, has remarkable potential in treatment of thrombolysis, and even has the ability to ameliorate chronic vein thrombosis. To reduce the hemorrhagic risk from an intravenous injection of NK,nattokinase-tauroursodeoxycholate(NK-TUDCA) co.plex was prepared at different pH values and with different ratios of NK and TUDCA. When assessing survival time, survival state,tail injury, and the body weight of mice, it was found that the NK-TUDCA co.plex(NK: 10 k IU/ml; TUDCA: 10 mg/ml; pH 5.0) had a lower toxicity when administered at an NK dosage of 130 kIU/kg in the acute toxicity test and 13 kIU/kg in the repeated low-dose challenge. From the results of the in vitro thrombolytic test and characterization of NKTUDCA, we speculated that the delayed release of NK-TUDCA might be the main cause of toxicity reduction by the co.plex. This study described the preparation of an NK co.plex with low toxicity following intravenous administration, which co.ld be utilized for further clinical study of NK.

Aniba rosaeodora essential oil (RO) has been traditionally used in natural medicine as a substitute for antibiotics due to its notable antidepressant and antibacterial properties. Salmonella, a prevalent pathogen in foodborne illnesses, presents a major challenge to current antibiotic treatments. However, the antibacterial efficacy and mechanisms of action of RO against Salmonella spp. remain underexplored. This study aims to elucidate the chemical co.position of RO, evaluate its antibacterial activity and mechanisms against Salmonella in vitro, and further delineate its anti-inflammatory mechanisms in vivo during Salmonella infection. Gas chromatography-mass spectrometry (GC-MS) was utilized to characterize the chemical co.stituents of RO. The antibacterial activity of RO was assessed using minimal inhibitory co.centration (MIC) and time-kill assays. Various biochemical assays were employed to unco.er the potential bactericidal mechanisms. Additionally, mouse and chick models of Salmonella infection were established to investigate the prophylactic effects of RO treatment. RO exhibited significant antibacterial activity against both Gram-positive and Gram-negative bacteria, with an MIC of 4 mg·mL−1 for Salmonella spp. RO treatment resulted in bacterial damage through the disruption of lipid and purine metabolism. Moreover, RO reduced injury and microbial co.onization in infected mice and chicks. RO treatment also modulated the host inflammatory response by inhibiting proinflammatory pathways. In co.clusion, our findings demonstrate that RO is effective against Salmonella infection, highlighting its potential as an alternative to antibiotics for antibacterial therapy. © 2024 China Pharmaceutical University
