A dual-protease-triggered chemiluminescent probe for precise tumor imaging

Imaging detection of interlinked dual proteases is imperative for precise tumor imaging, which remains challenging due to limited modification position of specific substrate and possible steric ***, we have developed a unimolecular chemiluminescent probe(LGP-CL) tandemly activated by two proteases interlinked with liver cancer to achieve precise tumor imaging. Probe LGP-CL consists of a phenoxy-dioxetane scaffold caged by a tripeptide substrate(LGP, leucine-glycine-proline) as the sensing layer, which can be cleaved sequentially by aminopeptidase N(APN) and dipeptidyl peptidase IV(DPPIV)to turn on a strong chemiluminescent signal, and silenced by specific inhibitor of each enzyme, which accounts for an integrated logic gate(AND, OR and INHIBIT). The successful cleavage of dual proteases on the metabolic site depends on the proper structure of the tripeptide substrate, as confirmed by two probes design. Probe LGP-CL(LGP as the substrate) enables the excellent “dual-lock-dual-key” fit with a382-fold enhancement of chemiluminescent emission while no obvious signal is observed by using GPLCL(GPL as the substrate). By virtue of its rapid response(several minutes), high sensitivity and good cell viability, probe LGP-CL has been utilized to evaluate upregulated levels of proteases in vitro and in living systems, especially to distinguish liver tumor cells(Hep G2) from others(LO2, MCF-7, MCF-10a and RAW264.7). Overall, the newly developed CL probe may facilitate rapid investigation into the role played by proteases in liver diseases, enabling timely selection appropriate treatment. Therefore, our work not only sheds light on the rational design of optical probes for dual protease imaging, but provides a promising tool for clinical diagnosis and even drug discovery.

Isothermal reduction kinetics and reduction prediction for iron ore pellets

Iron ore pellets,as one of the main charges of blast furnaces,have a greater impact on the CO_(2)emission reduction and stable operation of blast *** isothermal reduction behavior of the pellets obtained from a Chinese steel plant was studied in the gas mixtures of CO and N_(2).The results showed the reduction process is divided into two *** reduction in the initial stage(time t≤40 min)is cooperatively controlled by internal diffusion and interface chemical reactions with the activation energy of 30.19 and 16.67 kJ/mol,*** controlling step of the reduction in the final stage(t>40 min)is internal diffusion with the activation energy of 34.60 kJ/*** reduction process can be described by two equations obtained from kinetic *** reduction degree can be predicted under different temperatures and time,and the predicted results showed an excellent correlation with the experimental *** reduction mechanisms were confirmed by the analysis of the scanning electron microscope equipped with an energy dispersive spectrometer and optical microscope.

Low Complexity Successive Cancellation List Decoding of U-UV Codes

Constituted by BCH component codes and its ordered statistics decoding(OSD),the successive cancellation list(SCL)decoding of U-UV structural codes can provide competent error-correction performance in the short-to-medium length ***,this list decoding complexity becomes formidable as the decoding output list size *** is primarily incurred by the *** this challenge,this paper proposes the low complexity SCL decoding through reducing the complexity of component code decoding,and pruning the redundant SCL decoding *** the former,an efficient skipping rule is introduced for the OSD so that the higher order decoding can be skipped when they are not possible to provide a more likely codeword *** is further extended to the OSD variant,the box-andmatch algorithm(BMA),in facilitating the component code ***,through estimating the correlation distance lower bounds(CDLBs)of the component code decoding outputs,a path pruning(PP)-SCL decoding is proposed to further facilitate the decoding of U-UV *** particular,its integration with the improved OSD and BMA is *** results show that significant complexity reduction can be ***,the U-UV codes can outperform the cyclic redundancy check(CRC)-polar codes with a similar decoding complexity.

Performance Evaluation of Damaged T-Beam Bridges with External Prestressing Reinforcement Based on Natural Frequencies

As an evaluation index,the natural frequency has the advantages of easy acquisition and quantitative *** this paper,the natural frequency is used to evaluate the performance of external cable reinforced *** examples show that compared with the natural frequencies of first-order modes,the natural frequencies of higher-order modes are more sensitive and can reflect the damage situation and external cable reinforcement effect of T-beam *** damaged bridges,as the damage to the T-beam increases,the natural frequency value of the bridge gradually *** the degree of local damage to the beam reaches 60%,the amplitude of natural frequency change exceeds 10%for the first *** natural frequencies of the firstorder vibration mode and higher-order vibration mode can be selected as indexes for different degrees of the damaged T-beam *** damaged bridges reinforced with external cables,the traditional natural frequency of the first-order vibration mode cannot be used as the index,which is insensitive to changes in prestress of the external *** natural frequencies of higher-order vibration modes can be selected as indexes,which can reflect the reinforcement effect of externally prestressed damaged T-beam bridges,and its numerical value increases with the increase of external prestressed cable force.

Liquid chromatography-tandem mass spectrometry method for the estimation of adefovir in human plasma:Application to a pharmacokinetic study

An analytical method based on solid phase extraction was developed and validated for analysis of adefovir in human plasma. Adefovir-d4 was used as an internal standard and Synergi MAX RP80A （150rnmx4.6mm. 41am） column provided the desired chromatographic separation of compounds followed by detection with mass spectrometry. The method used simple isocratic chromato- graphic condition and mass spectrometric detection in the positive ionization mode. The calibration cuives were linear over the range of 0.50-42.47 ng/mL with the lower limit of quantitation validated at 0.50 ng/mL. Matrix effect was assessed by post-column infusion experiment to monitor phospholipids and post- extraction addition experiment was performed. The degree of matrix effect for adefovir was determined as 7.5% and ion-enhancement in five different lots of human plasma was 7.1% and had no impact on study samples analysis with 4.5 rain run time. The intra- and inter-day precision values were within 7.7% and 7.8%, respectively, for adefovir at the lower limit of quantification level. Validated bioanalytical method was successfully applied to clinical sample analysis.

Impacts of near-M_(s)austempering treatment on microstructure evolution and bainitic transformation kinetics of a medium Mn steel

The microstructure evolution and bainitic transformation of an Fe-0.19C-4.03Mn-1.48Si steel subjected to near-M_(s)austempering treatment were systematically investigated by combining dilatometer,X-ray diffraction,and electron *** additional austempering treatments with isothermal temperatures above M_(s)were used as *** show that the incubation period for the bainitic transformation occurs when the medium Mn steel is treated with the austempering temperature above M_(s).However,when subjected to near-M_(s)isothermal treatment,the medium Mn steel does not show an incubation period and has the fastest bainitic transformation ***,the largest volume fraction of bainite with a value of 74.7%is obtained on the condition of near-M_(s)austempering treatment after cooling to room *** and microstructure evolution analysis indicates that the elimination of the incubation period and the fastest rate of bainitic transformation are related to the preformed *** advent of preformed martensite allows the specimen to generate more bainite in a limited *** bainitic ferrite nucleation at austenite grain boundaries and through autocatalysis at ferrite/austenite interfaces,a model is established to understand the kinetics of bainite formation and it can describe the nucleation rate of bainitic transformation well when compared to the experimental results.

Characterization of petrophysical and seismic properties for CO2 storage with sensitivity analysis

Saline aquifers are considered as highly favored reservoirs for CO2sequestration due to their favorable properties. Understanding the impact of saline aquifer properties on the migration and distribution of CO2plume is crucial. This study focuses on four key parameters—permeability, porosity, formation pressure, and temperature—to characterize the reservoir and analyse the petrophysical and elastic response of CO2. First, we performed reservoir simulations to simulate CO2saturation, using multiple sets of these four parameters to examine their significance on CO2saturation and the plume migration ***, the effect of these parameters on the elastic properties is tested using rock physics *** established a relationship of compressional wave velocity（Vp） and quality factor（Qp） with the four key parameters, and conducted a sensitivity analysis to test their sensitivity to Vpand Qp. Finally, we utilized visco-acoustic wave equation simulated time-lapse seismic data based on the computed Vpand Qpmodels, and analysed the impact of CO2saturation changes on seismic data. As for the above numerical simulations and analysis, we conducted sensitivity analysis using both homogeneous and heterogeneous models. Consistent results are found between homogeneous and heterogeneous models. The permeability is the most sensitive parameter to the CO2saturation, while porosity emerges as the primary factor affecting both Qpand Vp. Both Qpand Vpincrease with the porosity, which contradicts the observations in gas reservoirs. The seismic simulations highlight significant variations in the seismic response to different parameters. We provided analysis for these observations, which serves as a valuable reference for comprehensive CO2integrity analysis, time-lapse monitoring, injection planning and site selection.

The effect of typical seawater cations on the adhesion of mussel protein Mefp-1 based on dissipative quartz crystal microbalance

Mussels are common anchoring organisms that adhere to the surfaces of various substrates with their *** adhesion of mussel to substrates is contingent upon the presence of mussel foot proteins,of which Mytilus edulis foot protein-1(Mefp-1)has been identified as the most abundant *** has been found that lipids are involved in the mussel adhesion process and can facilitate *** this research,the adhesion behavior of Mefp-1 on various substrate surfaces under the effect of typical seawater cations with or without the presence of lipid were investigated using a quartz crystal microbalance with dissipation(QCM-D).Results indicate that the presence of cations Ca^(2+),Mg^(2+),Na^(+),and K^(+)leads to varying degrees of reduction in the adhesion performance of Mefp-1 on different *** degree of this reduction,however,was much alleviated in the presence of palmitic acid,which is involved in the mussel adhesion ***,the involvement of palmitic acid is advantageous for mussel protein adhesion to the substrate surface in the marine *** study illustrated the significant contribution of palmitic acid to mussel adhesion,which can help to better understand biofouling mechanisms and develop biomimetic adhesive materials.
