Metabolome and RNA-seq reveal discrepant metabolism and secretory metabolism profile in skeletal muscle between obese and lean pigs at different ages

Metabolites and metabolism-related gene expression profiles in skeletal muscle change dramatically under obesity, aging and metabolic disease. Since obese and lean pigs are ideal models for metabolic research. Here, we compared metabolome and transcriptome of Longissimus dorsi(LD) muscle between Taoyuan black(TB, obese) and Duroc(lean) pigs at different ages. We defined the “window phase” of intramuscular fat(IMF) deposition in TB pig, which has significantly higher IMF than Duroc pig. Our results displayed discrepant lipid composition and different expression genes(DEGs) enriched in lipid metabolism, and both metabolome and transcriptome analyses revealed stronger energy expenditure and more active amino acid and protein metabolism in Duroc pig. 10 up-and 51 down-regulated biomarker metabolites with age-and breed-specificity were identified. Potential secretory metabolites, including organic acid(fumaric acid, succinate,malic acid, and gamma-aminobutyric acid), amino acid(L-lysine, and L-glutamic acid), lipid(2-hydroxyisovaleric acid, and L-carnitine) were demonstrated a significant correlation with IMF deposition. Our research highlights the huge difference of metabolic spectrum in skeletal muscle between obese and lean model and muscle-derived secretory metabolites might act as an ambassador of intercellular communication to regulate systematic metabolism.

Exploring the spatiotemporal heterogeneity of vegetation changes in subtropical humid karst region under CO2 fertilization

Terrestrial ecosystems heavily depend on vegetation,which responds to carbon dioxide （CO2） fertilization in hot and humid *** subtropical humid karst region is a hot and humid region;whether and to what extent CO2fertilization affects vegetation changes in such regions remains *** this study,we investigated the degree to which CO2fertilization influences vegetation changes,along with their spatial and temporal differences,in the subtropical humid karst region using time-lag effect analysis,a random forest model,and multiple regression *** showed that CO2fertilization plays an important role in vegetation changes,exhibiting clear spatial variations across different geomorphological zones,with its degree of influence ranging mainly between 11%and 25%.The highest contribution of CO2fertilization was observed in the karst basin and non-karstic region,whereas the lowest contribution was found in the karst plateau *** studies have primarily attributed vegetation changes in subtropical humid karst region to ecological engineering,leading to an overestimation of its contribution to these *** findings of this study enhance the understanding of the mechanism of vegetation changes in humid karst region and provide theoretical and practical insights for ecological and environmental protection in these regions.

Evaluating the impacts of converting grain to vegetable fields on nitrate transport in the deep vadose zone of the North China Plain

Nitrate （NO3-） accumulation and transport processes in the thick vadose zone affect the evolution of the groundwater NO3-content in intensive agricultural *** land-use change （ALUC）,typically accompanied by substantial alterations in nitrogen fertilizer application and irrigation practices,is an important influencing *** study evaluated the changes in NO3-accumulation and transport in the deep vadose zone （DVZ,below the root zone）,and the groundwater NO3-content associated with ALUC from grain to vegetable fields in the North China Plain （NCP）.The ALUC from grain to vegetable resulted in nitrate–nitrogen （NO3--N） accumulation in DVZ increased by 235.5 kg ha-1m-1（163.2%） in the piedmont plain and 224.9 kg ha-1m-1（102.7%） in the central plain,*** change accelerated downward transport velocity in the DVZ (from 0.81±0.47 to 0.89±0.55 m yr-1in the piedmont plain,and from 0.24±0.12 to 0.92±0.12 m yr-1in the central plain) and increased NO3-leaching *** transport velocity and leaching fluxes resulted in chemical N-fertilizer entering the aquifer in several areas in the piedmont *** impact of the agricultural activity intensity changes,accompanied by the ALUC,on groundwater quantity and quality should be considered in similar regions.

Enteromorpha prolifera polysaccharide-Fe(Ⅲ) complex promotes intestinal development as a new iron supplement

Iron is a crucial micronutrient, and its deficiency can have detrimental effects on the health of infants. Dietary polysaccharide-iron(Ⅲ) complexes(PICs) are promising for addressing iron deficiency due to their minimal adverse reactions and high iron absorption rate. This study aimed to investigate the effects of dietary Enteromorpha prolifera polysaccharide-Fe(Ⅲ) complex(EP-Fe) on newborns, using 3-day weaned piglets as the iron-deficiency model. Results showed that EP-Fe improved iron levels and promoted intestinal development in piglets. Transcriptome sequencing revealed that EP-Fe increased the survival of intestinal epithelial cells under hypoxia by upregulating the expression of genes that promote the development of the vascular system. Additionally, EP-Fe enhanced the mucosal barrier functions by inhibiting myosin light chain kinase(MLCK)/phosphorylated myosin light chain(p-MLC) signaling pathway to increase the expression of intestinal tight junction proteins. Furthermore, the 16S r RNA gene sequencing of gut microbiota showed that EP-Fe promoted the enrichment of Bacteroides＿fragilis and other gut microbes that can metabolize carbohydrates. In conclusion, EP-Fe is an effective iron supplement for newborns, and it can be developed as a comprehensive nutritional supplement.

Contribution of atmospheric nitrogen deposition to diffuse pollution in a typical hilly red soil catchment in southern China

Atmospheric nitrogen （hi） deposition is currently high and meanwhile diffuse N pollution is also serious in China. The correlation between N deposition and riverine N export and the contribution of N deposition to riverine N export were investigated in a typical hilly red soil catchment in southern China over a two-year period. N deposition was as high as 26.1 to 55.8 kg N/（ha-yr） across different land uses in the studied catchment, while the riverine N exports ranged from 7.2 to 9.6 kg N/（ha-yr） in the forest sub-catchment and 27.4 to 30.3 kg N/（***） in the agricultural sub-catchment. The correlations between both wet N deposition and riverine N export and precipitation were highly positive, and so were the correlations between NH-N or NO2-N wet deposition and riverine NH4-N or NO3-N exports except for NH-N in the agricultural sub-catchment, indicating that N deposition contributed to riverine N export. The monthly export coefficients of atmospheric deposited N from land to river in the forest sub-catchment （with a mean of 14%） presented a significant positive correlation With precipitation, while the monthly contributions of atmospheric deposition to riverine N export （with a mean of 18.7% in the agricultural sub-catchment and a mean of 21.0% in the whole catchment） were significantly and negatively correlated with precipitation. The relatively high contribution of N deposition to diffuse N pollution in the catchment suggests that efforts should be done to control anthropogenic reactive N emissions to the atmosphere in hilly red soil regions in southern China.

Effects of Land Cover on Soil Organic Carbon Stock in a Karst Landscape with Discontinuous Soil Distribution

Land cover type is critical for soil organic carbon （SOC） stocks in territorial ecosystems. However, impacts of land cover on SOC stocks in a karst landscape are not fully understood due to discontinuous soil distribution. In this study, considering soil distribution, SOC content and density were investigated along positive successional stages （cropland, plantation, grassland, scrubland, secondary forest, and primary forest） to determine the effects of land cover type on SOC stocks in a subtropical karst area. The proportion of continuous soil on the ground surface under different land cover types ranged between 0.0% and 79.8%. As land cover types changed across the positive successional stages, SOC content in both the 0-20 cm and 20-50 cm soil layers increased significantly. SOC density （SOCD） within O-lOO cm soil depth ranged from 1.45 to 8.72 kg m^-2, and increased from secondary forest to primary forest, plantation, grassland, scrubland, and cropland, due to discontinuous soil distribution. Discontinuous soil distribution had a negative effect on 8OC stocks, highlighting the necessity for accurate determination of soil distribution in karst areas. Generally, ecological restoration had positive impacts on SOC accumulation in karst areas, but this is a slow process. In the short term, the conversion of croplandto grassland was found to be the most efficient way for SOC sequestration.

Variations in seed size and seed mass related to tree growth over 5 years for 23 provenances of Quercus acutissima from across China

The origin of a seed strongly impacts its traits, and both origin and seed traits influence seed germination and seedling development. However, in many instances, this effect on the seedling does not persist into adulthood, and little is known about how seed traits and original environment affect seedling/tree growth over time. In this study, seed size, seed mass, seedling/tree growth and origins were collected and determined for 23 provenances of Quercus acutissima from across China. Origin variables correlated well with seed size and seed mass. In stepwise multiple regressions, a longitudinal aridity index explained 49.2-68.7% of the total variation in seed size and mass, while only seed width was correlated with seedling/tree height (H) and diameter at the ground (D) from seed traits and origins. The total variance in H and D explained by the models decreased over time, for example, the R 2 value of the models for H declined from 0.477 in the first year to 0.224 in the fourth year; no models was significant in the fifth year. These results indicate that seed size, regulated by the longitudinal aridity index strongly impacted seedling and tree growth, but the strength of the influence decreased over time, and disappeared after 4 years.

Genome-Wide Identification of Tomato (Solanum lycopersicum L.) CKX Gene Family and Expression Analysis in the Callus Tissue under Zeatin Treatment

The cytokinin oxidase/dehydrogenase(CKX)enzyme is essential for controlling theﬂuctuating levels of endogen-ous cytokinin(CK)and has a signiﬁcant impact on different aspects of plant growth and ***-less,there is limited knowledge about CKX genes in tomato(Solanum lycopersicum L.).Here we performed genome-wide identiﬁcation and analysis of nine SlCKX family members in tomatoes using bioinformatics *** results revealed that nine SlCKX genes were unevenly distributed onﬁve chromosomes(Chr.1,Chr.4,Chr.8,Chr.10,and Chr.12).The amino acid length,isoelectric points,and molecular weight of the nine SlCKX proteins ranged from 453 to 553,5.77 to 8.59,and 51.661 to 62.494 kD,*** localization analysis indi-cated that SlCKX2 proteins were located in both the vacuole and cytoplasmic matrix;SlCKX3 and SlCKX5 pro-teins were located in the vacuole;and SlCKX1,4,6,7,8,and 9 proteins were located in the cytoplasmic ***,we observed differences in the gene structures and phylogenetic relationships of SlCKX proteins among different ***1-9 were positioned on two out of the three branches of the CKX phylogenetic tree in the multispecies phylogenetic tree construction,revealing their strong conservation within phylogenetic *** patterns of expression of CKX genes were noticed in callus cultures exposed to varying con-centrations of exogenous ZT,suggesting their roles in speciﬁc developmental and physiological functions in the regeneration *** results may facilitate subsequent functional analysis of SlCKX genes and provide valu-able insights for establishing an efﬁcient regeneration system for tomatoes.

Nitrous Oxide Emissions from a Masson Pine Forest Soil in subtropical Central China

The forest ecosystem plays a pivotal role in contributing greenhouse gases to the *** order to characterize the temporal pattern of nitrous oxide(N_2O) emissions and identify the key factors affecting N_2O emissions from a Masson pine forest in a hilly red-soil region in subtropical central China,we measured the N_2O emissions in Jinjing of Hunan Province using the static chambergas chromatographic method for 3 years(2010-2012) and analyzed the relationships between the N_2O fluxes and the environmental *** results revealed that the N_2O fluxes over the 3 years varied from-36.0 to 296.7 μg N m^(-2) h^(-1),averaging 18.4±5.6 μg N m^(-2) h^(-1)(n=3).The average annual N_2O emissions were estimated to be 1.6±0.3 kg N ha^(-1) year^(-1).The N_2O fluxes exhibited clear intra-annual(seasonal) variations as they were higher in summers and lower in *** with other forest observations in the subtropics,N_2O emissions at our site were relatively high,possibly due to the high local dry/wet N deposition,and were mostly sensitive to variations in precipitation and soil ammonium N *** this work,a multiple linear regression model was developed to determine the influence of environmental factors on N_2O emissions,in which a category predictor of "Season" was intentionally used to account for the seasonal variation of the N_2O *** a model explained almost 40%of the total variation in daily N_2O emissions from the Masson pine forest soil studied(P<0.001).
