Nonclassical C-glycosides,distinguished by their unique glycosidic bond connection mode,represent a promising avenue for the development of carbohydrate-based ***,the accessibility of nonclassical C-glycosides hinders broader investigations into their structural features and modes of ***,we present the first example of Pd-catalyzed stereospecific glycosylation of nonclassical anomeric stannanes with aryl or vinyl *** method furnishes desired nonclassical aryl and vinyl C-glycosides in good to excellent yields,while allowing for exclusive control of nonclassical anomeric *** significant note is the demonstration of the generality and practicality of this nonclassical C-glycosylation approach across more than 50 examples,encompassing various protected and unprotected saccharides,deoxy sugars,oligopeptides,and complex ***,biological evaluation indicates that nonclassical C-glycosylation modifications of drug molecules can positively impact their biological ***,extensive computational studies are conducted to elucidate the rationale behind differences in reaction reactivity,unveiling a transmetalation transition state containing silver(Ag)within a six-membered *** its remarkable controllability,predictability,and consistently high chemical selectivity and stereospecificity regarding nonclassical anomeric carbon and Z/E configuration,the method outlined in this study offers a unique solution to the longstanding challenge of accessing nonclassical C-glycosides with exclusive stereocontrol.

Factors associated with coverage of praziquantel for schistosomiasis control in the communitydirect intervention (CDI) approach in Mali (West Africa)

Background:Despite the progress made in the control of Neglected Tropical Diseases(NTD),schistosome and soil-transmitted helminth infections are far from being effectively managed in many parts of the ***,the key element of all control strategies,is faced with some difficulties in terms of access to *** study aims to describe the factors involved in the success or failure of the community-directed intervention(CDI)approach through control programmes,which aims to achieve consistent high coverage at affordable and sustainable costs in endemic ***:The CDI approach was carried out from December 2007 to October 2008 in ten villages of the district of Diéma,*** inclusion,each child part of the study’s sample was interviewed and submitted for a physical *** study focused on:data collection,treatment of the eligible population,evaluation of the treatment coverage,performance of community drug distributors(CDDs),and the involvement and perception of ***:A total of 8,022 eligible people were studied with a mean coverage rate of 76.7%.Using multiple regression,it was determined that receiving praziquantel as treatment was associated with five factors:belonging to the Fulani or Moorish ethnic minority versus the Bambara/Soninke,use of the central versus the house-to-house drug distribution mode,the ratio of the population to the number of CDDs,the lack of supervision and belonging to the age group of 15 years or above(pprogrammes(HIV,tuberculosis)that provide financial incentives for community members discouraged many CDDs(who in most cases are volunteers)to participate in the CDI approach due to a lack of ***:The findings indicate that the success of the CDI approach depends on,amongst other things,the personal characteristics of the respondents,as well as on community factors.

Reliable and non-invasive diagnostic tools are highly valuable for successful therapeutic strategies for the treatment of Alzheimer's disease(AD). The existence of neurofibrillary tangles(NFTs) consisting of tau protein are one kind of the pathological features of AD, and its level of severity is correlated with the stage of ***, no clinically approved positron emission tomography(PET) probe is currently available for selective imaging of neurofibrillary tangles on patients. In this paper, we report our studies on biological characteristics of [18F]-THK523 as a novel tau imaging probe. With low molecular weight, [18F]-THK523 is stable, electrically neutral, lipophilic and non-mass concentration-dependent. Preliminary biological studies have shown the excellent properties of [18F]-THK523 as brain imaging tracer for further research.

Ginkgo biloba extract protects against depression-like behavior in mice through regulating gut microbial bile acid metabolism

Depression is a mental disorder with high morbidity,disability and relapse *** biloba extract(GBE),a traditional Chinese medicine,has a long history of clinical application in the treatment of cerebral and mental disorders,but the key mechanism remains incompletely *** we showed that GEB exerted anti-depressant effect in mice through regulating gut microbial *** protected against unpredictable mild stress(UMS)-induced despair,anxiety-like and social avoidance behavior in mice without sufficient brain *** microbiome transplantation transmitted,while antibiotic cocktail abrogated the protective effect of *** bacterial profiling and metabolomics assay revealed a potential involvement of Parasutterella excrementihominis and the bile acid metabolite ursodeoxycholic acid(UDCA)in the effect of *** administration induced depression-like behavior in ***,these findings suggest that GBE acts on gut microbiome-modulated bile acid metabolism to alleviate stress-induced depression.

Icariin ameliorates behavioral deficits and neuropathology in a mouse model of multiple sclerosis

Multiple sclerosis(MS) is a systemic inflammatory illness of the central nervous system that involves demyelinating lesions in the myelin-rich white matter and pathology in the grey matter. Despite significant advancements in drug research for MS, the disease′s complex pathophysiology makes it difficult to treat the progressive forms of the disease. In this study,we identified a natural flavonoid compound icariin(ICA)as a potent effective agent for MS in ameliorating the deterioration of symptoms including the neurological deficit score and the body weight in a murine experimental autoimmune encephalomyelitis(EAE) model. These improvements were associated with decreased demyelination in the corpus callosum and neuron loss in the hippo-campus and cortex confirmed by immunohistochemistry analysis. Meanwhile, it was observed that the activation of microglia in cerebral cortex and hippocampus were inhibited followed by the neuroinflammatory cytokines downregulation such as IL-1β, IL-6 and TNF-α after ICA treatment, which was probably attributable to the suppression of microglial NLRP3 inflammasome ***, molecular docking also revealed the binding force of ICA to NLRP3 inflammasome protein complexes in vitro. Taken together, our findings have demonstrated that ICA, as pleiotropic agent, prevents EAE-induced MS by improving demyelination and neuron loss, which interferes with the neuroinflammation via microglial NLRP3inflammasome activation.

Cancer stem cells(CSCs)play a pivotal role in tumor initiation,proliferation,metastasis,drug resistance,and ***,targeting CSCs has emerged as a promising avenue for cancer ***,3-phosphoglycerate dehydrogenase(PHGDH)has been identified as being intricately associated with the regulation of numerous cancer stem ***,reports detailing the functional regulators of PHGDH that can mitigate the stemness across cancer types are *** this study,the novel“molecular glue”LXH-3-71 was identified,and it robustly induced degradation of PHGDH,thereby modulating the stemness of colorectal cancer cells(CRCs)both in vitro and in ***,LXH-3-71 was observed to form a dynamic chimera,between PHGDH and the DDB1-CRL E3 *** insights not only elucidate the anti-CSCs mechanism of the lead compound but also suggest that degradation of PHGDH may be a more viable therapeutic strategy than the development of PHGDH ***,compound LXH-3-71 was leveraged as a novel ligand for the DDB1-CRL E3 ligase,facilitating the development of new PROTAC molecules targeting EGFR and CDK4 degradation.

Diagnostic delay and associated factors among patients with pulmonary tuberculosis in Dar es Salaam,Tanzania

Background:Tanzania is among the 30 countries with the highest tuberculosis(TB)*** TB has a long infectious period,early diagnosis is not only important for reducing transmission,but also for improving treatment *** assessed diagnostic delay and associated factors among infectious TB ***:We interviewed new smear-positive adult pulmonary TB patients enrolled in an ongoing TB cohort study in Dar es Salaam,Tanzania,between November 2013 and June *** patients were interviewed to collect information on socio-demographics,socio-economic status,health-seeking behaviour,and residential *** categorized diagnostic delay into≤3 or>3 *** used logistic regression models to identify risk factors for diagnostic delay,presented as crude(OR)and adjusted Odds Ratios(aOR).We also assessed association between geographical distance(incremental increase of 500 meters between household and the nearest pharmacy)with binary ***:We analysed 513 patients with a median age of 34 years(interquartile range 27-41);353(69%)were ***,444(87%)reported seeking care from health care providers prior to TB diagnosis,of whom 211(48%)sought care>2 *** six(1%)visited traditional healers before TB *** delay was positively associated with absence of chest pain(aOR=7.97,95%confidence intervals[CI]:3.15-20.19;P<0.001),and presence of hemoptysis(aOR=25.37,95%CI:11.15-57.74;P<0.001)and negatively associated with use of medication prior to TB diagnosis(aOR=0.31,95%CI:0.14-0.71;P=0.01).Age,sex,HIV status,education level,household income,and visiting health care facilities(HCFs)were not associated with diagnostic *** living far from pharmacies were less likely to visit a HCF(incremental increase of distance versus visit to any facility:OR=0.51,95%CI:0.28-0.96;P=0.037).Conclusions:TB diagnostic delay was common in Dar es Salaam,and was more likely among patients without prior use of medication and presenting with

Chronic kidney disease severely deteriorates the outcome of gastrointestinal bleeding: A meta-analysis

AIM To understand the influence of chronic kidney disease(CKD) on mortality, need for transfusion and rebleeding in gastrointestinal(GI) bleeding *** A systematic search was conducted in three databases for studies on GI bleeding patients with CKD or endstage renal disease(ESRD) with data on outcomes of mortality, transfusion requirement, rebleeding rate and length of hospitalization(LOH). Calculations were performed with Comprehensive Meta-Analysis software using the random effects model. Heterogeneity was tested by using Cochrane's Q and I2 statistics. Mean difference(MD) and OR(odds ratio) were ***1063 articles(EMBASE: 589; PubM ed: 459; Cochrane: 15) were found in total. 5 retrospective articles and 1 prospective study were available for analysis. These 6 articles contained data on 406035 patients, of whom 51315 had impaired renal function. The analysis showed a higher mortality in the CKD group(OR = 1.786, 95%CI: 1.689-1.888, P < 0.001) and the ESRD group(OR = 2.530, 95%CI: 1.386-4.616, P = 0.002), and a rebleeding rate(OR = 2.510, 95%CI: 1.521-4.144, P < 0.001) in patients with impaired renal function. CKD patients required more unit red blood cell transfusion(MD = 1.863, 95%CI: 0.812-2.915, P < 0.001) and spent more time in hospital(MD = 13.245, 95%CI: 6.886-19.623, P < 0.001) than the *** ESRD increases mortality, need for transfusion, rebleeding rate and LOH among GI bleeding patients. Prospective patient registries and observational clinical trials are crucially needed.

Light-responsive nanomedicine for cancer immunotherapy

Immunotherapy emerged as a paradigm shift in cancer treatments, which can effectively inhibit cancer progression by activating the immune system. Remarkable clinical outcomes have been achieved through recent advances in cancer immunotherapy, including checkpoint blockades, adoptive cellular therapy, cancer vaccine, and tumor microenvironment modulation. However, extending the application of immunotherapy in cancer patients has been limited by the low response rate and side effects such as autoimmune toxicities. With great progress being made in nanotechnology, nanomedicine has been exploited to overcome biological barriers for drug delivery. Given the spatiotemporal control,light-responsive nanomedicine is of great interest in designing precise modality for cancer immunotherapy. Herein, we summarized current research utilizing light-responsive nanoplatforms to enhance checkpoint blockade immunotherapy, facilitate targeted delivery of cancer vaccines, activate immune cell functions, and modulate tumor microenvironment. The clinical translation potential of those designs is highlighted and challenges for the next breakthrough in cancer immunotherapy are discussed.
