An overview of the physiopathology and various treatment strategies for polycystic ovary syndrome

Polycystic ovary syndrome (PCOS), a common endocrine disorder with metabolic and reproductive consequences, affects a significant percentage of women of reproductive age. Because of its complexity and impact on the quality of life, PCOS requires timely diagnosis and screening for complications. The most common clinical manifestations of PCOS are hirsutism, hyperandrogenism, acne, irregular cycles, amenorrhea, and anovulation. Although PCOS has different clinical manifestations in women, hyperandrogenism, and anovulation are the most common symptoms. A good medical history, examination, blood tests, and ultrasound findings facilitate the diagnosis of PCOS. Novel treatments have also undergone clinical trials to improve the symptoms of PCOS. However, comprehensive review articles investigating the different communities of this disease are limited, and this review aimed to collect data on the different etiologies of PCOS along with the latest diagnostic methods and the most common and up-to-date treatments for this disease.

Protective effect of co-enzyme Q10 on testicular tissue and sperm parameters in adult male rats treated with Sunset Yellow FCF

Objective:To determine the protective effect of co-enzyme Q10(CoQ10)on testicular tissue and sperm parameters in male rats treated with SunsetYellow ***:Sixty male Sprague-Dawley rats were randomly divided into 6.groups of the control,CoQ10(10 mg/kg/day),low dose of Sunset Yellow(2.5.mg/kg),high dose of Sunset Yellow(70 mg/kg),low dose of Sunset Yellow(2.5.mg/kg)plus CoQ10,and high dose of Sunset Yellow(70 mg/kg)plus *** drugs were administered via daily oral gavages for 6.*** the end of the experiment,sperm analysis,stereological and histological assessments of the testis were carried ***:The normal morphology(by 4..1%)and progressive spermatozoa(by 74.8%),testicle volume(by {6..4.),lumen volume(by {6..{6.),interstitial tissue volume(by 4..7%),seminiferous tubule volume(by 4..7%),and number of spermatogonia(by 5.4}9%)and Leydig cells(by 70.7%)reduced in the rats that received high doses of Sunset Yellow in comparison to the control ***,all these alterations were recovered by CoQ10 treatment in the CoQ10 plus high dose of Sunset Yellow ***,low doses of Sunset Yellow did not affect different parameters of the testis and ***:CoQ10 could,to some extent,prevent structural changes of the testis induced by the high dose of SunsetYellow.

Effect of Melatonin on Proliferative Activity and Apoptosis in Spermatogenic Cells in Mouse under Chemotherapy

Objective To investigate the possible protective role of melatonin on the proliferative activity, and apoptosis of germ cells in chemotherapy-induced spermiotoxicity. Methods Male adult NMRI mice were divided into four groups. Then each group was divided into two subgroups of a and b. Group A （control）： mice received vehicle （1% ethanol）for 5.d. Group B （busulfan）： mice received a single dose of 2. mg/kg busulfan. Group C （melatonin）： mice received melatonin （10 mg/kg） for 5.d. Group D （busulfan＋melatonin）： mice received a 5.day course of melatonin （10 mg/kg） following an initial dose of busulfan （2. mg/kg）. Evaluations were made using bromodeoxyuridine （BrdU） labeling and in situ TUNEL assay after 5.d （subgroup a） or {6. d （subgroup b）. Results Busulfan-treated mice both in subgroups a and b, showed a significant increase in the number of apoptotic cells （P〈0.01） and decrease in BrdU labeling index （P〈0.01） compared with controls. Melatonin in group C significantly decreased BrdU labeling index compared with the control （P〈0.01）. Melatonin in group D significantly reduced apoptosis rate and increased BrdU labeling index compared with group B. Conclusion Melatonin may have a protective effect against busulfan-induced testicular damage, partly by decreasing of apoptosis and alteration in proliferative activity of germ cells.

Nitric oxide(NO),a member of the reactive nitrogen species family,plays a role in several physiologic processes,including vasculogenesis and angiogenesis,growth and puberty,and senescence and *** plays an important role in the production of ovarian steroids,ovulation,and follicular *** other words,increased activity of nitric oxide synthase(NOS)leads to an increased amount of NO,which triggers production of prostaglandins and inflammatory cascades which facilitate follicular rupture and *** concentration elevation inhibits steroid synthesis in luteal and granulosa *** NO is a major paracrine mediator of various biological processes,as well as a key factor in both the reproductive cycle and embryo implantation,oversynthesis of NO in the uterus results in toxicity and inflammation in epithelial cells and immunorejection of *** the male physiological system,NO synthesized by NOS plays a major role in erectile function and androgen secretion,as well as semen parameters,and oocyte junction to the ***,this supposedly simple molecule is involved in a number of other functions,such as germ cell evolution,connections between sertoli cells and germ cells in the blood-testis barrier,homodynamic contraction,and germ cell ***,NO is considered a key factor in male fertility due to its widespread distribution in both normal and diseased testis *** difference of expression level of NOS in normal and pathological states is a probable cause of fertility destructive processes.

Background:Extensive loss of donor neural stem cell(NSCs)due to ischemic stress and low rate of differentiation at the site of cell graft are two of the major issues that hamper optimal outcome in NSCs transplantation *** that histone deacetylases(HDACs)modulate various cellular processes by deacetylating histones and non-histone proteins,we hypothesized that combined treatment with small molecules,sodium butyrate(NaB;a known HDAC inhibitor)and nicorandil,will enhance the rate neuronal differentiation of NSCs besides their preconditioning to resist oxidative ***:NSCs derived from 14.day old Sprague Dawley rat ganglion eminence were characterized for tri-lineage *** with 1 mM NaB significantly changed their culture characteristics while continuous treatment for 10 days enhanced their neural *** treatment also preconditioned the cells for their resistance to oxidative ***:The highest rate of neural differentiation and preconditioning effect was achieved when the NSCs were treated concomitantly with NaB and *** proliferation assay showed that concomitant treatment with NaB and nicorandil retarded their rate of ***:These data conclude that preconditioning of NSCs with NaB and nicorandil effectively enhances their differentiation capacity besides preconditioning the cells to support their survival under ischemic conditions.
