Actin,more than just a housekeeping protein at the scene of fertilization

Since the first demonstration of sperm entry into the fertilized eggs of Mediterranean sea urchin Paracentrotus lividus by Hertwig(1876),enormous progress and insights have been made on this topic.However,the precise molecular mechanisms underlying fertilization are largely unknown.The two most dramatic changes taking place in the zygote immediately after fertilization are:(i) a sharp increase of intracellular Ca2+ that initiates at the sperm interaction site and traverses the egg cytoplasm as a wave,and(ii) the concomitant dynamic rearrangement of the actin cytoskeleton.Traditionally,this has been studied most extensively in the sea urchin eggs,but another echinoderm,starfish,whose eggs are much bigger and transparent,has facilitated experimental approaches using microinjection and fluorescent imaging methodologies.Thus in starfish,it has been shown that the sperm-induced Ca2+ increase in the fertilized egg can be recapitulated by several Ca2+ -evoking second messengers,namely inositol 1,4,5-trisphosphate(InsP3) ,cyclic ADP-ribose(cADPr) and nicotinic acid adenine dinucleotide phosphate(NAADP) ,which may play distinct roles in the generation and propagation of the Ca2+ waves.Interestingly,it has also been found that the dynamic rearrangement of the actin cytoskeleton in the fertilized eggs plays pivotal roles in guiding monospermic sperm entry and in the fine modulation of the intracellular Ca2+ signaling.As it is well known that Ca2+ regulates the structure of the actin cytoskeleton,our finding that Ca2+ signaling can be reciprocally affected by the state of the actin cytoskeleton raises an intriguing possibility that actin and Ca2+ signaling may form a'positive feedback loop'that accelerates the downstream events of fertilization.Perturbation of the cortical actin networks also inhibits cortical granules exocytosis.Polymerizing actin bundles also compose the'acrosome process,'a tubular structure protruding from the head of fertilizing sperm. The adverse effect of environmental pollution,climate change,and bad lifestyles on human reproductive fitness and health is reported in a large body of literature and is a matter of attention from the scientific community and governments all over the world.Although several reproductive processes from gametogenesis up to embryo and fetal development are affected by a variety of environmental stressors,most of the data available in the literature are related to sperm physiology and quality.1 I read with interests the article by Dai et al.2 that reports an improvement of sperm quality along with environmental quality in Wenzhou,a highly populated area in the south of Zhejiang province,coastal eastern China.

The United Nations(UN)’s call for a decade of“ecosystem restoration”was prompted by the need to address the extensive impact of anthropogenic activities on natural ecosystems.Marine ecosystem restoration is increasingly necessary due to increasing habitat degredation in deep waters(>200 m depth).At these depths,which are far beyond those accessible by divers,only established and emerging robotic platforms such as remotely operated vehicles(ROVs),autonomous underwater vehicles(AUVs),landers,and crawlers can operate through manipulators and multiparametric sensor arrays(e.g.,optoacoustic imaging,omics,and environmental probes).The use of advanced technologies for deep-sea ecosystem restoration can provide:①high-resolution three-dimensional(3D)imaging and acoustic mapping of substrates and key taxa,②physical manipulation of substrates and key taxa,③real-time supervision of remote operations and long-term ecological monitoring,and④the potential to work autonomously.Here,we describe how robotic platforms with in situ manipulation capabilities and payloads of innovative sensors could autonomously conduct active restoration and monitoring across large spatial scales.We expect that these devices will be particularly useful in deep-sea habitats,such as①reef-building cold-water corals,②soft-bottom bamboo corals,and③soft-bottom fishery resources that have already been damaged by offshore industries(i.e.,fishing and oil/gas).

Neurodegenerative diseases are becoming a big challenge for modern society.Neurodegenerative disorders strongly impact on patient and their caregivers.Moreover, since the population is becoming older, these pathologies will deeply influence medical and socio-economic conditions in the next years.Therefore, efforts are needed to find new strategies devoted to define new protocols and identify novel substances able to prevent neurodegeneration or to improve the quality of life of people affected by neurodegenerative diseases(Alzheimer's Disease International, 2019).

Islands are special environments for studying fungi.On the Vivara island,an integral natural reserve,several taxonomic studies have been performed in the last decades but the taxonomic composition of the mycoflora is still not clear.For this reason,here we present an updated checklist of the mycoflora present on the island,part of a more complex work that allowed the mapping of all the living species present in this precious natural park,over the past decade.

The identification of wildlife-vehicle collision hotspots: Citizen science reveals spatial and temporal patterns

Linear infrastructures(e.g.,roads,railways,pipelines,and powerlines)pose a serious threat to wildlife,due to the risk of wildlife-vehicle collisions(roadkills).The placement of mitigation measures,such as crossing structures,should consider species’life cycles and ecological requirements.Such an assessment would require data collection over large areas,which may be possible by employing citizen science.In this study,we aimed to identify spatiotemporal trends of roadkill occurrence using citizen science data from one of the most urbanized and biodiversityrich regions of Italy.Temporal trends were analyzed using generalized additive models,while landscape patterns were assessed by identifying significant thresholds over land cover gradients,related to increases in relative roadkill abundance,by employing threshold indicator taxa analysis.Our approach recorded a total of 529 roadkills,including 33 different species,comprising 13 mammal,10 bird,6 reptile,and 2 amphibian species.Statistical analysis indicated significant temporal trends for the red fox,the European hedgehog,the stone marten and the European badger,with peaks in roadkill occurrence between the winter and spring months.Relative roadkill abundance increased mostly in landscapes with anthropogenic land cover classes,such as complex cultivations,orchards,or urban surfaces.Our results allowed us to develop a map of potential roadkill risk that could assist in planning the placement of mitigation measures.Citizen science contributions from highly populated areas allowed data collection over a large area and a dense road network,and also directly led to the evaluation of management decisional options.
