Transcription factors （TFs） play vital roles in various biological processes by binding to cis-acting elements to control expressions of their target genes. The MYB TF BplMYB46, from Betula platyphylla, is involved in abiotic stress responses and secondary wall deposition. In the present study, we used a TF-centered yeast onehybrid technology （TF-centered YIH） to identify the cis- acting elements bound by BplMYB46. We screened a shortinsert random library and identified three cis-elements bound by BplMYB46： an E-box （CA（A/T/C）（A/G/C）TG） and two novel motifs, a TO-box （T（GIA）TCG（C/G）） and a GT-box （A（G/T）T（AIC）GT（T/G）C）. Chromatin immunoprecipitation （CHIP） and effector-reporter coexpression assays inNicotiana tabacum confirmed binding of BplMYB46 to the TC-box, GT-box, and E-box motifs in the promoters of the phenylalanine ammonia lyase （PAL）, peroxidase （POD）, and superoxide dismutase （SOD） genes, which function in abiotic stress tolerance and secondary wall biosynthesis. This finding improves our understanding of potential regulatory mechanisms in the response to abiotic stress and secondary wall deposition of BplMYB46 in B. platyphylla.

Precise quantifi cation of climate-growth relationships can make a major contribution to scientifi c forest management.However,whether diff erences in the response of growth to climate at diff erent altitudes remains unclear.To answer this,264 trees of Larix kaempferi from 88 plots,representing diff erent altitudinal ranges(1000-2100 m)and tree classes were sampled and used to develop tree-ring chronologies.tree-ring growth(TRG)was either positively(dominant)or negatively(intermediate and suppressed)correlated with climate in diff erent tree classes at diff erent altitudes.TRG was strongly correlated with growing season at low altitudes,but was less sensitive to climate at middle altitudes.It was mainly limited by precipitation and was highly sensitive to climate at low altitudes.Climate-growth relationships at high altitudes were opposite compared to those at low altitudes.TRG of dominant trees was more sensitive to climate change compared to intermediate and suppressed trees.Climate factors(annual temperatures;moisture,the number of frost-free days)had diff erent eff ects on tree-ring growth of diff erent tree classes along altitudinal gradients.It was concluded that the increase in summer temperatures decreased water availability,resulting in a signifi cant decline in growth rates after 2005 at lower altitudes.L.kaempferi is suitable for planting in middle altitudes and dominant trees were the best sampling choice for accurately assessing climate-growth relationships.

Study on the variation in and selection of Fraxinus mandshurica provenances and families in northeast China

Fraxinus mandshurica Rupr.is one of the main afforestation species in northeast China,and there is great demand for improved F.mandshurica varieties.The results of an investigation into and analysis of the growth traits of F.mandshurica provenances and families showed that there were significant differences in different periods.However,variations in growth traits decreased year by year.There was a significant negative correlation between tree heights of the provenances and sunshine hours in their areas of origin.The provenances of Xinglong,Hailin and Wuchang were selected based on the volume of 18-year-old trees.The average genetic gain from the selection of fast-growing provenances was 19.4%.Ten superior fast-growing families were selected.The average volume of the selected families was22.6%,higher than that of all families.The correlation coefficient between heights at 6-year-old and at 18a was 0.838for provenances,and between heights at 4-year-old and at 18-year-old was 0.303 for families.These results indicate that early selection for height in provenances or families could be performed at 6 years or 4 years,respectively.Early selection for DBH and volume in families could start at 8 years.

With embryogenic callus of Larix olgensisis, we investigated the effects of inositol, glutamine, casein hydrolysate, carbohydrate, abscisic acid and silver nitrate concentration on the maturation of the somatic embryo.Three dominant factors emerged, and we developed a response surface model based on the Box-Behnken design.We defined the optimal conditions for the maturation of somatic embryos. The contents of abscisic acid, silver nitrate, sucrose and casein hydrolysis significantly affected the amount of maturing embryos, but inositol, maltose and glutamine had no effect. By establishing a response surface model with multiple factors, we predicted that the optimal number of L. olgensis somatic embryos was 204 ± 4 gon basal medium, containing 18.28 mg Labscisic acid,5.46 mg Lsilver nitrate and 82.67 g Lsucrose. In the verification experiments, the addition of 20 mg Labscisic acid, 5 mg Lsilver nitrate and 80 g Lsucrose to BM yielded an average of 202.06 somatic embryos per gram. These results should guide large-scale breeding of L. olgensis.

The role of plant eIF5A proteins in multiple biological processes, such as protein synthesis regulation, translation elongation, mRNA turnover, programmed cell death and stress tolerance is well known. Toward using these powerful proteins to increase stress tolerance in agricultural plants, in the present study, we cloned and characterized PsneIFSA2 and PsneIFSA4 from young poplar （P. simonii × P. nigra） leaves. The deduced amino acid sequences of PsneIF5A2 and PsneIF5A4 were 98 % similar to each other, and they are orthologs of eIF5A 1 in Arabidopsis. In a subcellular localization analysis, PsneIF5A2 and PsneIF5A4 proteins were localized in the nucleus and cytoplasm, qRT-PCR analysis showed that PsneIF5A2 and PsneIF5A4 were transcribed in poplar flowers, stem, leaves, and roots. In addition, they were also induced by abiotic stresses. Transgenic yeast expressing PsneIF5A2 and PsneIF5A4 had increased salt, heavy metal, osmotic, oxidative tolerance. Our results suggest that PsneIF5A2 and PsneIF5A4 are excellent candidates for genetic engineering to improve salt and heavy metal tolerance in agricultural plants.

This study explores CAMTA genes in the rare and endangered Chinese plant species,Liriodendron chinense.Despite the completion of whole-genome sequencing,the roles of CAMTA genes in calcium regulation and stress responses in this species remain largely unexplored.Within the L.chinense genome,we identiﬁed two CAMTA genes,Lchi09764 and Lchi222536,characterized by four functional domains:CG-1,TIG,ANK repeats,and IQ motifs.Our analyses,including phylogenetic investigations,cis-regulatory element analyses,and chromosomal location studies,aim to elucidate the deﬁning features of CAMTA genes in L.chinense.Applying Weighted Gene Co-Expression Network Analysis(WGCNA),we explored the impact of CAMTA genes on different organs and their regulation under abiotic stress conditions.The identiﬁcation of signiﬁcant gene modules and the prediction of promoter binding sites revealed co-expressed genes associated with CAMTA transcription factors.In summary,this study provides initial insights into CAMTA genes in L.chinense,laying the groundwork for future research on their evolution and biological roles.This knowledge enhancement contributes to a better understanding of plant responses to environmental stress—an essential aspect of plant biology.

Estimating genetic parameters of parental lines through progeny testing and choosing good hybrid parents are important for genetically improving seed orchard trees.In this study, 24 tetraploid progeny seedlings were used as experimental materials, which came from test cross design：six tetraploid Betula platyphylla lines（Q33, Q13, Q103,Q19, Q83 and Q14） as female parents and four individual B. platyphylla diploid lines（F3, F4, F9 and F11） as male parents were crossed. Variance analysis of height, diameter, height-to-diameter ratio, and internodal distance showed that the differences between hybrid combinations reached highly significant levels. Using multi-objective decisionmaking, we performed a comprehensive assessment of the various hybrid combinations. Using a selection rate of 20 % of the standard, five hybrids were selected;their genetic gains in average height, diameter, height-todiameter ratio, and internodal distance were 20.95, 6.07,13.07 and 8.96 %, respectively. We also analyzed the combining ability and genetic parameter effect values of parents and hybrid combinations. The combined analysis revealed that Q13, Q103, Q33 and Q83 were superior females; F3, F4, and F9 were superior males; and F3 × Q13, F4 × Q83 and F9 × Q33 were superior hybrid combinations. The heights and diameters of these progenies were 22.49 and 11.48 % greater than average,respectively.

The breaking of dormancy mediated by reac-tive nitrogen species(RNS)is related to the accumulation of reactive oxygen species(ROS)in germinating embryos but the underlying mechanism is unclear.The objectives of this study were:(1)to explore the relationship between RNS-mediated dormancy release and ROS accumulation in germinating embryos of Sorbus pohuashanensis;and,(2)to investigate the relationships among germination time,ROS metabolism,and endogenous hormone synthesis.We studied the effects of exogenous nitric oxide(NO)donor sodium nitroprusside(SNP),the NO scavenger(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide(PTIO),abscisic acid(ABA),the exogenous ethylene donor ethrel,and the ethylene receptor inhibitor 2,5-norbornadien(NBD)on embryo germination and seedling growth.Embryos were released from dormancy by pretreatment with NO or ethylene, which was related to increased ethylene biosyn-thesis and decreased ABA levels. Breaking of dormancy by SNP was related to increased levels of ethylene, hydrogen peroxide, and glutathione, increased activities of superoxide dismutase and glutathione peroxidase, and decreased levels of ABA, superoxide anions, and malondialdehyde. These effects of nitric oxide were especially significant in seedling hypocotyls and radicles. These results demonstrate that NO can break S. pohuashanensis embryo dormancy by induc-ing ethylene biosynthesis, and that this signalling pathway is closely related to ROS accumulation and the antioxidant defence response.

Genetic variation and superior provenances selection for wood properties of Larix olgensis at four trials

Larix olgensis,one of the most important timber species in northeastern China,is used for paper making and construction.In this study,10 wood properties(wood density,fiber length,fiber width,fiber length-to width ratio,hemicellulose content,cellulose content,holocellulose content,lignin content,ash content,and carbon content)of 10 provenances of L.olgensis planted at sites of CuoHai(CH),JiaGeDaQi(JGDQ),LiangShui(LS),and Mao'erShan(MES)were analyzed.The results of ANOVA showed that almost traits differed significantly among locations and provenances,with a significant interaction effect.Each trait also differed significant among provenances within sites.The phenotypic and genetic coefficient of variation(PCV and GCV)and provenance heritability(H 2)for wood properties ranged from 1.122 to 27.365%,from 0.564%to 21.113%and from 0.332 to 0.996,respectively.A correlation analysis showed that wood density was significantly negatively correlated with cellulose content and holocellulose content at sites CH,JGDQ,and LS,but were significantly positively correlated at site MES.Wood density was significantly negatively correlated with lignin content at CH and JGDQ,but not at LS and MES.Fiber width(FW)was negatively correlated with the ratio of fiber length(FL)to width across sites,and FW and FL/W were all positively correlated with FL.Lignin content was significantly positively correlated with hemicellulose content at site JGDQ and significantly negatively correlated with cellulose content and with holocellulose content at site MES.Interestingly,carbon content was positively correlated with cellulose content and holocellulose content at CH,but negatively correlated with these two traits at site MES.In a correlation analysis of wood properties with geographic,soil and climatic characteristics at the four sites,wood properties were mainly correlated with latitude and altitude of the site and affected by the average annual precipitation and temperature simultaneously.To select superior pulpwood provenances and high carbon storag

Betulin, oleanolic acid, and betulinic acid are naturally occurring pentacyclic triterpenoids that have significant medicinal value. Considerable amounts of these triterpenoids are available in the outer bark of white birch. In this study, we used ultrasound-assisted extraction （UAE） to extract triterpenoids from birch bark rapidly and with high efficiency. Using high performance liquid chro- matography （HPLC）, three types of triterpenoids were separated and detected. We examined the differences among triterpenoids extracted from diploid versus tetra- ploid white birch. Then, we used factor analysis to screen out tetraploid white birches with comprehensively excel- lent performance. The results indicate that the optimum conditions for extraction include the use of ethanol as an extraction solvent, a solid-to-liquid ratio of 0.1 g/10 ml, ultrasonic power set at 100 W, a temperature of 60 ℃ and an extraction time of 15 min. A reversed-phase C18 col- umn （4.6 mm × 250 mm × 5 μm） with a column tem- perature of 30 ℃ and the mobile phase composed of A （acetonitrile） and B （0.1% aqueous phosphoric acid, v/v） at a flow rate of 0.5 ml/min were used, and the detection wavelength was 195 nm. No significant difference wasobserved between diploid and tetraploid white birch in terms of the content of three types of triterpenoids （at a confidence level of 0.05）. As triterpenoid content, height, and DBH （diameter at breast height） are strongly interre- lated, we used factor analysis to evaluate all individuals, and we screened out six plus trees with excellent com- prehensive characters.
