Oral administration of the herbal oligonucleotide XKC-sRNA-h3 prevents angiotensinⅡ-induced hypertension in mice

Hypertension has become a growing public health concern worldwide.In fact,hypertension is commonly associated with increased morbidity and mortality.Currently,oligonucleotide drugs have proven to be promising therapeutic agents for various diseases.In the present study,we aimed to demonstrate that a herbal small RNA(s RNA),XKC-sRNA-h3(B55710460,F221.I000082.B11),exhibits potent antihypertensive effects by targeting angiotensin-converting enzyme(ACE)in mice.When compared with captopril,oral administration of the sphingosine(d18:1)-XKC-s RNA-h3 bencaosome more effectively prevented angiotensin II-induced hypertensive cardiac damage and alleviated kidney injury in mice.Such findings indicated that XKC-sRNA-h3 may be a novel orally available ACE inhibitor type oligonucleotide drug for hypertension.

The mode of scientific thinking is undergoing rapid and profound changes.In the 21st century,macro and micro civilizations go parallel.A systematic and scientific methodology is required for the study of complex things.The thinking mode in modern medicine is gradually shifting from analytical,reductive thinking to holistic and systematic thinking.As such Western medicine and traditional Chinese medicine are gradually approaching the epistemology of health and disease state.The importance of scientific thinking in innovation has been expounded in this study.The development trends in medicine in the current era are analyzed,the importance of systems theory in the study of human bodies is discussed,and a new medical model named Novel Systems Medicine is proposed.

Pneumoconiosis,a fatal lung disease caused by dust inhalation and deposition,is one of the leading occupational diseases worldwide[1].The dust is primarily inorganic particles,such as silica particles,coal dust,and asbestos fibers.If the diameter of dust is less than 5μm,it is defined as‘respirable dust’that can easily reach the distal airways and alveoli[2].

Spatial expression patterns of homeobox(HOX) genes delineate positional identity of primary fibroblasts from different topographic sites. The molecular mechanism underlying the establishing or maintaining of HOX gene expression pattern remains an attractive developmental issue to be addressed. Our previous work suggested a critical role of CTCF/cohesin-mediated higher-order chromatin structure in RA-induced HOXA activation in human teratocarcinoma NT2/D1 cells. This study investigated the recruitment of CTCF and cohesin, and the higher-order chromatin structure of the HOXA locus in fetal lung and adult foreskin fibroblasts, which display complementary HOXA gene expression patterns. Chromatin contacts between the CTCF-binding sites were observed with lower frequency in human foreskin fibroblasts. This observation is consistent with the lower level of cohesin recruitment and 5′ HOXA gene expression in the same cells. We also showed that CTCF-binding site A56(CBSA56) related chromatin structures exhibit the most notable changes in between the two types of cell, and hence may stand for one of the key CTCF-binding sites for cell-type specific chromatin structure organization. Together, these results imply that CTCF/cohesin coordinates HOXA cluster higher-order chromatin structure and expression during development, and provide insight into the relationship between cell-type specific chromatin organization and the spatial collinearity.

Since the recent discovery of cancer cell-intrinsic programmed cell death protein-1(PD-1),the mechanisms that manipulate PD-1 functions in tumor development beyond its immune checkpoint roles have become attractive research topics in oncology.Our previous study validated that PD-1 exists in lung cancer cells and is directly transactivated by p53 in a DNA-binding domain(DBD)acetylation-dependent manner.Here,we report that the carboxyl-terminal domain(CTD)of p53 likewise participates in PD-1 transcriptional regulation in cancer cells under different regulatory mechanisms.By mutating the lysine residues within the CTD to mimic either acetylationdeficient or fully acetylated status,we proved that acetylated CTD dramatically impeded p53-mediated transactivation of PD-1.Furthermore,we identified bromodomain-containing protein 4(BRD4)as a transcriptional coactivator of p53 that facilitates p53-mediated PD-1 transcription.Mechanistically,BRD4 specifically bound to the unacetylated CTD of p53,while CTD acetylation almost completely destroyed the BRD4-p53 interaction and thus led to compromised PD-1 expression.Collectively,this study unveils an alternative mechanism of p53 acetylation-directed PD-1 transcriptional regulation,which would broaden our current understanding of the molecular regulatory network of cancer cell-intrinsic PD-1.

Polypyrimidine Tract-Binding Protein Regulates Enterovirus 71 Translation Through Interaction with the Internal Ribosomal Entry Site

Enterovirus 71(EV71), a major causative agent of hand, foot, and mouth disease, has caused periodic infection outbreaks in children in the Asia–Pacific region. In order to describe the largely unknown life cycle of EV71, the molecular basis of its virus-host interactions must first be determined. The 50 untranslated region of EV71 contains a cloverleaf-like structure and internal ribosomal entry site(IRES), which play an important role in transcription and translation of viral protein. We found that polypyrimidine tract-binding protein 1(PTB) bound to the IRES of EV71. RNA recognition motifs 1 and 2 of PTB were responsible for its binding to the EV71 IRES. Moreover, PTB protein was shuttled from nucleus to cytoplasm after EV71 infection. Additionally, IRES activity and viral protein production were inhibited by PTB knockdown. These results suggest that PTB interacts with the EV71 IRES, and positively regulates viral protein translation.

Cardiac hypertrophy is the strongest predictor of the development of heart failure, and anti-hypertrophic treatment holds the key to improving the clinical syndrome and increasing the survival rates for heart failure. The paraoxonase(PON) gene cluster(PC) protects against atherosclerosis and coronary artery diseases. However, the role of PC in the heart is largely unknown. To evaluate the roles of PC in cardiac hypertrophy, transgenic mice carrying the intact human PON1, PON2, and PON3 genes and their flanking sequences were studied. We demonstrated that the PC transgene(PC-Tg) protected mice from cardiac hypertrophy induced by Ang II; these mice had reduced heart weight/body weight ratios, decreased left ventricular wall thicknesses and increased fractional shortening compared with wild-type(WT) control. The same protective tendency was also observed with an Apoe^(-/-)background. Mechanically, PC-Tg normalized the disequilibrium of matrix metalloproteinases(MMPs)/tissue inhibitors of MMPs(TIMPs) in hypertrophic hearts, which might contribute to the protective role of PC-Tg in cardiac fibrosis and, thus, protect against cardiac remodeling. Taken together, our results identify a novel anti-hypertrophic role for the PON gene cluster, suggesting a possible strategy for the treatment of cardiac hypertrophy through elevating the levels of the PON gene family.

Non-integrating lentiviral vectors based on the minimal S/MAR sequence retain transgene expression in dividing cells

Safe and efficient gene transfer systems are the basis of gene therapy applications. Non-integrating lentiviral(NIL) vectors are among the most promising candidates for gene transfer tools, because they exhibit high transfer efficiency in both dividing and non-dividing cells and do not present a risk of insertional mutagenesis. However, non-integrating lentiviral vectors cannot introduce stable exogenous gene expression to dividing cells, thereby limiting their application. Here, we report the design of a non-integrating lentiviral vector that contains the minimal scaffold/matrix attachment region(S/MAR) sequence(SNIL), and this SNIL vector is able to retain episomal transgene expression in dividing cells. Using SNIL vectors, we detected the expression of the eGFP gene for 61 days in SNIL-transduced stable CHO cells, either with selection or not. In the NIL group without the S/MAR sequence, however, the transduced cells died under selection for the transient expression of NIL vectors. Furthermore,Southern blot assays demonstrated that the SNIL vectors were retained extrachromosomally in the CHO cells. In conclusion,the minimal S/MAR sequence retained the non-integrating lentiviral vectors in dividing cells, which indicates that SNIL vectors have the potential for use as a gene transfer tool.

Pneumoconiosis refers to a spectrum of pulmonary diseases caused by inhalation of mineral dust,usually as the result of certain occupations.The main pathological features include chronic pulmonary inflammation and progressive pulmonary fibrosis,which can eventually lead to death caused by respiratory and/or heart failure.Pneumoconiosis is widespread globally,seriously threatening global public health.Its high incidence and mortality lie in improper occupational protection,and in the lack of early diagnostic methods and effective treatments.This article reviews the epidemiology,safeguard procedures,diagnosis,and treatment of pneumoconiosis,and summarizes recent research advances and future research prospects.
