A Putative Chloroplast-Localized Ca2＋/H＋ Antiporter CCHA1 Is Involved in Calcium and pH Homeostasis and Required for PSII Function in Arabidopsis

Calcium is important for chloroplast, not only in its photosynthetic but also nonphotosynthetic functions. Mul- tiple Ca2＋/H＋ transporters and channels have been described and studied in the plasma membrane and organ- elle membranes of plant cells; however, the molecular identity and physiological roles of chloroplast Ca2＋/H＋ antiporters have remained unknown. Here we report the identification and characterization of a member of the UPFO016 family, CCHA1 （a chloroplast-localized potential Ca2＋/H＋ antiporter）, in Arabidopsis thaliana. We observed that the ccha I mutant plants developed pale green leaves and showed severely stunted growth along with impaired photosystem II （PSII） function. CCHA1 localizes to the chloroplasts, and the levels of the PSII core subunits and the oxygen-evolving complex were significantly decreased in the ccha I mutants compared with the wild type. In high Ca2＋ concentrations, Arabidopsis CCHA1 partially rescued the growth defect of yeast gdtl3 null mutant, which is defective in a Ca2＋/H＋ antiporter. The cchal mutant plants also showed significant sensitivity to high concentrations of CaCI2 and MnCI2, as well as variation in pH. Taken these results together, we propose that CCHA 1 might encode a putative chloroplast-localized Ca2＋/H＋ antiporter with critical functions in the regulation of PSII and in chloroplast Ca2＋ and pH homeostasis in Arabidopsis.

Earliest fossil fruit record of the genus Paliurus(Rhamnaceae) in Eastern Asia

The genus Paliurus is characterized by its distinctive orbicular-winged fruits and a trilocular(or bilocular) ovary. Macrofossil records suggested this genus was distributed widely in North America, Europe, and Asia during the Paleogene and Neogene, before its present limitation to Eurasia. In this study, we describe some Paliurus fossil winged fruits from the lower part of Youganwo Formation(middle Eocene) and Huangniuling Formation(late Eocene) of the Maoming Basin, South China. These fruits are recognized as Paliurus favonii Unger based on a detailed study on the fossil morphology and cuticle characteristics. This represents the earliest occurrence of Paliurus fossil fruits in eastern Asia, and to date is the world's lowest latitude appearance for this genus. The most ancient records of Paliurus throughout the world are from the middle Eocene of North America and the late Eocene of Japan, leading some researchers to conclude the Paliurus may have originated in North America. Molecular data, however, do not support this hypothesis. The present Paliurus fossil fruits provide new evidence for the origin and phytogeographic history of this genus.

Grass phasiRNAs and male fertility

Recent studies have indicated that a special type of small noncoding RNAs, phased small-interfering RNAs(phasiRNAs) play crucial roles in many cellular processes of plant development. PhasiRNAs are generated from long RNA precursors at intervals of 21 or 24 nt in plants, and they are produced from both protein-coding gene and long noncoding RNA genes. Different from those in eudicots, grass phasiRNAs include a special class of small RNAs that are specifically expressed in reproductive *** grass phasiRNAs are associated with gametogenesis, especially with anther development and male fertility. In this review,we summarized current knowledge on these small noncoding RNAs in male germ cells and their possible biological functions and mechanisms in grass species.

Identification of rice cultivars with low brown rice mixed cadmium and lead contents and their interactions with the micronutrients iron,zinc,nickel and manganese

Paddy fields in mining areas are usually co-contaminated by a cocktail of mixed toxic heavy metals （e.g., Cd and Pb in Pb/Zn mines）. However, previous studies on rice cultivars screened for effective metal exclusion have mostly focused on individual metals, and have been conducted under pot-trial or hydroponic solution conditions. This study identified rice cultlvars with both low Cd and Pb accumulation under Cd- and Pb-contaminated field conditions, and the interactions of the toxic elements Cd and Pb with the micronutrient elements Fe, Zn, Mn and Ni were also studied. Among 32 rice cultivars tested, there were significant differences in Cd （0.06-0.59 mg/kg） and Pb （0.25-3.15 mg/kg） levels in their brown rice, and similar results were also found for the micronutrient elements. Significant decreases in concentrations of Fe and Mn were detected with increasing Cd concentrations and a significant elevation in Fe, Mn and Ni with increasing Pb concentrations. A similar result was also shown by Cd and Ni. Three cultivars were identified with a combination of low brown rice Cd and Pb, high micronutrient and grain yield （Wufengyou 2168, Tianyou 196 and Guinongzhan）. Present results suggest that it is possible to breed rice eultivars with low mixed toxic element （Cd, Pb） and high micronutrient contents along with high grain yields, thus ensuring food safety and quality.

Two decades of suspect evidence for adaptive molecular evolution——negative selection confounding positive-selection signals

There has been a large literature in the last two decades affirming adaptive DNA sequence evolution between *** main lines of evidence are from(i) the McDonald-Kreitman(MK) test,which compares divergence and polymorphism data,and(ii) the phylogenetic analysis by maximum likelihood(PAML) test,which analyzes multispecies divergence ***,we apply these two tests concurrently to genomic data of Drosophila and *** our surprise,the> 100 genes identified by the two tests do not overlap beyond random *** the non-concordance could be due to low powers leading to high false negatives,we merge every 20-30 genes into a 'supergene'.At the supergene level,the power of detection is large but the calls still do not *** rule out methodological reasons for the *** particular,extensive simulations fail to find scenarios whereby positive selection can only be detected by either MK or PAML,but not *** molecular evolution is governed by positive and negative selection concurrently,a fundamental assumption for estimating one of these(say,positive selection) is that the other is ***,in a broad survey of primates,birds,Drosophila and Arabidopsis,we found that negative selection rarely stays constant for long in *** a consequence,the variation in negative selection is often misconstrued as a signal of positive *** conclusion,MK,PAML and any method that examines genomic sequence evolution has to explicitly address the variation in negative selection before estimating positive *** a companion study,we propose a possible path forward in two stages—first,by mapping out the changes in negative selection and then using this map to estimate positive *** now,the large literature on positive selection between species has to await reassessment.

Asplenium sanshuiense sp. nov.:the Lowest Latitude Fossil Record of the Genus

Extant Asplenium is one of the most widespread fern groups and occurs in the temperate and tropical regions. However, the fossil records of this genus are poorly documented, especially in the low latitudes. Here, a new species, Asplenium sanshuiense sp. nov. is described from the early Eocene of Sanshui Basin, Guangdong Province, South China. This is the lowest modern latitude fossil record of Asplenium and the first fossil assignment of A. section Darea （Jussieu） Bak., as well as the first fossil record of Asplenium reported from South China. This new species shows that Asplenium had already spread into South China by the early Eocene and the section Darea （Jussieu） Bak. was identifiable within the genus Asplenium during that time. This new species, combined with previous fossil spore-pollen records indicates a warm and humid climate in the Sanshui Basin of the early Eocene.

Bromodomain-containing proteins BRD1, BRD2, and BRD13 are core subunits of SWI/SNF complexes and vital for their genomic targeting in Arabidopsis

Switch defective/sucrose non-fermentable(SWI/SNF)chromatin remodeling complexes are multi-subunit machines that play vital roles in the regulation of chromatin structure and gene ***,the mechanisms by which SWI/SNF complexes recognize their target loci in plants are not fully ***,we show that the Arabidopsis thaliana bromodomain-containing proteins BRD1,BRD2,and BRD13 are core subunits of SWI/SNF complexes and critical for SWI/SNF genomic *** three BRDs interact directly with multiple SWI/SNF subunits,including the BRAHMA(BRM)catalytic *** and transcriptomic analyses of the brd1 brd2 brd13 triple mutant revealed that these BRDs act largely redundantly to control gene expression and developmental processes that are also regulated by ***-wide occupancy profiling demonstrated that these three BRDs extensively colocalize with BRM on *** loss of function of three BRD genes results in reduced BRM protein levels and decreased occupancy of BRM on chromatin across the ***,we demonstrated that the bromodomains of BRDs are essential for genomic targeting of the BRD subunits of SWI/SNF complexes to their target ***,these results demonstrate that BRD1,BRD2,and BRD13 are core subunits of SWI/SNF complexes and reveal their biological roles in facilitating genomic targeting of BRM-containing SWI/SNF complexes in plants.
