Biomonitoring for traditional herbal medicinal products using DNA metabarcoding and single molecule, real-time sequencing

Global concerns have been paid to the potential hazard of traditional herbal medicinal products(THMPs). Substandard and counterfeit THMPs, including traditional chinese patent medicine, health foods, dietary supplements, etc. are potential threats to public health. Recent marketplace studies using DNA barcoding have determined that the current quality control methods are not sufficient for ensuring the presence of authentic herbal ingredients and detection of contaminants/adulterants. An efficient biomonitoring method for THMPs is of great needed. Herein, metabarcoding and single-molecule, realtime(SMRT) sequencing were used to detect the multiple ingredients in Jiuwei Qianghuo Wan(JWQHW), a classical herbal prescription widely used in China for the last 800 years. Reference experimental mixtures and commercial JWQHW products from the marketplace were used to confirm the method. Successful SMRT sequencing results recovered 5416 and 4342 circular-consensus sequencing(CCS) reads belonging to the ITS2 and psb A-trn H regions. The results suggest that with the combination of metabarcoding and SMRT sequencing, it is repeatable, reliable, and sensitive enough to detect species in the THMPs, and the error in SMRT sequencing did not affect the ability to identify multiple prescribed species and several adulterants/contaminants. It has the potential for becoming a valuable tool for the biomonitoring of multi-ingredient THMPs.

O3 is not only greenhouse gas but also a primary gaseous contaminant in the *** has long-lasting effects on crop growth,yield and quality,and brings a series of ecological and environmental problems.A free-air controlled enrichment（FACE） system was applied to study the effect of elevated ozone concentration on activities of key enzymes of starch synthesis of Yangmai 16 in *** main-plot treatment had two levels of O 3 ：ambient level（A-O 3） and 50% higher than ambient level（E-O 3）.The main results were that accumulation rate of amylose,amylopectin and starch were represented in a single peak curve,and their content and accumulation amount rose *** O 3 elevation decreased the accumulation rate of amylose,amylopectin and starch amylase,reduced the accumulation amount of amylopectin and starch,and decreased the content of amylopectin and starch,but increased the content of *** the increase of O 3 concentration,the enzyme activity of grain granule-bound starch synthase（GBSS）,soluble starch synthase（SSS） and starch branching enzyme（SBE） decreased after *** activities of GBSS and SSS had highly significant correlations with amylose,amylopectin and starch accumulation rate,and the activity of SBE had significant correlations with these *** the O 3 elevation decreased the activity of key enzymes of starch synthesis,which led to the variation of starch synthesis.

Impedance Analysis of Forging Process and Strategy Study on Compliance for Forging Manipulator

In the field of heavy forging, there are numerous researches on deformation rule in forging process by FEM simulation, however, not many scholars take the equipment constraint and the mutual reaction load between the forging manipulator clamp and the forging blank into account, which will impact on safety of manipulator body and quality of forging blank. This paper presents an impedance model to describe the load and formulates compliance strategies correspondingly to reduce the mutual reaction load for forging manipulator. Firstly, an FEM model of forging process is built. Meanwhile, the clamp of forging manipulator is added to the model as movement constraint and interaction part between the manipulator and the forming process. Secondly, a typical forging process is simulated by changing the movement constraint, and then an impedance model is established to describe the relationship between the load and movement constraint. Finally, two kinds of compliance strategies are formulated according to the impedance model, one is called free compliance, and the other is initiative/passive compliance. The simulation results show that compliance strategies reduce the load amounting to 5 000 kN in z direction between the manipulator clamp and the forging blank obviously, which may lead to serious accidents, such as the capsizing of forging manipulator, the fracture of manipulator clamp, and so on. The proposed research simulates the more real forging process, gets the initiative/passive compliance strategy which is more simple and suitable to the real producing and better for forming a forging process planning and control system in the modern production, and improves the quality and efficiency of heavy forging.

We demonstrate an all-solid quasi-continuous-wave （QCW） narrow-band source tunable to sodium D2a line at 589.159 nm. The source is based on sum-frequency mixing between lasers at 1064 nm and 1319 nm in a LBO crystal. The 1064 nm and 1319 nm lasers are produced from two diode side-pumped Nd：YAG master oscillator power amplifier （MOPA） laser systems, respectively. A 33 W output of 589 nm laser is obtained with beam quality factor M^2 = 1.25, frequency stability better than ±0.2 GHz and linewidth less than 0.44 GHz. A prototype 589 nm laser system is assembled, and a sodium laser guided star has been successfully observed in the field test.

LSL1 controls cell death and grain production by stabilizing chloroplast in rice

Objective:Taxifolin is a natural flavonoid compound that can be isolated from onions,grapes,oranges and *** also acts as a medicine food homology with extraordinary antioxidant and antiinflammatory *** study aims to explain the protective effects and potential mechanisms of taxifolin against inflammatory ***:Levels of interleukin(IL)-6,IL-1βand intracellular reactive oxygen species(ROS)were assessed in different time after the treatment of taxifolin in RAW264.7 cells induced by lipopolysaccharide(LPS).Subsequently,the m RNA and protein levels of inducible nitric oxide synthase(i NOS),vascular endothelial growth factor(VEGF),cyclooxygenase(COX)-2,tumor necrosis factor(TNF)-a and the phosphorylation expression levels of the MAPK signal pathway were also evaluated.A silico analysis was used to explain the binding situation for the investigation of taxifolin and MAPK signal *** then MAPK inhibitors were used to reveal the expression level of i NOS,VEGF,COX-2 and TNF-a in RAW264.7 ***:It was demonstrated that cell inflammatory damage induced by LPS was significantly alleviated after the treatment of ***,the m RNA and protein levels of i NOS,VEGF,COX-2 and TNF-a were reduced and the phosphorylation expression levels of the MAPK signal pathway were down-regulated remarkably as *** silico analysis,taxifolin could form a relatively stable combination with MAPK signal *** inhibitors showed increasing or decreasing effect in the m RNA levels of i NOS,VEGF,COX-2 and TNF-a,which suggesting that taxifolin down-regulated i NOS,VEGF,COX-2 and TNF-a expressions were not entirely through the MAPK ***:This finding demonstrated that taxifolin improved the inflammatory responses that partly involved in the phosphorylation expression level of MAPK signal pathway in RAW264.7 cells exposed to acute stress.

Omega-3 and omega-6 polyunsaturated fatty acids（PUFAs）were major derived from arachidonic acid（AA）and eicosapentaenoic acid（EPA）,which plays important roles in many physiological processes,especially *** of eicosanoids is important for understanding their homeostatic and pathophysiological roles in inflammatory disease.A new analytical approach focus on polyunsaturated fatty acids metabolome was established using isotope dilution,solid phase extraction and 2D nano liquid chromatography-nanospray ionization-time of flight-mass spectrometry（2D nano LC-NSI-Q-TOF/MS）.Within 35 min,58 PUFAs,including free fatty acids,prostaglandins,lipoxins,resolvins,leukotrienes,thromboxans,maresins,hydroxyeicosatetraenoic acids as well as epoxyeicosatrienoic acids were quantified along with seven corresponding isotope dilution internal *** limits of quantification were between 0.008 and 20 pg per injection for *** efficiency of separation were perfect,all of the standards were separated well（Figure 1）.The method validations for linearity,accuracy,precision,recovery were ***,this approach was performed to analyze both of targeted and non-targeted metabolomics studies in serum samples of women with polycystic ovarian syndrome（PCOS）and health women,the results indicated that the significant differences noted in the levels of heptadecanoic acid,mysristic acid,pentadecanoic acid,9E,11E-octadecadienoic acid,arachidolic acid,palmitic acid,oleic acid and stearic acid（p＜0.001）as well as palmitelaidic acid（p＜0.01）（Figure 1）.Women with PCOS may have a unique metabolomic finger print and a definitive study is *** findings may indicate that the PCOS were positive correlation between patient and health woman in fatty acids syntheses.
