A novel molecular classification method for osteosarcoma based on tumor cell differentiation trajectories

Subclassification of tumors based on molecular features may facilitate therapeutic choice and increase the response rate of cancer ***,the highly complex cell origin involved in osteosarcoma(OS)limits the utility of traditional bulk RNA sequencing for OS ***-cell RNA sequencing(sc RNA-seq)holds great promise for identifying cell ***,this technique has rarely been used in the study of tumor *** analyzing sc RNA-seq data for six conventional OS and nine cancellous bone(CB)samples,we identified 29 clusters in OS and CB samples and discovered three differentiation trajectories from the cancer stem cell(CSC)-like subset,which allowed us to classify OS samples into three *** classification model was further examined using the TARGET *** subgroup of OS had different prognoses and possible drug sensitivities,and OS cells in the three differentiation branches showed distinct interactions with other clusters in the OS *** addition,we verified the classification model through IHC staining in 138 OS samples,revealing a worse prognosis for Group B ***,we describe the novel transcriptional program of CSCs and highlight the activation of EZH2 in CSCs of *** findings provide a novel subclassification method based on sc RNA-seq and shed new light on the molecular features of CSCs in OS and may serve as valuable references for precision treatment for and therapeutic development in OS.

Cytoskeletal microtubule rearrangement and movement are crucial in the repair of spinal cord *** plays an important role in the regulation of microtubule *** spastin and collapsin response mediator proteins can regulate neurite growth and branching;however,whether spastin interacts with collapsin response mediator protein 3(CRMP3)during this process remains unclear,as is the mechanism by which CRMP3 participates in the repair of spinal cord *** this study,we used a proteomics approach to identify key proteins associated with spinal cord injury *** then employed liquid chromatography-mass spectrometry to identify proteins that were able to interact with glutathione ***,co-immunoprecipitation and staining approaches were used to evaluate potential interactions between spastin and ***,we co-transfected primary hippocampal neurons with CRMP3 and spastin to evaluate their role in neurite *** spectrometry identified the role of CRMP3 in the spinal cord injury repair *** chromatography-mass spectrometry pulldown assays identified three CRMP3 peptides that were able to interact with ***3 and spastin were co-expressed in the spinal cord and were able to interact with one another in vitro and in ***,CRMP3 overexpression was able to enhance the ability of spastin to promote neurite growth and ***,our results confirm that spastin and CRMP3 play roles in spinal cord injury repair by regulating neurite growth and *** proteins may therefore be novel targets for spinal cord injury *** Institutional Animal Care and Use Committee of Jinan University,China approved this study(approval ***-20181008-03)on October 8,2018.

Vik: A Chatbot to Support Patients with Chronic Diseases

Background: Chatbots are easy to use and simulate a human conversation through text or voice via smartphones or computers. In the field of health, chatbots can improve patient information, monitoring, or treatment adherence. Method: The objective of this article is to describe how a chatbot dedicated to disease monitoring and support of patients can interact with them and how data are exploited to be safe. Results: Wefight designed a chatbot named Vik to empower patients with cancers or chronic diseases and their relatives via personalized text messages. Natural Language Processing models were used. We built several Vik for each disease. Each Vik has its contents, its own NLP model and interacts its way with the patient. Conclusion: Conversational agents may help patients with minor health concerns without seeing a real physician. If the quality of these softwares is not thoroughly assessed, they could be dangerous. If chatbots are effective and safe, they could be prescribed like a drug to improve patient information, monitoring, or treatment adherence.

Acellular nerve allograft promotes selective regeneration

Acellular nerve ailograft preserves the basilar membrane tube and extracellular matrix, which promotes selective regeneration of neural defects via bridging. In the present study, a Sprague Dawley rat sciatic nerve was utilized to prepare acellular nerve allografts through the use of the chemical extraction method. Subsequently, the allograft was transplanted into a 10-mm sciatic nerve defect in Wistar rats, while autologous nerve grafts from Wistar rats served as controls. Compared with autologous nerve grafts, the acellular nerve allografts induced a greater number of degenerated nerve fibers from sural nerves, as well as a reduced misconnect rate in motor fibers, fewer acetylcholine esterase-positive sural nerves, and a greater number of carbonic anhydrase-positive sensory nerve fibers. Results demonstrated that the acellular nerve allograft exhibited significant neural selective regeneration in the process of bridging nerve defects.

Natural Postoperative Bone Metabolic Changes after Total Knee Arthroplasty Determined by Positron Emission Tomography Scans

Artificial joint replacement surgery is the orthopedic procedure of choice to relieve pain, correct joint deformities, and help patients resume everyday activities. However, the detailed mechanisms regulating peri-implant bone remodeling at the bone-prosthesis interface remain elusive. To address this gap in knowledge, we evaluated the natural postoperative course of bone metabolism by using [18F] NaF positron emission tomography (PET)/computed tomography in 57 patients (104 joints) who underwent asymptomatic total knee arthroplasty (TKA). First, we measured total tracer uptake around TKA to determine the total bone metabolism (TBM) and used the ischial tuberosity as the reference tissue for normalization of PET images. Second, we calculated the TBM ratio (TBMR) by dividing the TBM by tracer uptake in the reference tissue. A moderate increase in TBMR was observed 4 - 5 days after implantation, and its intensity reached the maximum on the seventh postoperative day. The TBMR showed no differences until 12 weeks, after which it decreased slowly and returned to the basal levels. The new parameter TBMR and the unique pattern of postoperative metabolic changes in the bone around the prosthesis may help set accurate interpretation criteria to diagnose complications such as loosening or infections.

PTEN/PI3K and MAPK signaling in protection and pathology following CNS injuries

Brain and spinal cord injuries initiate widespread temporal and spatial neurodegeneration, through both necrotic and programmed cell death mechanisms. Inflammation, reactive oxidation, excitotoxicity and cell-specific dysregulation of metabolic processes are instigated by traumatic insult and are main contributors to this cumulative damage. Successful treatments rely on prevention or reduction of the magnitude of disruption, and interfering with injurious cellular responses through modulation of signaling cascades is an effective approach. Two intracellular signaling pathways, the phosphatase and tensin homolog （PTEN）/phosphatidylinositol 3-kinase （PI3K） and mitogen- activated protein kinase （MAPK） signaling cascades play various cellular roles under normal and pathological conditions. Activation of both pathways can influence anatomical and functional outcomes in multiple CNS disorders. However, some mechanisms involve inhibiting or enhancing one pathway or the other, or both, in propagating specific downstream effects. Though many intraceHular mechanisms contribute to cell responses to insult, this review examines the evidence exploring PTEN/PI3K and MAPK signaling influence on pathology, neuroprotection, and repair and how these pathways may be targeted for advancing knowledge and improving neurological outcome after injury to the brain and spinal cord.

Rarely,penetrating injuries to the spinal cord result from wooden objects,creating unique challenges to mitigate neurological injury and high rates of infection and foreign body *** report a man who sustained a penetrating cervical spinal cord injury from a sharpened *** initially tetraparetic,he rapidly recovered *** risks of neurological deterioration during surgical removal made the patient reluctant to consent to surgery despite the impalement of the spinal cord.A repeat MRI on day 3 showed an extension of edema indicating progressive *** the 7~(th)day after injury,fever and paresthesias occurred with a large increase in serum inflammatory indicators,and the patient agreed to undergo surgical removal of the wooden *** discuss the management nuances related to wood,the longitudinal evolution of MRI findings,infection risk,surgical risk mitigation and technique,an inflammatory marker profile,long-term recovery,and the surprisingly minimal neurological deficits associated with low-velocity midline spinal cord *** patient had an excellent clinical *** main lessons are that a wooden penetrating central nervous system injury has a high risk for infection,and that surgical removal from the spinal cord should be performed soon after injury and under direct visualization.
