Metagenomic perspectives on antibiotic resistance genes in tap water:The environmental characteristic,potential mobility and health threat

As an emerging environmental contaminant,antibiotic resistance genes(ARGs)in tap water have attracted great *** studies have provided ARG profiles in tap water,research on their abundance levels,composition characteristics,and potential threat is still ***,9 household tap water samples were collected from the Guangdong-Hong Kong-Macao Greater Bay Area(GBA)in ***,75 sets of environmental sample data(9 types)were downloaded from the public *** was then performed to explore the differences in the abundance and composition of ARGs.221 ARG subtypes consisting of 17 types were detected in tap *** the ARG abundance in tap water was not significantly different from that found in drinking water plants and reservoirs,their composition *** tap water samples,the three most abundant classes of resistance genes were multidrug,fosfomycin and MLS(macrolide-lincosamidestreptogramin)ARGs,and their corresponding subtypes ompR,fosX and macB were also the most abundant ARG *** the potential mobility,vanS had the highest abundance on plasmids and viruses,but the absence of key genes rendered resistance to vancomycin ***,the majority of ARGs present in tap water were those that have not been assessed and are currently not listed as high-threat level ARG families based on the World Health Organization *** the current potential threat to human health posed by ARGs in tap water is limited,with persistent transfer and accumulation,especially in pathogens,the potential danger to human health posed by ARGs should not be ignored.

An application of a seismic nodal system with seismic ambient noise near Kunlun Station, Antarctica: estimating ice thickness and firn structure

The thickness and upper densification structure of an ice sheet are important parameters for dynamic ice sheet modeling and glacier mass balance studies. Seismic ambient noise methods, such as the horizontal-to-vertical spectral ratio(H/V) method and ambient noise cross-correlation method, are becoming increasingly popular in glacier structure investigations. During China's 39th expedition to Antarctica, seismic ambient noise experiments were conducted to investigate the structure of the ice sheet at Kunlun Station, Dome A,using a seismic nodal system. We obtained a broad band(0.1–10 Hz) H/V curve with a 1-hour noise record from a seismic node. In addition, we extracted the Rayleigh wave dispersion curve with 5-day noise cross-correlation functions from a linear dense seismic *** clear peaks were observed in the H/V curve — a lower peak at ～0.17 Hz and two higher frequency peaks at ～3 Hz and ～6 Hz. We inverted the ice sheet thickness using the lower frequency portion of the H/V curve and inverted the upper structure of the ice sheet using the higher frequency portion of the H/V curve jointly with the dispersion curve. Our estimations from ambient noise observations were consistent with those derived from the BedMachine ice sheet thickness dataset and the density profile determined by ground-penetrating radar investigations at the same site.

Terahertz Frequency Domain Response for Insulation State Assessment of Vehicle Cable Terminal Under Electro-Thermal Aging

To accurately diagnose the aging condition of the vehicle-mounted high-voltage cable terminals under the combined electro-thermal stress,this paper proposes a terahertz frequency spectrum technology-based ***,an electro-thermal aging platform is established in the laboratory to obtain the test samples of ethylene-propylene rubber(EPR)cable terminals with different aging ***,the degree of electro-thermal aging is characterized by frequency spectrum,absorption coefficient spectrum,absorption spectrum,refractive index and dielectric constant in the terahertz ***,the micro-morphology and micro-area structure of the test samples under different aging gradients are also observed by scanning electron microscopy,and both the material and chemical properties are *** findings demonstrate that terahertz frequency spectra offer significant benefits in non-destructively detecting and identifying the insulation condition of vehicle cable terminals during electro-thermal *** tests confirm the feasibility of utilizing the terahertz frequency spectrum to assess the insulation aging state of EPR cable terminals,making it potentially applicable for on-site purposes.

Optimal Coupling Height of the Atmosphere and Land Surface——An Earth System Modeling Perspective

In Earth system modeling,the land surface is coupled with the atmosphere through surface turbulent *** fluxes are computed using mean meteorological variables between the surface and a reference height in the ***,the dependence of flux computation on the reference height,which is usually set as the lowest level in the atmosphere in Earth system models,has not received much *** on high-resolution large-eddy simulation(LES)data under unstable conditions,we find the setting of reference height is not trivial within the framework of current surface layer *** a reasonable prescription of aerodynamic roughness length(following the setting in LESs),reference heights near the top of the surface layer tend to provide the best estimate of surface fluxes,especially for the momentum ***,this conclusion for the sensible heat flux is insensitive to the ratio of roughness length for momentum versus *** results are robust,whether using the classical or revised surface layer *** provide a potential guide for setting the proper reference heights for Earth system modeling and can be further tested in the near future using observational data from land–atmosphere feedback observatories.

Influence of Gravity on Structure of Colloidal Crystal Using Simulated Microgravity

Liquid mixtures of water and deuterium oxide as the liquid phase, were used to match the density of charged colloidal particles. Kossel diffraction method was used to detect the crystal structures. The experiments under the density-matched （g=0） and unmatched （g=1） conditions are compared to examine the influence of gravity on the crystal structures formed by self-assembly of 110 nm （in diameter） polystyrene microspheres. The result shows that the gravity tends to make the lattice constants of colloidal crystals smaller at lower positions, which indicates that the effect of gravity should be taken into account in the study of the colloidal crystals.

Discovering Climate Change during the Early 21st Century via Wasserstein Stability Analysis

Climate change is an essential topic in climate science,and the accessibility of accurate,high-resolution datasets in recent years has facilitated the extraction of more insights from big-data ***,current research predominantly focuses on mean-value changes and largely overlooks changes in the probability *** this study,a novel method called Wasserstein Stability Analysis(WSA)is developed to identify probability density function(PDF)changes,especially the extreme event shift and nonlinear physical value constraint variation in climate *** is applied to the early 21st century and compared with traditional mean-value trend *** results indicate that despite no significant trend,the equatorial eastern Pacific experienced a decline in hot extremes and an increase in cold extremes,indicating a La Nina-like temperature *** analysis at two Arctic locations suggests sea ice severely restricts the hot extremes of surface air *** impact is diminishing as the sea ice *** revealing PDF shifts,WSA emerges as a powerful tool to re-examine climate change dynamics,providing enhanced data-driven insights for understanding climate evolution.

It is critical to assess the extent and progression of liver fibrosis for patients to receive suitable treatments,but its diagnostic methods remain *** matrix protein 1(ECM1)has previously been reported to be a key factor in the induction and progression of liver ***,little is known about the use of ECM1 as a biomarker to evaluate *** a CCl_(4)-induced mouse model of liver fibrosis,the present study demonstrated that ECM1 decreased with gradually increasing *** biopsy as a reference,the serum ECM1 levels decreased with increasing fibrosis stage in 247 patients with liver fibrosis,but there were no significant changes between fibrosis stage 2 and stage 0–*** improve the performance of ECM1,age,platelet count,and ECM1 concentration were combined to calculate an EPA(ECM1-platelet-age)score(ranging from 0 to 10).The areas under the receiver operating characteristic curve of the EPA scores for the detection of F≥2,F≥3,and F4 were 0.6801,0.7377,and 0.8083,respectively,which showed a comparable or significantly greater diagnostic performance for assessing fibrosis than that of the AST/ALT ratio,APRI score,or FIB-4 *** HBV patients following antiviral treatment,the dynamics of the EPA score depended on the status of liver fibrosis *** accuracy of the EPA score in predicting fibrosis regression and progression was 66.00%and 71.43%,respectively,while that of the LSM,another useful method for monitoring hepatic fibrosis changes during treatment,was only 52.00%and 7.14%,*** with healthy controls,there were lower levels of serum ECM1 in HBV patients and individuals with HCV infection,MAFLD,ALD,PBC,and *** findings suggested that individuals with reduced ECM1 levels may have a risk of developing liver injury,and further examinations or medical care are *** conclusion,the ECM1-containing EPA score is a valuable noninvasive test for staging fibrosis and predicting the progression of liv

Morphological alterations in dendritic spines have been linked to changes in functional communication between neurons that affect learning and ***-4 KIF21A helps organize the microtubule-actin network at the cell cortex by interacting with KANK1;however,whether KIF21A modulates dendritic structure and function in neurons remains *** this study,we found that KIF21A was distributed in a subset of dendritic spines,and that these KIF21A-positive spines were larger and more structurally plastic than KIF21A-negative ***,the interaction between KIF21A and KANK1 was found to be critical for dendritic spine morphogenesis and synaptic *** of either KIF21A or KANK1 inhibited dendritic spine morphogenesis and dendritic branching,and these deficits were fully rescued by coexpressing full-length KIF21A or KANK1,but not by proteins with mutations disrupting direct binding between KIF21A and KANK1 or binding between KANK1 and *** down KIF21A in the hippocampus of rats inhibited the amplitudes of long-term potentiation induced by high-frequency stimulation and negatively impacted the animals’cognitive *** together,our findings demonstrate the function of KIF21A in modulating spine morphology and provide insight into its role in synaptic function.

Severe combined immunodeficiency disease(SCID),characterized by profound immune system dysfunction,can lead to life-threatening infections and death. Animal models play a pivotal role in elucidating biological processes and advancing therapeutic strategies. Recent advances in gene-editing technologies, including zincfinger nucleases(ZFNs), transcription activator-like effector nucleases(TALENs), CRISPR/Cas9, and base editing, have significantly enhanced the generation of SCID models. These models have not only deepened our understanding of disease pathophysiology but have also driven progress in cancer therapy, stem cell transplantation, organ transplantation, and infectious *** comprehensive overview of current SCID models generated using novel gene-editing approaches,highlighting their potential applications in translational medicine and their role in advancing biomedical research.
