Taurine in semen extender modulates post–thaw semen quality,sperm kinematics and oxidative stress status in mithun(Bos frontalis)spermatozoa

Objective:To assess the effect of taurine on post-thaw semen quality parameters,sperm kinematics,antioxidant and oxidative stress profiles and sperm cholesterol efflux in mithun(Bos frontalis).Methods:A total of 50 ejaculates(n=25 samples)were selected based on biophysical *** sample was split into four equal aliquots after dilution with the Tris-citrate-glycerol ***Ⅰ,Ⅱ,ⅢandⅣcontained 0 mM(the control),25 mM,50 mM and 100 mM of taurine,***-thawed samples were analysed for motility parameters(progressive forward and in bovine cervical mucus penetration test),kinetic and velocity parameters by computer-assisted sperm analyzer,viability,sperm and nuclear abnormalities,acrosome integrity,plasma membrane and nuclear integrities,sperm enzymatic leakage and biochemical(sperm cholesterol and oxidative stress)***:The extender containing 50 mM taurine led to a significant enhancement in viability,acrosomal integrity,plasma membrane integrity,motility(progressive and in cervical mucus),and sperm cholesterol content and notably reduced sperm morphological and nuclear abnormalities,and leakage of intracellular enzymes compared to other taurine treated and untreated control groups(P<0.05).Moreover,in addition to significant improvement in kinetic and velocity profiles,50 mM taurine protected the integrity of acrosome and biochemical membranes than in the untreated control and other taurine treated *** of 50 mM taurine held a clear advantage over the control or 25 mM or 100 mM taurine in cryopreservation of mithun ***:Taurine(50 mM)supplementation in semen extender can be effectively utilized to reduce oxidative stress and improve post-thaw semen quality in mithun.

Outcome of repair of chronic tear of the pectoralis major using corkscrew suture anchors by box suture sliding technique

AIM To assess the functional and clinical results of repair of chronic tears of pectoralis major using corkscrew and sliding suture technique. METHODS In this retrospective study, we reviewed the results of pectoralis major repair in 11 chronic cases(> 6 wk) done between September 2011 and December 2014 at our institute. In all cases repair was done by same surgeon using corkscrew suture anchors and box suture sliding technique. At 6 mo, after surgery magnetic resonance imaging was done to see the integrity of the repair. Functional evaluation was done using Penn and ASES scores. Pre and postoperative Isokinetic strength was *** Average follow-up was 48.27 ± 21.0 mo. The Wilcoxon signed rank test was used to evaluate the outcome scores. The average ASES score increased from an average of 54.63 ± 13.0 preoperatively to 95.09 ± 2.60 after surgery at their last follow-up. The average Penn score also increased from 5.72 ± 0.78, 2.81 ± 1.32 and 45.81 ± 1.72 to 9.36 ± 0.80, 8.27 ± 0.90 and 59 ± 1.34 for pain, satisfaction and function respectively. Follow up magnetic resonance imaging(MRI)(at 6 mo) showed continuity and the bulk of pectoralis major muscle in all cases. Average isokinetic strength deficiency in horizontal adduction at 60° was 13.63% ± 6.93% and at 120° was 10.18% ± 4.93% and in flexion at 60° was 10.72% ± 5.08% and at 120° was 6.63% + 3.74%. Results showed that both ASES and Penn score improved significantly(2 tailed P value = 0.0036).CONCLUSION We could conclude from this series that pectoralis major repair even in chronic cases using 5.5 mm corkscrew anchors give excellent functional and cosmetic results. In chronic cases the repairable length of the tendon is not available and sliding suture technique allows for fixation of worn out tendomuscular junction to bone without letting cutting through the muscle.

evaluation of acute and subacute toxicity of whole-plant aqueous extract of Vernonia mespilifolia Less. in Wistar rats

Objective： This study investigated the acute and subacute toxicity of whole-plant aqueous extract of Vernonia mespilifolia Less. （AEVM） in rats for evaluating its safety profile. Methods： AEVM for the acute （2000 and 5000 mg/kg） and subacute （200, 400 and 600 mg/kg） toxicity studies was administered orally to rats according the guidelines 425 and 407 of Organization for Economic Cooperation and Development, respectively. Food and water intake as well as body and organ weight of animals were recorded. Signs of toxicity were assessed, and hematological, biochemical and histopathological analyses were performed. Results： In the acute toxicity study, a single dose of the aqueous extract at 2000 or 5000 mg/kg caused no mortality in the animals, suggesting that the median lethal dose is greater than 5000 mg/kg. In the subacute toxicity study, administration of the extract for 28 d, at all doses, caused no significant changes in the body weights or organ weights of rats in the treated groups when compared with the control group. In addition, hematological and biochemical parameters also revealed no toxic effects of the extract on rats. Histological sections of the heart, liver and kidney from test animals showed no signs of degen- eration. Conclusion： These results showed that AEVM at dosage levels up to 600 mg/kg is nontoxic and could also offer protection on some body tissues. AEVM could, therefore, be considered safe.

Credibility protection for resource sharing and collaborating in non-center network computing environments

Non-center network computing environments have some unique characteristics, such as instability, heterogeneity, autonomy, distribution and openness, which bring serious issues of security and reliability. This article proposes a brand-new credibility protection mechanism for resource sharing and collaboration in non-center network computing environments. First, the three-dimensional hierarchical classified topology （3DHCT） is proposed, which provides a basic framework for realizations of the identity credibility, the behavior credibility and the capability credibility. Next, the agent technology is utilized to construct the credibility protection model. This article also proposes a new comprehensive credibility evaluation algorithm with simple, efficient, quantitative and able to meet the requirements of evaluating behavior credibility and the capability credibility evaluation as well. The Dempster-Shafer theory of evidence and the combination rule are used to achieve the evaluation of the capability credibility. The behavior credibility is evaluated with the current and historical performance of nodes for providers and consumers to realize more accurate prediction. Based on the non-center network computing simulation test platform, simulation is been conducted to test the performance and validity of the proposed algorithms. Experiment and analysis show that the proposed algorithms are suitable for large-scale, dynamic network computing environments, and able to maintain the credibility for networks without relying on central node, make a non-center network gradually evolve into an orderly, stable and reliable computing environment efficiently.

In vitro evaluation of artificial joints: a comprehensive review

Development of artificial joints is a great progress for joint replacement operations of human being, but the short longevity of prostheses has concerned both industries and researchers since the advent of modem implants. Thus, continuous improvements have been made alongside the clinical applications. The methodologies to inspect the potential properties of new designs have also seen many advances. The test machines have evolved from easy pin-on-disk configuration to the modem joint simulator. Besides, various wear traces provided by testing machines are investigated greatly, and multi-directional files are recommended as motion profiles for in vitro testing. The typical testing parameters, like sliding speed or loading profile, are discussed in this article, and their working mechanisms are described in detail. Meanwhile, the calf or bovine serum has been regarded as the gold standard for testing lubricant, and the future trend of fluid will be focused on investigating the effects of lubricant composites on the tribological properties. Hard particles and proteins are discussed as well, and their effects on the wear mechanisms are the focus. Finally, various approaches to measure the wear rate, wear factor and the thickness of lubricant film are presented, and suggestions are given for the development in future.

An evaluation method for internal erosion potential of gravelly soil based on particle size distribution

Internal erosion occurs when fine particles escape from the soil driven by seepage flow,which is considered to be the crucial factor causing the failure of earth structures filled with gravelly *** objective of this paper is to suggest an appropriate method to assess internal erosion potential of gravelly *** analyzing the sensitivity of soil material to internal erosion,the variable(Dc15/df85)max and the content of coarse particles(Pc)are selected as the evaluation indexes(Dc15 and df85 are the diameters of 15%mass passing in the coarse component and 85%mass passing in the fine component,respectively).A series of gravelly soils with different particle size distributions are tested for internal erosion by the self-made *** on the test results,an evaluation method for the internal erosion of gravelly soil is *** soil is prone to internal erosion when 60%≤Pc<95%and(Dc15/df85)max≥*** proposed method shows good accuracy in evaluating the internal erosion of 36 soil samples from other studies,which confirms the reliability of the *** proposed method makes it possible to accurately assess internal erosion of gravelly soil,and an alternative method is provided for engineers to determine whether there is a risk of internal erosion in earth structures consisting of gravelly soil.

Sperm chromatin structure assay （SCSA）： a tool in diagnosis and treatment of infertility

Diagnosis of male infertility has mainly been based on the World Health Organization （WHO） manual-based semen parameter＇s concentration, motility and morphology. It has, however, become apparent that none of these parameters are reliable markers for evaluation of the fertility potential of a couple. A search for better markers has led to an increased focus on sperm chromatin integrity testing in fertility work-up and assisted reproductive techniques. During the last couple of decades, numerous sperm DNA integrity tests have been developed. These are claimed to be characterized by a lower intraindividual variation, less intralaboratory and interlaboratory variation and thus less subjective than the conventional sperm analysis. However, not all the sperm chromatin integrity tests have yet been shown to be of clinical value. So far, the test that has been found to have the most stable clinical threshold values in relation to fertility is the sperm chromatin structure assay （SCSA）, a flow cytometric test that measures the susceptibility of sperm DNA to acid-induced DNA denaturation in situ. Sperm DNA fragmentation as measured by SCSA has shown to be an independent predictor of successful pregnancy in first pregnancy planners as well as in couples undergoing intrauterine insemination, and can be used as a tool in investigation, counseling and treatment of involuntary childlessness. More conflicting data exist regarding the role of sperm DNA fragmentation in relation to fertilization, pre-embryo development and pregnancy outcome in in vitro fertilization and intracytoplasmic sperm injection （ICSI）.

evaluation and Relevance of Indications for Primary Caesarean Section: A Five-Year Experience Report from Nevers Hospital Center

Context: The caesarean section rate continues to increase in our different health structures specially for women who have not had a scar in the uterus. Objectives: The aim of this study was to analyze the key factors and main indications for primary caesarean sections and to find ways to reduce the increasing rates. Patients and Method: This is a longitudinal and retrospective study carried out from June 1, 2018 to July 31, 2022. The study included all patients who had a cesarean-section for the first time (primary caesarean). An anterior uterine scar was a non-inclusion criterion. Data were collected prospectively using Synfonievre and Agopra software via patients’ files and information collection sheet. Data were analyzed with SPSS 21 software, Mac version. Averages were calculated for quantitative data and percentages for qualitative data. The statistical tests used were the Pearson Chi2 test. The observed differences were considered significant when the p-value was less than 0.05. Results: During the study period, we recorded 8832 deliveries and 3148 caesarean sections (35.6%). Primary CS concerned 70% of overall C-section rate. The main indications were FHR Fetal Heart Rate abnormalities (FHRA) (27%), followed by the other indications (including preterm delivery, umbilical cord dystocia, malpresentation of fetus, foetal abnormalities, elective CS, triple gestation, mother abnormalities);dystocia or prolonged labor (18.7%), breech presentation in a twin pregnancy with 11.3% and 9.6% respectively. We recorded more vaginal deliveries with labor induction: 81.4% against 75.2%. An obstetrical audit led to better labor management and a reduction in the cesarean section rate. Conclusion: We need to focus on diagnosis of fetal distress, management of breech presentation during of a twin birth and a singleton. Induction of labor can be an effective alternative in certain indications. An obstetrical audit is needed to reverse the caesarean section rate.

Loss of FHIT expression in gastric mucosa of patients with family histories of gastric cancer and Helicobacter pylori infection

AIM: To answer the question whether FHIT gene expression is affected by the family history of gastric carcinoma and the presence of Helicobacter pylori (Hpylori) in the gastric mucosa of patients with ***: FHIT gene expression in two different topographic sites of the gastric mucosa of twenty-one patients with dyspepsia and with or without familial gastric carcinoma, infected or not infected with H pylori, was evaluated by reverse transcription-PCR (RT-PCR) and IMAGE QUANT methods. A rapid urease test and histopathological examination were used to determine H pylori ***: In the gastric mucosa of patients with family histories of gastric carcinoma, the amount of FHIT protein mRNA was reduced down to 32%, and for patients with H pylori colonization, to 24% in comparison to controls with dyspepsia and without cancer in the family. FHIT expression was independent of the topography of specimens (corpus vsantrum), and for the control patients it was less sensitive to infection with H pylori. A considerable statistical difference in FHIT levels was observed in the gastric mucosa from the corpus of patients with family histories of gastric carcinoma in respect to H pylori colonization (P = 0.06). Macroscopic evaluation of the gastric mucosa demonstrated that pathologic changes classified according to the Sydney system had no significant influence on FHIT expression within each tested group of ***: Loss of FHIT expression was observed in patients with dyspepsia and family histories of gastric carcinoma, especially those infected with H pylori. Such results may constitute an early indication of the development of gastric carcinoma, which is associated with family factors including heredity and H pylori infection. The loss of the FHIT gene may serve as a marker for early diagnosis and prevention of gastric carcinoma, especially in context of early monitoring of H pylori infection in individuals with a record of familial stomach ca
