The Role of the European Pharmacopoeia(Ph Eur)in Quality Control of Traditional Chinese Herbal medicine in European Member States

In order to assure the safety and efficacy of the Chinese medicines in Europe, the quality of TCM herbals should be guaranteed so that they can be freely imported in the European Union and other Western European Countries which are signatories of the European Pharmacopoeia Convention. Consequently, new Ph Eur TCM herbal drug Monographs should be elaborated, based on preexisting Monographs in the Chinese Pharmacopoeia(Ch P) *** a program has been inaugurated in 2005 by the Ph Eur Groups of Experts 13 A and B(Phytochemistry). Since then good progress has been made, elaborating of about one third of the originally proposed 100 TCM herbals being identified as important monographs for the European Market. Taking into account the many challenges still laying ahead, the establishment of a specialized Working Party(WP) on TCM with specialists and experts from many EU Member States has been decided by the Ph Eur Commission in 2008 which is highly active ever since in the examination and elaboration of new TCM herbal drug monographs, primarily to assure the safety of the European patient and further to provide quality parameters extremely important for all registration and licensing procedures of the respective National Authorities all over *** paper is a survey of results and difficulties obtained so far which has been encountered meanwhile in the elaboration process by the Ph Eur TCM WP of these monographs and will discuss these in detail. Moreover the role of Ph Eur TCM monographs in the European community is addressed.

Objective: Digoxin is a therapeutic cardenolide widely used to treat various heart conditions such as atrial flutter, atrial fibrillation and heart failure in both Western as well as Chinese medicine. Digoxin is extracted from cultivated Digitalis lanata Ehrh. plants, known as Mao Hua Yang Di Huang in Chinese medicine. This manuscript presents two studies that were conducted to optimize the cultivation conditions for digoxin production in the TCM Mao Hua Yan Di Huang in a greenhouse under GAP ***: Two experiments were designed in which 4 growth conditions were compared. Levels of digoxin, gitoxin, digitoxin, α-acetyldigoxin, β-acetyl-digoxin were measured using HPLC-UV and compared between the ***: Normal soil, no CO_2 enrichment combined with a cold shock was found to be the optimal condition for producing digoxin in the first experiment. Gitoxin content was significantly lower in plants grown in this condition. Mechanical stress as well as the time of harvesting showed no statistically significant differences in the production of cardenolides. In the second experiment the optimal condition was found to be a combination of cold nights, sun screen, fertilizer use and no milled ***: This study shows that digoxin production can be increased by controlling the growth conditions of D. lanata Ehrh. The effect of cold was important in both experiments for improving digoxin production. Cultivating Chinese herbal medicines in optimized greenhouse conditions might be an economically attractive alternative to regular open air cultivation.

Objective: To explore the effects of two Rehmanniae Radix formulae in patients with metabolic syndrome(Met S), a randomized controlled study was ***: Met S patients were randomly assigned to receive either a classic Rehmanniae Six Formula(R6, or ‘Liu Wei Di Huang Wan') or a novel multi-herbal Rehmanniae Radix containing formula SUB889 for 8 weeks. Western medicine related clinical parameters, Chinese medicine defined symptoms and syndromes as well as metabolomic profiles were evaluated at different time ***: R6(n = 20) and SUB889(n = 20) showed similar effects on Met S regarding the improvement of clinical parameters(waist circumference, body mass index, LDL-cholesterol, systolic blood pressure) and Qi/Yin deficiency(p medicine and Western medicine defined clinical outcomes. R6 is more effective in improving lipid profiles compared to SUB889. The exact mechanisms of the two formulae on Met S remain to be elucidated.

Exploring the mechanism of neural-function reconstruction by reinnervated nerves in targeted muscles

A lack of myoelectric sources after limb amputation is a critical challenge in the control of multifunctional motorized prostheses. To reconstruct myoelectric sources physiologically related to lost limbs, a newly proposed neural-function construc- tion method, targeted muscle reinnervation （TMR）, appears promising. Recent advances in the TMR technique suggest that TMR could provide additional motor command information for the control of multifimctional myoelectric prostheses. However, little is known about the nature of the physiological functional recovery of the reinnervated muscles. More understanding of the under- lying mechanism of TMR could help us fine tune the technique to maximize its capability to achieve a much higher performance in the control of multifunctional prostheses. In this study, rats were used as an animal model for TMR surgery involving transferring a median nerve into the pectoralis major, which served as the target muscle. Intramuscular myoelectric signals reconstructed following TMR were recorded by implanted wire electrodes and analyzed to explore the nature of the neural-fimction recon- struction achieved by reinnervation of targeted muscles. Our results showed that the active myoelectric signal reconstructed in the targeted muscle was acquired one week after TMR surgery, and its amplitude gradually became stronger over time. These pre- liminary results from rats may serve as a basis for exploring the mechanism of neural-function reconstruction by the TMR tech- nique in human subjects.

Transferring deep neural networks for the differentiation of mammographic breast lesions

Machine learning can help differentiating benign and malignant lesions seen on mammographic images. Conventional models require handcrafting features for lesion representation. Due to insufficient medical instances, the performance of convolutional neural networks(CNNs) can be further increased. This study makes use of transfer learning for mammographic breast lesion diagnosis and deep neural network(DNN) models pre-trained with large-scale natural images are employed. The diagnosis performance is evaluated with the prediction accuracy(ACC) and the area under the curve(AUC) on average. A histologically verified database is analyzed which contains 406 lesions(230 benign and 176 malignant). Involved models include transferred DNNs(GoogLeNet and AlexNet), shallow CNNs(CNN2 and CNN3) that are fully trained with medical instances and boosted by support vector machine(SVM), and two conventional methods which combine handcrafted features and SVM for lesion diagnosis. Experimental results indicate that GoogLeNet achieves the best performance(ACC=0.81, AUC=0.88), followed by AlexNet(ACC=0.79, AUC=0.83) and CNN3(ACC=0.73, AUC=0.82). Knowledge transfer can improve the mammographic breast cancer diagnosis, while its wide application still requires further verification in medical imaging domain.

Ballistocardiogram signal monitoring system based on GSM network was put forward in this paper. The system included a BCG signal acquisition module, a data processing module, a display module and a GSM module. The STM32F103 VB microprocessor was used as the controlling core of the signal acquisition module. BCG signal acquisition, amplification, filtering and A/D conversion were completed by the resistance strain sensor and high precision A/D conversion chip of TM7708; VB6.0software was used to realize the BCG signal analysis and processing; the SD card and LCD completed data storage and waveform display; the BCG data remote transmission and alarm function were realized through the GSM module. The system cannot only real-time monitor the changes of heart rate of patients by non-contact means, and can process data automatically, timely detection of arrhythmia and automatic alarm. The system is particularly suitable for heart disease patients receiving long-term home care; therefore, it has a broad application prospect.

Self-incompatibility(SI),which has recurred during the evolution of plants,is one of the most important cross-pollination mating *** S-loci have been reported in Brassicaceae,namely,Arabidopsis lyrata(Al),Brassica(Br),and Leavenworthia alabamica(La)***,through multi-genomic comparative analysis of 20 species,we revealed that the most ancient S-locus was formed prior to the divergence of Brassicaceae lineage I and *** retained and inherited by Arabidopsis,as the Al S-locus in Brassicaceae lineage ***,we found that the Br S-locus,which has been widely used in the breeding of Brassica crops to generate hybrid seeds,was formed through segmental translocation(ST)in the hexaploid ancestor of Brassica in Brassicaceae lineage *** Br S-locus was evolved through a ST from one of the triplicated ancestral S-locus paralogs in the Brassica hexaploidy ancestor,while the other two S-locus paralogs were *** with the previous discovery that the La S-locus was formed through a secondary origin in Brassicaceae lineage I,we conclude the monophyletic origin of Al and Br S-loci and clarify the evolutionary route of S-loci in the Brassicaceae *** findings will contribute to evolutionary studies and breeding applications of the S-locus in Brassicaceae.

Plant Polyploidy: Origin, Evolution, and Its Influence on Crop Domestication

The prevalence and recurrence of polyploidization in plant species make it one of the most important evolutionary events in plants, and as a result, polyploidization is an extensively investigated research field. Due to the rapid development of sequencing technologies, there is increased evidence to support that polyploidization plays an important role in the diversification of plant species, evolution of genes, and the domestication of crops. Here, we reviewed the influence of polyploidization on various aspects of plant evolution, mainly focused on polyploid origin, characteristics, subsequent genome divergence, and its impact on gene function innovation and crop domestication. The occurrence of many independent polyploidization events in plants was found to be tightly associated with the timing of extreme climate events or natural disasters on earth, leading to mass extinction while possibly facilitating increased polyploidization. Following allo-polyploidization, a distinct phenomenon known as sub-genome dominance occurred during sub-genome evolution, which was found to be associated with the methylation of transposons. Extensive gene fractionations(lost) following polyploidization were reported in almost all paleo-polyploids, and the evolutionary fates of multi-copy genes, such as sub-/neo-functionalization, were further proposed to illustrate their underlying mechanisms. Moreover,polyploidization was found to significantly impact species diversification, with subsequent effects on crop domestication and the development of traits with agronomic importance. Based on the progress of plant polyploidization studies, we discussed several main topics that might further improve our understanding of polyploid evolution and that are likely contribute to the application of polyploidization in crop breeding in the near future.
