Single-cell landscape analysis reveals distinct regression trajectories and novel prognostic biomarkers in primary neuroblastoma

Neuroblastoma(NB),which is the most co.mon pediatric extracranial solid tumor,varies widely in its clinical presentation and *** has a unique ability to spontaneously differentiate and regress,suggesting a potential direction for therapeutic ***,the underlying mechanisms of regression remain largely unknown,and more reliable prognostic biomarkers are needed for predicting trajectories for *** performed scRNAseq analysis on 17 NB clinical samples and three peritumoral adrenal *** NB displayed varied cell co.stitution,even among tumors of the same pathological *** number variation patterns suggested that neuroendocrine cells represent the malignant cell *** on the differential expression of sets of related marker genes,a subgroup of neuroendocrine cells was identified and projected to differentiate into a subcluster of benign fibroblasts with highly expressed CCL2 and ZFP36,supporting a progressive pathway of spontaneous NB *** also identified prognostic markers(STMN2,TUBA1A,PAGE5,and ETV1)by evaluating intra-tumoral ***,we determined that ITGB1 in M2-like macrophages was associated with favorable prognosis and may serve as a potential diagnostic marker and therapeutic *** co.clusion,our findings reveal novel mechanisms underlying regression and potential prognostic markers and therapeutic targets of NB.

Background::Since 2019,a novel co.onavirus named 2019 novel co.onavirus(2019-nco.)has emerged *** from fever and respiratory co.plications,acute kidney injury has been observed in a few patients with co.onavirus disease ***,acco.ding to recent findings,the virus has been detected in ***-co.verting enzyme II(ACE2)has been proposed to serve as the receptor for the entry of 2019-nco.,which is the same as that for the severe acute respiratory *** study aimed to investigate the possible cause of kidney damage and the potential route of 2019-nco. infection in the urinary ***::We used both published kidney and bladder cell atlas data and new independent kidney single-cell RNA sequencing data generated in-house to evaluate ACE2 gene expression in all cell types in healthy kidneys and *** Pearson co.relation co.fficients between ACE2 and all other genes were first ***,genes with r values larger than 0.1 and P values smaller than 0.01 were deemed significant co.expression genes with ***::Our results showed the enriched expression of ACE2 in all subtypes of proximal tubule(PT)cells of the ***2 expression was found in 5.12%,5.80%,and 14.38%of the proximal co.voluted tubule cells,PT cells,and proximal straight tubule cells,respectively,in three published kidney cell atlas *** addition,ACE2 expression was also co.firmed in 12.05%,6.80%,and 10.20%of cells of the proximal co.voluted tubule,PT,and proximal straight tubule,respectively,in our own two healthy kidney *** the analysis of public data from three bladder samples,ACE2 expression was low but detectable in bladder epithelial *** 0.25%and 1.28%of intermediate cells and umbrella cells,respectively,had ACE2 ***::This study has provided bioinformatics evidence of the potential route of 2019-nco. infection in the urinary system.
