Analysis of DNA methylation variation in wheat genetic background after alien chromatin introduction based on methylation-sensitive amplification polymorphism

During the process of alien germplasm introduced into wheat genome by chromosome engineering, extensive genetic variations of genome structure and gene expression in recipient could be induced. In this study, we performed GISH (genome in situ hybridization) and AFLP (amplified fragment length po-lymorphism) on wheat-rye chromosome translocation lines and their parents to detect the identity in genomic structure of different translocation lines. The results showed that the genome primary struc-ture variations were not obviously detected in different translocation lines except the same 1RS chromosome translocation. Methylation sensitive amplification polymorphism (MSAP) analyses on genomic DNA showed that the ratios of fully-methylated sites were significantly increased in translo-cation lines (CN12, 20.15%; CN17, 20.91%; CN18, 22.42%), but the ratios of hemimethylated sites were significantly lowered (CN12, 21.41%; CN17, 23.43%; CN18, 22.42%), whereas 16.37% were fully-me-thylated and 25.44% were hemimethylated in case of their wheat parent. Twenty-nine classes of me-thylation patterns were identified in a comparative assay of cytosine methylation patterns between wheat-rye translocation lines and their wheat parent, including 13 hypermethylation patterns (33.74%), 9 demethylation patterns (22.76%) and 7 uncertain patterns (4.07%). In further sequence analysis, the alterations of methylation pattern affected both repetitive DNA sequences, such as retrotransposons and tandem repetitive sequences, and low-copy DNA.

Because the oilfields in eastern China are in the very high water cut development stage, accurate forecast of oilfield development indices is important for exploiting the oilfields efficiently. Regarding the problems of the small number of samples collected for oilfield development indices, a new support vector regression prediction method for development indices is proposed in this paper. This method uses the principle of functional simulation to determine the input-output of a support vector machine prediction system based on historical oilfield development data. It chooses the kernel function of the support vector machine by analyzing time series characteristics of the development index; trains and tests the support vector machine network with historical data to construct the support vector regression prediction model of oilfield development indices; and predicts the development index. The case study shows that the proposed method is feasible, and predicted development indices agree well with the development performance of very high water cut oilfields.

Integrated development of industrial cooperation is the core content in the Bohai economic *** structure convergence of Bohai economic rim mainly exist in macro three industries while it rarely exist in the manufacturing industry and other internal meso level. Learning from changing tendency, industrial structure convergence in the five provinces and municipality in the Bohai Economic Rim tend to be increasingly intense. For integration of industry cooperation development in Bohai Economic Rim, it is faced with massive separation, industrial development convergence and is restricted by institutional and ideological barriers. As strategic choice based on interest tending, integration of industry cooperation development in Bohai Economic Rim should be further integrated on development concept of industrial transformation to promote institutional system of the integration of industrial development and optimize the industrial layout and division system of Bohai economic rim.

In this paper, the method of generalized moments of data and systems for 101 counties in Jiangsu and Zhejiang Provinces is used to analyze the impact of industrial agglomeration and industrial structure upgrading on county economic growth. The real diagnosis result indicated that, the industry gathers with the industrial structure promotes has the forward function to the county territory economic growth rate, in the sample time that had not discovered the industry gathers to the economical growth existence non-linearity in？ uence. On the other hand, the industrial structure promotes to the county territory economy long-term growth power is stronger, later period should break through the present stage in the manufacturing industry mature county territory through the industrial structure promotion strategy the industry to glide down.

Neoadjuvant chemoradiotherapy(nCRT)and total rectal mesenteric excision are the main standards of treatment for locally advanced rectal cancer(LARC).Lymph node regression grade(LRG)is an indicator of prognosis and response to preoperative nCRT based on postsurgical metastatic lymph node *** histopathological findings in metastatic lymph nodes after nCRT include necrosis,hemorrhage,nodular fibrosis,foamy histiocytes,cystic cell reactions,areas of hyalinosis,residual cancer cells,and pools of mucin.A number of LRG systems designed to classify the amount of lymph node regression after nCRT is mainly concerned with the relationship between residual cancer cells and regressive fibrosis and with estimating the number of lymph nodes existing with residual cancer *** offers significant prognostic information,and in most cases,LRG after nCRT correlates with patient *** this review,we describe the systematic classification of LRG after nCRT,patient prognosis,the correlation with tumor regression grade,and the typical histopathological findings of lymph *** work may serve as a reference to help predict the clinical complete response and determine lymph node regression in patients based on preservation strategies,allowing for the formulation of more accurate treatment strategies for LARC patients,which has important clinical significance and scientific value.
