Objective:This study aimed to describe,optimize and evaluate a method for preparing multivalent co.jugate vaccines by simultaneous co.jugation of two different bacterial capsular polysaccharides(CPs)with tetanus toxoid(TT)as bivalent co.jugates.Methods:Different molecular weights(MWs)of polysaccharides,activating agents and capsular polysaccharide/protein(CP/Pro)ratio that may influence co.jugation and immunogenicity were investigated and optimized to prepare the bivalent co.jugate bulk.Using the described method and optimized parameters,a 20-valent pneumoco.cal co.jugate vaccine and a bivalent meningoco.cal vaccine were developed and their effectiveness was co.pared to that of co.responding licensed vaccines in rabbit or mouse models.Results:The immunogenicity test revealed that polysaccharides with lower MWs were better for Pn1-TT-Pn3 and MenA-TT-MenC,while higher MWs were superior for Pn4-TT-Pn14,Pn6A-TT-Pn6B,Pn7F-TT-Pn23F and Pn8-TT-Pn11A.For activating polysaccharides,1-cyano-4-dimethylaminopyridinium tetrafluoroborate(CDAP)was superior to cyanogen bromide(CNBr),but for Pn1,Pn3 and MenC,N-(3-dimethylaminopropyl)-N’-ethylcarbodiimide hydrochloride(EDAC)was the most suitable option.For Pn6A-TT-Pn6B and Pn8-TT-Pn11A,rabbits immunized with bivalent co.jugates with lower CP/Pro ratios showed significantly stronger CP-specific antibody responses,while for Pn4-TT-Pn14,higher CP/Pro ratio was better.Instead of interfering with the respective immunological activity,our bivalent co.jugates usually induced higher IgG titers than their monovalent co.nterparts.co.clusion:The result indicated that the described co.jugation technique was feasible and efficacious to prepare glyco.onjugate vaccines,laying a solid foundation for developing extended-valent multivalent or co.bined co.jugate vaccines without potentially decreased immune function.

Areco.ine is an alkaloid with important pharmaco.ogical effects in the plant areca nut,which has been demonstrated to be an agonist of muscarinic receptors(M receptor).This study explored the influences of dietary areco.ine on growth performance,intestinal digestion and absorption abilities,antioxidant capacity,and the apical junction co.plex(AJC)of adult grass carp(Ctenopharyngodon idella).Adult grass carp(608to 1512 g)were fed at 6 graded levels of dietary areco.ine(0,0.5,1.0,1.5,2.0,and 2.5 mg/kg diet)for 9weeks.The results suggested that appropriate dietary supplementation of areco.ine(1.0 mg/kg)increased growth parameters and intestinal growth in adult grass carp(Pco.tent of neuropeptide fish substance P(Pco.al permeability(Pco.ine supplementation for adult grass carp was determined to be 1.20,1.21,1.07,and 1.19 mg/kg based on percentage weight gain,lipase activity,serum diamine oxidase,and protein carbonyl,respectively.Overall,to the best of our knowledge,we investigated for the first time the effects and possible mechanisms of dietary areco.ine on intestinal digestive and absorptive capacities and structural integrity in fish and evaluated the appropriate level of supplementation.

DEAR EDITOR,During the breast-feeding period,infants undergo remarkable changes,including rapid physiological and developmental growth.However,little is known about gene expression features and sex-specific gene expression in breast-feeding infants.In this study,we sequenced 32 blood transcriptomes from 16 breast-feeding rhesus macaque(Macaca mulatta)infants and their lactating mothers.We identified 218 differentially expressed genes(DEGs)between infants and mothers,including 91 up-regulated and 127 down-regulated DEGs in the infant group.

Angiokinases, such as vascular endothelial-, fibroblast-and platelet-derived growth factor receptors(VEGFRs, FGFRs and PDGFRs) play crucial roles in tumor angiogenesis. Anti-angiogenesis therapy using multi-angiokinase inhibitor has achieved great success in recent years. In this study, we presented the design, synthesis, target identification, molecular mechanism, pharmaco.ynamics(PD)and pharmaco.inetics(PK) research of a novel triple-angiokinase inhibitor WXFL-152. WXFL-152,identified from a series of 4-oxyquinoline derivatives based on a structureeactivity relationship study,inhibited the proliferation of vascular endothelial cells(ECs) and pericytes by blocking the angiokinase signals VEGF/VEGFR2, FGF/FGFRs and PDGF/PDGFRβ simultaneously in vitro. Significant anticancer effects of WXFL-152 were co.firmed in multiple preclinical tumor xenograft models, including a patientderived tumor xenograft(PDX) model. Pharmaco.inetic studies of WXFL-152 demonstrated high favourable bioavailability with single-dose and co.tinuous multi-dose by oral administration in rats and beagles.In co.clusion, WXFL-152, which is currently in phase Ib clinical trials, is a novel and effective tripleangiokinase inhibitor with clear PD and PK in tumor therapy.

Apoptosis is an important process for maintaining tissue homeostasis and eliminating abnormal cells in multicellular organisms.Abnormality in apoptosis often leads to severe diseases such as cancers. Better understanding of its mechanisms and processes is therefore important. Acco.panying molecular biology events of apoptosis is a series of cellular morphology changes: nucleus co.densation, cell shrinkage and rounding, cell surface blebbing, dynamic blebbing, apoptotic membrane protrusions and nucleus fragmentations and finally, the formation and release of apoptotic bodies. It is difficult to detect cellular changes in the early phase of apoptosis due to the subtle changes at this phase. In the current study, we induced apoptosis in He La cells with H2 O2 and used nuclear dye Hoechst 33258, mitochondria, lysosome and cytoplasmic protein specific aggregation-induced emission fluorogens(AIEgens), TPE-Ph-In, 2 M-DABS and BSPOTPE to successfully perform live cell multiplexed imaging to investigate early apoptosis cellular events. We showed the gradual dissipation of mitochondria membrane potential until it is nondetectable by TPE-Ph-In. Increased mitophagy detected by TPE-Ph-In and 2 M-DABS, co.densed nucleus detected by Hoechst33258, increased permeability and/or reduced integrity of nuclear membrane, and increased intracellular vesicles detected by2 M-DABS are some of the early events of apoptosis.

Among the various scanning techniques, spot and raster scanning are the most frequently adopted. Raster scanning turns off the beam only when each isoenergy slice irradiation is co.pleted. This feature intrinsically solves the leakage dose and frequent beam-switching problems enco.ntered during spot scanning. However, to shorten the delivery time of raster scanning, a sophisticated dose co.trol strategy is required to guarantee dose distribution.In this study, a real-time co.pensation method with raster scanning for synchrotron systems was designed. It is characterized by a small spot-spacing planning strategy and real-time subtraction of the transient number of particles delivered between two planning-spot positions from the planned number of particles of the subsequent raster point.The efficacy of the co.pensation method was demonstrated by performing accurate raster scanning simulations with an in-house simulation co.e and accurate final dose evaluations with a co.mercial treatment planning system.Given the similar dose evaluation criteria under a practical high scanning speed, co.pared with the spot scanning method, the total delivery time of the co.pensated raster scanning method was significantly shortened by 53.3% in the case of irradiating a cubical target and by 28.8% in a pelvic case. Therefore, it can be co.cluded that real-time co.pensated raster scanning with a fast scanning co.figuration can significantly shorten the delivery time co.pared to that of spot scanning. It is important to reduce the pressure on patients caused by prolonged immobilization and to improve patient throughput capacity at particle therapy centers.

Background: Despite the recent development of new therapies, multiple myeloma(MM) remains an incurable disease. Thus, new, efective treatments are urgently needed, particularly for relapsed or refractory MM(RRMM). In an earlier phase I study, a novel form of reco.binant human Apo2L/tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) that is currently in clinical development for the treatment of hematologic malignancies, i.e., circularly permuted TRAIL(CPT), was well tolerated at a dose of 2.5 mg/kg per day and showed promising preliminary activity in patients with RRMM. This phase II, open-label, multicenter study further investigated the eicacy and safety of 2.5-mg/kg per day CPT as single-agent therapy for patients with RRMM.Methods: Patients with RRMM were treated once daily with CPT(2.5 mg/kg, intravenously) for 14 co.secutive days for each 21-day cycle. Clinical response and toxicity were assessed after each treatment cycle.Results: Twenty-seven patients received CPT. Using the European Group for Blood and Marrow Transplantation criteria, we calculated the overall response rate of 33.3% with 1 near-co.plete response(n CR) and 8 partial responses(PRs). The clinical beneit rate(48.1%) included 1 nCR, 8 PRs, and 4 minimal responses. The most co.mon treatmentrelated adverse events(TRAEs) were fever, aspartate aminotransferase elevation, alanine aminotransferase elevation, leuco.enia, rash, neutropenia, and thrombocytopenia. We graded toxicity using the co.mon Toxicity Criteria for Adverse Events, version 3.0, and determined that 37.0% of patients had at least 1 grade 3–4 TRAE.co.clusions: CPT as a single agent can elicit a response in patients with RRMM and is well tolerated. Further clinical investigation is warranted.

To assess the health benefit of polydextrose(PDX) and its blends with inulin Methods The in vitro co.onic model of batch fermentation was used to assess the fermentation performance of PDX and its blends with inulin, as well as fructooligosaccharides and resistent dextrose. 16 S rRNA gene sequencing was performanced to reveal the effects of PDX, inulin and their blends on the fecal microbiota. Results After 24 h of fermentation, PDX stimulated production of more gas, ammonia and several short chain fatty acids(SCFAs), including propionate and butyrate, when co.pared to inulin and fructo-oligosaccharides. Furthermore, PDX degradation showed slowest rate among all the carbohydrates tested, co.sistent with PDX’ property of partly fermentation in distal co.on. On the other hand, the blends of PDX with inulin, with a ratio of 50/50 or 67/33, showed reduced gas production and a similar bifidogenicity to inulin along suggesting a potential bifidogenic factor. Furthermore, 16 S rRNA gene sequencing suggested that the fermented microbial co.munity structures of the blends were closer to inulin but not PDX. Exept for gas production, the metabolic feature of PDX fermentation was due to the significantly higher abundance of both ClostridiumVIII and Erysipelotrichaceaencertaeedis in PDX than other carbohydrates. co.clusion Blends of polydextrose with inulin has similar bifidogenic effect and metabolic profiling, and microbial co.position and structure as inulin, suggesting prebiotic potential.
