Two-sample Mendelian randomization analysis of causal relationship between eczema and autoimmune diseases

Objective:The causal relationship between eczema and autoimmune diseases has not been previously *** study aims to evaluate the causal relationship between eczema and autoimmune ***:The two‐sample Mendelian randomization(MR)method was used to assess the causal effect of eczema on autoimmune *** data from the Genome-Wide Association study Catalog(GWAs)were obtained from the Integrative Epidemiology Unit(IEU)*** eczema and autoimmune diseases,genetic instrument variants(GIVs)were identified according to the significant difference(Psal effect estimates were generated using the inverse‐variance weighted(IVW)*** Egger,maximum likelihood,MR-PREssO,and MR-RAPs methods were used for alternative *** tests,including heterogeneity,horizontal pleiotropy,and leave-one-out analyses,were ***,reverse causality was ***:Genetic susceptibility to eczema was associated with an increased risk of Crohn’s disease(OR=1.444,95%CI 1.199 to 1.738,Ps(OR=1.002,95%CI 1.001 to 1.003,P=0.002).However,no causal relationship was found for the other 6 autoimmune diseases,including systemic lupus erythematosus(sLE)(OR=0.932,P=0.401),bullous pemphigoid(BP)(OR=1.191,P=0.642),vitiligo(OR=1.000,P=0.327),multiple sclerosis(Ms)(OR=1.000,P=0.965),ankylosing spondylitis(As)(OR=1.001,P=0.121),rheumatoid arthritis(RA)(OR=1.000,P=0.460).Additionally,no reverse causal relationship was found between autoimmune diseases and ***:Eczema is associated with an increased risk of Crohn’s disease and ulcerative *** causal relationship is found between eczema and sLE,Ms,As,RA,BP,or vitiligo.

Invasive breast carcinoma(BRCA)is associated with poor prognosis and high risk of ***,it is critical to identify novel biomarkers for the prognostic assessment of ***:The expression data of polo-like kinase 1(PLK1)in BRCA and the corresponding clinical information were extracted from TCGA and GEO ***1 expression was validated in diverse breast cancer cell lines by quantitative real-time polymerase chain reaction(qRT-PCR)and western *** sample gene set enrichment analysis(ssGsEA)was performed to evaluate immune infiltration in the BRCA microenvironment,and the random forest(RF)and support vector machine(sVM)algorithms were used to screen for the hub infiltrating cells and calculate the immunophenoscore(IPs).The RF algorithm and COX regression model were applied to calculate survival risk scores based on the PLK1 expression and immune cell ***,a prognostic nomogram was constructed with the risk score and pathological stage,and its clinical potential was evaluated by plotting calibration charts and DCA *** application of the nomogram was further validated in an immunotherapy ***:PLK1 expression was significantly higher in the tumor samples in TCGA-BRCA ***,PLK1 expression level,age and stage were identified as independent prognostic factors of *** the IPs was unaffected by PLK1 expression,the TMB and MATH scores were higher in the PLK1-high group,and the TIDE scores were higher for the PLK1-low *** also identified 6 immune cell types with high infiltration,along with 11 immune cell types with low infiltration in the PLK1-high tumors.A risk score was devised using PLK1 expression and hub immune cells,which predicted the prognosis of BRCA *** addition,a nomogram was constructed based on the risk score and pathological staging,and showed good predictive ***:PLK1 expression and immune cell infiltration can predict post-immunotherapy pr

Ubiquitously transcribed tetratricopeptide repeat on chromosome X(UTX),also known as lysine(K)-specific demethylase 6A(KDM6A),functions as a tumor suppressor gene or oncogene depending on the tumor type and ***,its tumor-suppressive mechanisms remain largely ***,we investigated the clinical significance and biological effects of UTX expression in pancreatic ductal adenocarcinoma(PDA)and determined the potential mechanisms of its *** expression and its association with clinicopathologic characteristics of PDA patients were analyzed using *** mRNA and protein expression and their regulation in PDA cell lines were measured using quantitative polymerase chain reaction and Western blot *** biological functions of UTX in PDA cell growth,migration,and invasion were determined using gain-and loss-of-function assays with both in vitro and in vivo animal *** expression was reduced in human PDA cell lines and *** UTX expression was associated with poor differentiation and prognosis in *** UTX expression inhibited PDA proliferation,migration,and invasion in vitro and PDA growth and metastasis in vivo,whereas knockdown of UTX expression did the ***,UTX expression was trans-activated by GATA6 ***6-mediated PDA progression could be blocked,at least partially,by silencing UTX *** conclusion,loss of GATA6-mediated UTX expression was evident in human PDA and restored UTX expression suppressed PDA growth and ***,UTX is a tumor suppressor in PDA and may serve as a prognostic biomarker and therapeutic target.

The Impact of seasonal Variation on Clinical Pregnancy and Live Birth Rates in Assisted Reproductive Technology: A Retrospective Cohort study in Hainan

Objective:To investigate the influence of season on live birth and clinical pregnancy rates,as well as assisted reproductive technology(ART)outcomes,in the Hainan ***:Patients were categorized into four groups based on the dates of artificial insemination and transplantation:spring,summer,autumn,or *** main outcome measures were clinical pregnancy rates and live birth *** outcomes included body mass index(BMI),oocyte number,two pronuclei(2PN)cleavage rate,total gonadotropin(Gn)dosage and days,age,2PN fertilization rate,sperm concentration,sperm PR rate,anti-Müllerian hormone(AMH),and endometrial *** semen quality indicators included sperm PR rate,total sperm count,sperm concentration,and total sperm ***:This retrospective cohort study analyzed 2,016 artificial insemination cycles and 1,783 ovarian retrieval cycles from January 2017 to October 2022,and assessed the semen quality of 6,651 outpatients from May 2017 to October *** artificial insemination cycles,sperm PR rate and clinical pregnancy rate were highest in winter,with a statistically significant difference between groups(Ps influenced by both age and sperm PR rate(Ps,the winter group had significantly higher clinical pregnancy,2PN fertilization,and 2PN cleavage rates than the other *** autumn group had higher live birth rates,though not significantly ***,winter months showed higher total sperm concentration and total sperm number compared to other ***:seasonality affected clinical pregnancy and live birth rates in artificial insemination cycles but not in ovarian retrieval cycles in the Hainan *** findings suggest that while there is no need to choose a specific season for ovarian retrieval cycles,artificial insemination in winter may be preferable for patients.

Background:Neuroblastoma is one of the most common extracranial malignant solid tumors in *** homolog 5(ALKBH5)is an RNA N6-methyladenosine(m6A)demethylase that plays a critical role in tumorigenesis and *** assessed the association between single nucleotide polymorphisms(sNPs)in ALKBH5 and the risk of neuroblastoma in a case-control study including 402 patients and 473 non-cancer ***:Genotyping was determined by the TaqMan *** association between ALKBH5 polymorphisms(rs1378602 and rs8400)and the risk of neuroblastoma was evaluated using the odds ratio(OR)and 95%confidence interval(CI).Results:We found no strong association of ALKBH5 rs1378602 and rs8400 with neuroblastoma *** stratification analysis by age,sex,primary site,and clinical stage showed that the rs1378602 AG/AA genotype was associated with a lower risk of neuroblastoma in males(adjusted OR=0.58,95%CI=0.35–0.97,p=0.036)and children with retroperitoneal neuroblastoma(adjusted OR=0.58,95%CI=0.34–0.98,p=0.040).Conclusions:ALKBH5 sNPs do not seem to be associated with neuroblastoma *** studies are required to confirm this negative result and reveal the relationship between gene polymorphisms of the m6A modifier ALKBH5 and neuroblastoma.
