An Overview of Potential Cardioprotective Benefits of Xanthophylls in Atherosclerosis: An Evidence-based Review

Atherosclerosis， as the most prevalent form of cardiovascular disease， is characterized by oxidized low-density lipoprotein (ox-LDL) accumulation in the vascular wall， increased inflammation of the large arteries， dysfunction of the endothelial cells (ECs) and vascular smooth muscle cells (VSMCs)， which may eventually lead to the formation of plaques. Xanthophylls， one of the main groups of carotenoids， have been proposed as preventive agents or adjunct therapies to prevent and slow the progression of atherosclerosis due to their cardioprotective properties. However， the underlying preventive mechanism of action of xanthophylls on the pathogenesis of atherosclerosis remains unclear， and clinical evidence of the effect of xanthophylls on atherosclerosis have not yet been summarized and critically reviewed. In this regard， we conducted a comprehensive literature search in four scientific databases (PubMed， Google Scholar， ScienceDirect and Web of Science) and carefully analyzed the existing evidence to provide meaningful insights on the association between xanthophylls and atherosclerosis from various aspects. Based on the evidence from in vitro and in vivo studies， we explored several potential mechanisms， including antioxidant effect， anti-inflammatory effect， regulation of lipid metabolism， and modulation of ECs and VSMCs dysfunction， and we found that a clear picture of regulatory pathways of xanthophylls on atherosclerosis prevention and treatment is still lacking. In addition， epidemiological studies suggested the possible relationship among high dietary intake of xanthophylls， high plasma/serum xanthophylls and a reduced risk of atherosclerosis. Direct evidence from interventional studies investigating the effect of xanthophylls on atherosclerosis is very sparse， whilst indirect clinical evidence was only limited to astaxanthin and lutein. Therefore， well-designed long-term randomized controlled trials (RCTs) are highly recommended for future studies to investigate the effective dose of different xanthophyll

The oral cavity of each person is home to hundreds of bacterial species.While taxa for oral diseases have been studied using culture-based characterization as well as amplicon sequencing,metagenomic and genomic information remains scarce compared to the fecal microbiome.Here,using metagenomic shotgun data for 3346 oral metagenomic samples together with 808 published samples,we obtain 56,213 metagenome-assembled genomes(MAGs),and more than 64%of the 3589 species-level genome bins(SGBs)contain no publicly available genomes.The resulting genome collection is representative of samples around the world and contains many genomes from candidate phyla radiation(CPR)that lack monoculture.Also,it enables the discovery of new taxa such as a genus Candidatus Bgiplasma within the family Acholeplasmataceae.Large-scale metagenomic data from massive samples also allow the assembly of strains from important oral taxa such as Porphyromonas and Neisseria.The oral microbes encode genes that could potentially metabolize drugs.Apart from these findings,a strongly male-enriched Campylobacter species was identified.Oral samples would be more user-friendly collected than fecal samples and have the potential for disease diagnosis.Thus,these data lay down a genomic framework for future inquiries of the human oral microbiome.

The vagina contains at least a billion microbial cells,dominated by lactobacilli.Here we perform metagenomic shotgun sequencing on cervical and fecal samples from a cohort of 516 Chinese women of reproductive age,as well as cervical,fecal,and salivary samples from a second cohort of 632 women.Factors such as pregnancy history,delivery history,cesarean section,and breastfeeding were all more important than menstrual cycle in shaping the microbiome,and such information would be necessary before trying to interpret differences between vagino-cervical microbiome data.Greater proportion of Bifidobacterium breve was seen with older age at sexual debut.The relative abundance of lactobacilli especially Lactobacillus crispatus was negatively associated with pregnancy history.Potential markers for lack of menstrual regularity,heavy flow,dysmenorrhea,and contraceptives were also identified.Lactobacilli were rare during breastfeeding or post-menopause.Other features such as mood fluctuations and facial speckles could potentially be predicted from the vagino-cervical microbiome.Gut and salivary microbiomes,plasma vitamins,metals,amino acids,and hormones showed associations with the vagino-cervical microbiome.Our results offer an unprecedented glimpse into the microbiota of the female reproductive tract and call for international collaborations to better understand its long-term health impact other than in the settings of infection or pre-term birth.

Disturbances in the microbiota-gut-brain axis may contribute to the development of Alzheimer's disease. Magnesium-L-threonate has recently been found to have protective effects on learning and memory in aged and Alzheimer's disease model mice. However, the effects of magnesium-L-threonate on the gut microbiota in Alzheimer's disease remain unknown. Previously, we reported that magnesium-L-threonate treatment improved cognition and reduced oxidative stress and inflammation in a double-transgenic line of Alzheimer's disease model mice expressing the amyloid-β precursor protein and mutant human presenilin 1(APP/PS1). Here, we performed 16S r RNA amplicon sequencing and liquid chromatography-mass spectrometry to analyze changes in the microbiome and serum metabolome following magnesium-Lthreonate exposure in a similar mouse model. Magnesium-L-threonate modulated the abundance of three genera in the gut microbiota, decreasing Allobaculum and increasing Bifidobacterium and Turicibacter. We also found that differential metabolites in the magnesiumL-threonate-regulated serum were enriched in various pathways associated with neurodegenerative diseases. The western blotting detection on intestinal tight junction proteins(zona occludens 1, occludin, and claudin-5) showed that magnesium-L-threonate repaired the intestinal barrier dysfunction of APP/PS1 mice. These findings suggest that magnesium-L-threonate may reduce the clinical manifestations of Alzheimer's disease through the microbiota-gut-brain axis in model mice, providing an experimental basis for the clinical treatment of Alzheimer's disease.

Magnesium(Mg)based alloys are promising candidates for many applications,but their untreated surfaces usually have low strength and hardness.In this study,a single point diamond turning(SPDT)technique was applied to refine the grain size and improve the mechanical properties of the surface layers of Mg-Li alloys.By refining grains in the topmost layer to the nanometer scale(~60 nm),the surface hardness was found to be enhanced by approximately 60%.The atomic plastic deformation process during the SPDT was then studied by the real-time atomistic molecular dynamics(MD)simulations.A series of MD simulations with different combinations of parameters,including rake angle,cutting speed and cutting depth,were conducted to understand their influences on the microstructural evolution and associated plastic deformation mechanisms on the surface layer of the workpieces.The MD simulation results suggest that using increased rake angle,cutting speed and cutting depth can help to achieve better grain refinement.These simulation results,which provide atomic-level details of the deformation mechanism,can assist the parameter design for the SPDT techniques to achieve the high-performance heterogeneous nanostructured materials.

As one of the deadliest viruses,Ebola virus(EBOV)causes lethal hemorrhagic fevers in humans and nonhuman primates.The suppression of innate immunity leads to robust systemic virus replication of EBOV,leading to enhanced transmission.However,the mechanism of EBOV-host interaction is not fully understood.Here,we identified multiple dysregulated genes in early stage of EBOV infection through transcriptomic analysis,which are highly clustered to Jak-STAT signaling.EBOV VP35 and VP30 were found to inhibit type I interferon(IFN)signaling.Moreover,exogenous expression of VP35 blocks the phosphorylation of endogenous STAT1,and suppresses nuclear translocation of STAT1.Using serial truncated mutations of VP35,N-terminal 1–220amino acid residues of VP35 were identified to be essential for blocking on type I IFN signaling.Remarkably,VP35 of EBOV suppresses type I IFN signaling more efficiently than those of Bundibugyo virus(BDBV)and Marburg virus(MARV),resulting in stable replication to facilitate the pathogenesis.Altogether,this study enriches understanding on EBOV evasion of innate immune response,and provides insights into the interplay between filoviruses and host.

Carbon-supported Pt-based materials are highly promising electrocatalysts.The carbon support plays an important role in the Pt-based catalysts by remarkably influencing the growth,particle size,morphology,dispersion,electronic structure,physiochemical property and function of Pt.This review summarizes recent progress made in the development of carbon-supported Pt-based catalysts,with special emphasis being given to how activity and stability enhancements are related to Pt–C interactions in various carbon supports,including porous carbon,heteroatom doped carbon,carbon-based binary support,and their corresponding electrocatalytic applications.Finally,the current challenges and future prospects in the development of carbon-supported Pt-based catalysts are discussed.

Impact statement The applications of marker gene concatenation have been advanced to resolve the key questions in the Tree of Life.However,the interphylum evolutionary relationship between Korarchaeota of TACK(Thaumarchaeota,Aigarchaeota,Crenarchaeota,Korarchaeota)and Asgard archaea remains uncertain.We applied a marker gene ranking procedure to examine their evolutionary history.Our updated trees showed confident placements of(1)Korarchaeota as the basal branch to other TACK archaea and as a sister group to Asgard archaea;(2)Njordarchaeota at basal branch to Korarchaeota instead of within Asgard archaea.They highlight the importance of evaluating marker genes for phylogeny inference of the Archaea domain.

High-energy-density-batteries working at a wide-temperature range are urgently required in many performance-critical areas.Lithium-sulfur batteries(LSB)are promising high-energy-density batteries that have the potential to maintain high performance at extreme temperatures.However,some problems like severe shuttling and safety issues at high temperatures or sluggish reaction kinetics and charge-transfer process at low temperatures decrease the performance and hinder their practical uses in extreme temperature conditions.Therefore,broadening the working temperature of LSB with stable electrochemical performance becomes a crucial topic.In this paper,the key stumbling blocks for high and low-temperature LSB are comprehensively discussed.The solutions from the aspects of electrolyte and electrode materials are discussed to solve the aggravating shuttle effect and thermal safety issues under high temperature and the sluggish reaction kinetics under low temperature.Moreover,some specific promising solutions to extend the operating temperature range of LSB are also proposed and highlighted,which provide potential research directions on the practical LSB application in future.
