Acceptance and Factors Associated With Participation in Functional Cure–Related Trials Among People Living With HIV: A Cross-sectional Study in Southern China

HIV remains a global health challenge,and research efforts directed towards a functional cure require people living with HIV(PLHIV)in-volvement in clinical trials.Our study assessed willingness to participate in HIV functional cure–related clinical trials and associated factors among PLHIV in Guangzhou,China,using a questionnaire survey approach.We analyzed responses from 718 questionnaires,finding that 71.2%were willing to participate in Phase Ⅲtrials,while 51.7%were willing to participate in Phase I trials and 42.9%expressed acceptability for analytic treatment interruption.Multivariate logistic regression demonstrated that male PLHIV,those with awareness of functional cure,and PLHIV,who had been on antiretroviral therapy(ART)for less than 1 year,were more willing to partic-ipate in Phase Ⅲtrials.Those with a body mass index greater than 24,and those without resistance to ART drug were more willing to participate in Phase I trials.The major motivations for participation in Phase Ⅲtrials were access to cutting-edge treatments(62.6%)and supporting research(55.3%).Safety was the main concern contributing to hesitancy.Our study revealed a high willingness to participate in HIV functional cure–related trials among PLHIV in Guangzhou,China,and willingness varied across different trial phases and was influenced by multiple factors.This study provides valuable references for future clinical trial recruitment strategies and public health policy formulation.

Development of RPA-Cas12a-fluorescence assay for rapid and reliable detection of human bocavirus 1

Human bocavirus(HBoV)1 is considered an important pathogen that mainly affects infants aged 6–24 months,but preventing viral transmission in resource-limited regions through rapid and affordable on-site diagnosis of individuals with early infection of HBoV1 remains somewhat challenging.Herein,we present a novel faster,lower cost,reliable method for the detection of HBoV1,which integrates a recombinase polymerase amplification(RPA)assay with the CRISPR/Cas12a system,designated the RPA-Cas12a-fluorescence assay.The RPA-Cas12a-fluorescence system can specifically detect target gene levels as low as 0.5 copies of HBoV1 plasmid DNA per microliter within 40 min at 37℃without the need for sophisticated instruments.The method also demonstrates excellent specificity without cross-reactivity to non-target pathogens.Furthermore,the method was appraised using 28 clinical samples,and displayed high accuracy with positive and negative predictive agreement of 90.9%and 100%,respectively.Therefore,our proposed rapid and sensitive HBoV1 detection method,the RPA-Cas12a-fluorescence assay,shows promising potential for early on-site diagnosis of HBoV1 infection in the fields of public health and health care.The established RPA-Cas12a-fluorescence assay is rapid and reliable method for human bocavirus 1 detection.The RPA-Cas12a-fluorescence assay can be completed within 40 min with robust specificity and sensitivity of 0.5 copies/μl.

The reaction of carbonyl-to-imine/hemiaminal conversion in the atmospheric aqueous phase is a critical pathway to produce the light-absorbing N-containing secondary organic compounds(SOC).The formation mechanism of these compounds has been wildly investigated in bulk solutions with a low ionic strength.However,the ionic strength in the aqueous phase of the polluted atmosphere may be higher.It is still unclear whether and to what extent the inorganic ions can affect the SOC formation.Here we prepared the bulk solution with certain ionic strength,in which glyoxal and ammonium were mixed to mimic the aqueous-phase reaction.Molecular characterization by High-resolution Mass Spectrometry was performed to identify the N-containing products,and the light absorption of the mixtures was measured by ultraviolet-visible spectroscopy.Thirty-nine N-containing compounds were identified and divided into four categories(N-heterocyclic chromophores,high-molecular-weight compounds with N-heterocycle,aliphatic imines/hemiaminals,and the unclassified).It was observed that the longer reaction time and higher ionic strength led to the formation of more N-heterocyclic chromophores and the increasing of the lightabsorbance of the mixture.The added inorganic ions were proposed to make the aqueous phase somewhat viscous so that the molecules were prone to undergo consecutive and intramolecular reactions to form the heterocycles.In general,this study revealed that the enhanced ionic strength and prolonged reaction time had the promotion effect on the lightabsorbing SOC formation.It implies that the aldehyde-derived aqueous-phase SOC would contribute more light-absorbing particulate matter in the industrial or populated area where inorganic ions are abundant.

This paper comprehensively analyses the first-ever monkeypox outbreak in shenzhen,China,encompassing clinical symptomatology,therapeutic approaches,epidemiological research,and comprehensive laboratory tests,aiming to establish a robust reference for future monkeypox mitigation and management strategies.The investigation involved a thorough investigation of all identified positive cases,including extensive molecular analysis by nucleic acid detection and whole-genome sequencing of the monkeypox virus.Contact tracing and containment of the infected individuals were also undertaken.Three distinct monkeypox cases were identified in this unique outbreak,exhibiting mild and atypical clinical manifestations,primarily fever and rash.All cases were associated with a single transmission chain,primarily facilitated through close contact and homosexual behavior,indicative of a high-risk factor for monkeypox transmission.

What is already known about this topic?Campylobacter is a significant foodborne pathogen that leads to global outbreaks of acute gastroenteritis(AGE)usually affecting less than 30 individuals.Human sapovirus(HuSaV)is an enteric virus responsible for sporadic cases and outbreaks of AGE worldwide.In a study conducted in Beijing,HuSaV detection ranked second after norovirus.What is added by this report?We present a discussion of the first large-scale outbreak of AGE caused by both Campylobacter coli(C.coli)and HuSaV.The outbreak involved a total of 996 patients and exhibited two distinct peaks over a period of 17 days.Through case-control studies,we identified exposure to raw water from a secondary water supply system as a significant risk factor.Among 83 patients,49 samples tested positive for C.coli,39 samples tested positive for HuSaV,and 27 samples tested positive for both pathogens using real-time polymerase chain reaction detection.Furthermore,whole-genome sequencing of 17 C.coli isolates obtained from 17 patients revealed that all isolates belonged to a highly clonal strain of C.coli.What are the implications for public health practice?Outbreaks of AGE resulting from multiple pathogen infections warrant increased attention.This report emphasizes the significance of ensuring the safety of drinking water,particularly in secondary supply systems.

Sensitively detecting antigen of SARS-CoV-2 by NIR-Ⅱ fluorescent nanoparticles

Early detection of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection is an efficient way to prevent the spread of coronavirus disease 2019(COVID-19).Detecting SARS-CoV-2 antigen can be rapid and convenient,but it is still challenging to develop highly sensitive methods for effective diagnosis.Herein,a lateral flow assay(LFA)based on fluorescent nanoparticles emitting in the second near-infrared(NIR-II)window is developed for sensitive detection of SARS-CoV-2 antigen.Benefiting from the NIR-II fluorescence with high penetration and low autofluorescence,such NIR-II based LFA allows enhanced signal-to-background ratio,and the limit of detection is down to 0.01 ng·mL^(−1)of SARS-CoV-2 antigen.In the clinical swab sample tests,the NIR-II LFA outperforms the colloidal gold LFA with higher overall percent agreement with the polymerase chain reaction test.The clinical samples with low antigen concentrations(~0.015–~0.068 ng·mL^(−1))can be successfully detected by the NIR-II LFA,but fail for the colloidal gold LFA.The NIR-II LFA can provide a promising platform for highly sensitive,rapid,and cost-effective method for early diagnosis and mass screening of SARS-CoV-2 infection.
