Oleanolic acid inhibits colon cancer cell stemness and reverses chemoresistance by suppressing JAK2/sTAT3 signaling pathway

Background:Oleanolic acid(OA),a pentacyclic triterpenoid exhibiting speciﬁc anti-cancer properties and highly effective antioxidant activity,was isolated from traditional Chinese medicinal ***,the OA that impacts colon cancer(CC)cells and its underlying mechanisms remain poorly ***:The cytotoxic effect of OA alone or OA-5-Fluorouracil(5-FU)combination on normal and CC cells was analyzed by methyl thiazolyl diphenyl-tetrazolium bromide(MTT).Then,the impact of OA on CC cell lines(LoVo and HT-29)proliferation and stemness were measured using colon formation and tumorsphere formation ***-binding transcription factor 4(Oct4),Prominin-1(CD133),Nanog,and transcription factor sOX-2(sOX2)are cell stemness-related indicators whose expression was assessed usingﬂuorescence qPCR assay,Western blotting,and *** effect of OA on the proliferative potency of CC cells was evaluated using an in vivo ***:The stem-like characteristics and clone production of colon cancer cells were markedly reduced by OA alone or in combination with ***,OA increases the susceptibility of CC cells to 5-FU by blocking the cell stemness-related markers(CD133,Nanog,sOX2,and Oct4)expression levels both in vitro and in vivo,as well as by inactivating the activator of transcription 3(sTAT3 signaling)and Janus kinase 2/signal transducer(JAK2).Conclusion:Theseﬁndings imply that oleanolic acid,both in vitro and in vivo,suppresses the JAK2/sTAT3 pathway,which in turn reverses chemoresistance and decreases colon cancer cell ***,by reducing the recommended amount of 5-FU,this strategy may improve chemotherapeutic effectiveness and minimize undesired side effects.

BACKGROUND:Heatstroke has become a common emergency event in ***(PCT)is used as a biomarker of infection in the emergency department(ED),but its role in rhabdomyolysis(RM)following exertional heatstroke(EHs)remains ***:A retrospective cohort study enrolled patients with EHs from the intensive care unit(ICU).We collected RM biomarkers,inflammation markers,critical disease scores at admission,24 h,48 h,and discharge,and 90-day *** analysis,linear regression and curve fi tting were used to identify the relationship between PCT and ***:A total of 162 patients were recruited and divided into RM(n=56)and non-RM(n=106)*** was positively correlated with myoglobin(Mb),acute hepatic injury,disseminated intravascular coagulation(DIC),sequential Organ Failure Assessment(sOFA)score,and Acute Physiology and Chronic Health Evaluation II(APACHE II)score,with correlation coefficients of 0.214,0.237,0.285,0.454,and 0.368,respectively(all Pstingly,the results of curve fi tting revealed a nonlinear relationship between PCT and RM,and a two-piecewise linear regression model showed that PCT was related to RM with an odds ratio of 1.3 and a cut-off ofsis revealed that RM was associated with higher mortality compared to non-RM cases(P=0.0093).CONCLUsION:High serum PCT concentrations are associated with RM after EHs in critically ill *** PCT concentrations should be interpreted cautiously in patients with EHs in the ED.

The bipolar ionic liquid thruster employs ionic liquid as a propellant to discharge positively and negatively charged high-energy particles under an alternating current(AC)power source,effectively suppressing electrochemical reaction and ensuring charge *** an optimal AC supply power source frequency is critical for sustained stable thruster *** study focuses on the emission characteristics of the ionic liquid thruster under varied AC *** AC power supply was set within the frequency range of 0.5-64 Hz,with eight specific frequency conditions selected for *** experimental results indicate that the thruster operates steadily within a voltage range of±1470 to±1920 V,with corresponding positive polarity current ranging from 0.41 to 4.91μA and negative polarity current ranging from−0.49 to−4.10μ*** voltage polarity switching,an emission delay occurs,manifested as a prominent peak signal caused by circuit capacitance characteristics and a minor peak signal resulting from liquid *** emission test was conducted at 16 Hz,demonstrating approximately 1 h and 50 min of consistent emission before intermittent *** findings underscore the favorable impact of AC conditions within the 8-16 Hz range on the self-neutralization capability of the ionic liquid thruster.

Exploring causality with biliary atresia at different levels:two-sample Mendelian randomization study

Background In recent years,Mendelian randomization(MR)has been widely used to infer causality of related disease risk ***,this strategy has not been applied to biliary atresia(BA).Methods Genome-wide association studies(GWAs)data of 41 inflammatory cytokines,731 immune cell traits,and 1400 metabolites were obtained from public databases as exposure *** outcome information was obtained from a GWAs meta-analysis of 499 children with BA and 1928 normal *** variance weighting was the primary causality *** Q-test,MR-Egger intercept,MR pleiotropy residual sum and outlier,and'leave-one-out'analyses were used for sensitivity *** MR,MR-steiger,and Linkage Disequilibrium score were used to exclude the effects of reverse causality,genetic association,and linkage *** MR results showed that a total of seven traits had potential causal relationships with BA,including three inflammatory cytokines:eotaxin(odds ratio(OR)=1.45,95%confidence interval(CI):1.08 to 1.95,pFDR=0.18),G-CsF(OR=4.21,95%CI:1.75 to 10.13,pFDR=0.05)and MCP-1/MCAF(OR=1.53,95%CI:1.12 to 2.10,pFDR=0.14);three immune cell traits:CD8dim NKT/T cells ratio(OR=0.59,95%CI:0.45 to 0.77,pFDR=0.06),CD8dim NKT counts(OR=0.58,95%CI:0.43 to 0.78,pFDR=0.06),CD8dim NKT/lymphocyte ratio(OR=0.63,95%CI:0.49 to 0.81,pFDR=0.06);one metabolite:X-12261 levels(OR=2.86,95%CI:1.73 to 4.74,pFDR=0.06).Conclusions In this study,eotaxin,G-CsF,MCP-1/MCAF,and X-12261 levels were shown to be risk factors for ***,CD8dim NKT/T cells ratio,CD8dim NKT counts,and CD8dim NKT/lymphocyte ratio were protective factors for *** findings provided a promising genetic basis for the etiology,diagnosis,and treatment of BA.

Diabetic nephropathy(DN)is a severe complication of diabetes,characterized by changes in kidney structure and *** natural product rosmarinic acid(RA)has demonstrated therapeutic effects,including anti-inflammation and anti-oxidative-stress,in renal damage or *** this study,we characterized the heterogeneity of the cellular response in kidneys to DN-induced injury and RA treatment at single cell *** results demonstrated that RA significantly alleviated renal tubular epithelial injury,particularly in the proximal tubular s1 segment and on glomerular epithelial cells known as podocytes,while attenuating the inflammatory response of macrophages,oxidative stress,and cytotox-icity of natural killer *** findings provide a comprehensive understanding of the mechanisms by which RA alleviates kidney damage,oxidative stress,and inflammation,offering valuable guidance for the clinical application of RA in the treatment of DN.

Chronic subdural hematoma(CsDH)is a disease characterized by capsuled blood products that progressively occupy the intracranial space,causing intracranial hypertension and compression in the *** frequently occurs in all demographics,especially in the elderly,but the pathogenesis of CsDH remains *** this review,we discuss the origin,development,and current treatment strategies of *** theﬁrst time,we analyzed the cellular and molecular compositions of hematoma membranes with a focus on neomembrane formation,a complex early-stage interactive event in hematoma *** hypothesize that in patients with CsDH,dural border cells(DBCs)might be induced to synthesize collagen or serum proteins might accumulate at the dura and arachnoid layers at the site of injury,thereby encapsulating the *** formation may trigger inﬂammatory responses after subdural hemorrhage,promotingﬁbroblast-involved extracellular matrix(ECM)deposition and aberrant angiogenesis within the outer ***,ECM deposition and angiogenesis mutually inﬂuence each other and are modulated by inﬂammatory *** illustrating the complex and interactive mechanism of neomembrane formation,we aim to provide a novel insight into CsDH pathogenesis and propose directions for future research as well as advancements in treatment strategies for this disease.

Association of initial assessment variables and mortality in severe pediatric traumatic brain injury

Background Predictive scales have been used to prognosticate long-term outcomes of traumatic brain injury(TBI),but gaps remain in predicting mortality using initial trauma resuscitation *** sought to evaluate the association of clinical variables collected during the initial resuscitation of intubated pediatric severe patients with TBI with in-hospital *** Intubated pediatric trauma patientss with severe TBI(Glasgow coma scale(GCs)score≤8)from January 2011 to December 2020 were *** between initial trauma resuscitation variables(temperature,pulse,mean arterial blood pressure,GCs score,hemoglobin,international normalized ratio(INR),platelet count,oxygen saturation,end tidal carbon dioxide,blood glucose and pupillary response)and mortality were evaluated with multivariable logistic *** Among 314 patients,median age was 5.5 years(interquartile range(IQR):2.2-12.8),GCs score was 3(IQR:3-6),Head Abbreviated Injury score(hAIs)was 4(IQR:3-5),and most had a severe(25-49)Injury severity score(Iss)(48.7%,153/314).Overall mortality was 26.8%.GCs score,hAIs,Iss,INR,platelet count,and blood glucose were associated with in-hospital mortality(all ps age and GCs score increased,the odds of mortality *** 1-point increase in GCs score was associated with a 35%decrease in odds of *** hAIs,INR,and blood glucose increased,the odds of mortality *** each 1.0 unit increase in INR,the odds of mortality increased by 1427%.Conclusions Pediatric patients with severe TBI are at substantial risk for in-hospital *** are needed to examine whether earlier interventions targeting specific parameters of INR and blood glucose impact mortality.
