Isolation and Characterization of Transcriptionally Active Ty1-copia Retrotransposons in Fragaria × ananassa

One possible mechanism suggested for somaclonal variation is the activation of transposable elements. The activation of retrotransposons by stresses and external changes is commonly observed in plants. In previous study, we isolated the reverse transcriptase （RT） gene sequences of Ty 1-copia retrotransposons from tissue culture strawberry （Fragaria x ananassa） plant, but not the transcriptionally active sequence. For further understanding the relationship between retrotransposon and somaclonal varation, in this study, we isolated the transcriptionally active RT gene sequences from strawberry plants subjected to different abiotic stresses. These retrotransposons were activated by spraying strawberry leaves with 2 mmol L^-1 salicylic acid （SA）, 50 mmol L^-1 methyl jasmonate （MeJA）, 50 mmol L^-1 abscisic acid （ABA）, 50 mmol L^-1 2,4- dichlorophenoxyacetic acid （2,4-D） or by inducing callus growth in 2 types of MS media： first medium supplemented with 0.5 mg L^-1 6-benzylaminopurine （6-BA）, 0.5 mg L^-1 gibberellic acid （GA3）, 1.0 mg L^-1 thidiazuron （TDZ）, and 0.1 mg L^-1 2,4-D, and the second medium supplemented with 0.5 mg L^-1 6-BA, 0.5 mg L^-1 GA3, 2.0 mg L^-1 TDZ, and 0.02 mg L^-1 indole butyric acid （1BA）. Analysis of gene sequences of 17 RTs revealed that none of them contained stop codons and/or indels disrupting the reading frame. These different stress-origin transcriptionally active RTs were remarkably similar to each other- FATEXP2-8 and FATEYS9-7 showed 100% sequence identity. Analysis of pylogenetic of these transcriptionally active RTs and the RT sequences from genome showed that there were close phylogenetic relationships of most of the transcriptionally active RTs. The results of this study have contributed to the background information necessary for future studies for evaluating the relationship between retrotransposons and somaclonal variation.

To investigate the impact of particle size on in vitro/vivo performance of praziquantel(PZQ), nanocrystals(NCs) and microcrystals(MCs) of PZQ were prepared using the methods of wet milling and jet milling, respectively. PZQ NCs and MCs were characterized with dynamic light scattering, laser particle size analyzer, transmission electron microscopy, differential scanning calorimetry, X-ray powder diffraction and fourier transform infrared spectroscopy. The average diameters of PZQ NCs and MCs were 364.4 nm and 3.7 μm, respectively. No change in crystalline form was observed. Dissolution tests were performed in two different media, where the cumulative dissolution and dissolution rate of NCs were significantly improved in comparison with those of MCs and KANGQING~? in non-sink condition. Similarly, oral bioavailability of PZQ NCs in beagle dogs was 1.68( P < 0.05) and 1.83 fold( P < 0.01) higher than that of MCs and KANGQING~? . Considering the advantages of in vitro/vivo performance and facile preparation, PZQ NCs may have a great application in the treatment of schistosomiasis.

Reference and Application of Peking Opera Aria in National Vocal music Singing

Peking Opera is one of ancient opera arts in China, known as the ＂national essence＂ , which has a very high artistic achievement. Because of its unique and systematic aria, phonology, performance and modeling, Peking Opera is favored by people, which is an art that vocal music artists mutually research and learn. The Peking Opera aria has already formed a set of professional theories, which has practical significance to guide the national vocal music. It not only can inherit the Peking Opera but also can carry forward the national vocal music. This paper illustrates the reference and application of Peking Opera aria in national vocal music singing according to examples, which provides the reference advice for the development of national vocal music.

When a ferromagnet(FM)couples to a spin glass(SG),the hidden nature of SG may imprint on the FM via their interfacial coupling,probably resulting in elongated magnetic relaxation and enhanced unidirectional and uniaxial *** parallel with the study of interfacial roles,*** coupling and matching,we focused on the influence of the competing interactions in SG(JSG)on the magnetic relaxation properties of SG/FM bilayer and the FM magnetization reversal involving exchange bias field(HE)and coercivity(HC).A fastest decay of relaxation in SG/FM bilayer is found at an intermediate JSG^5.2 erg/cm2 and the relaxation parameters related to JSG are *** the other hand,a pronounced HE of^0.26 kOe is observed at small JSG,and as JSG increases larger than 2.275 erg/cm2,HE decreases by an order of magnitude and levels off,i.e.,a net EB *** is sensitive to JSG only at JSG above 2.275 erg/cm2 and exhibits a minimum value at JSG=4.55 erg/cm2,just below 5.2 erg/cm2 where the relaxation decay is the *** analysis unravels that the net magnetization associated with unidirectional magnetic stabilization and the large-area FM-like domains in SG are favored at small JSG,while with increasing JSG,the SG spins spontaneously reverse and thus the energy barriers are smeared out.A high susceptibility and a strong interfacial coupling lead to FM magnetization reversal under weaker fields and thus a decrease in *** too large JSG(exceeding 5.2 erg/cm2),magnetic frustration extends over the domains in SG meanwhile fierce energy competition occurs frequently,resembling enhanced quantum *** a result,dynamic relaxation and statistic coercivity are both *** work tells us that the independent modulations of unidirectional and uniaxial anisotropies are possible in SG-based systems through precisely controlling JSG,which is desired for application in the read/write head in future data storage media.

Solid dispersion(SD)systems have been extensively used to increase the dissolution and bioavailability of poorly water-soluble *** circumvent the limitations of polyvinylpyrrolidone(PVP)dispersions,HPMC E5 was applied in the formulation process and scaling-up techniques,*** this study,SD of nimodipine(NMP)and corresponding tablets were prepared through solvent method and fluid bed granulating one step technique,*** dissolution media were used to obtain reliable dissolution ***,the stability study of SDs was investigated with storage under high temperature and humidity ***,the solubility of SDs was measured to explore the effect of *** preparations were characterized by DSC,PXRD,and *** improvements in the dissolution rate of NMP were achieved by the ingenious combination of the two *** NMP/PVP/HPMC-SDs released steadily,while the dissolution of single NMP/PVP-SDs decreased rapidly in *** fluid-bed tablets(FB-T)possessed a similar dissolution behavior to the commercial Nimotop TM *** characterization patterns implied that NMP existed in an amorphous state in our ***,the results of stability tests suggested a better stability of the binary SDs.A special cooperative effect of PVP and HPMC was discovered on dissolution characteristics of NMP SDs and tablets,which could be extended to other drugs ***,the bioavailability of FB-T was evaluated in beagle dogs with Nimotop TM as the reference,and the results showed a higher AUC 0–12h value for FB-T.

Objective:To study the chemical constituents in leaves of Cebera ***:Chemical constituents were isolated by using various column chromatography and tho structures were elucidated on basis of physicochemical constants and spectral data ***:Nine compounds were obtained including p-hydroxybenzaldehyde(1),benzamide(2),n-hexadecane acid monoglyceride(3),loliolide(4),β-sitosterol(5),cerberin(6),neriifolin(7),cerleaside A(8), daucosterol(9).Conclusions:Compounds 1-4 are obtained from this genus for the first time.

Improved dissolution and oral absorption by co-grinding active drug probucol and ternary stabilizers mixtures with planetary beads-milling method

The objective of this work is to construct a nanosuspension drug delivery system of probucol,a BCS II drug,in order to improve its dissolution and oral *** wet milling procedure using planetary beads-milling equipment was utilized to grind the raw probucol to ultrafine nanoparticle/nanocrystal aqueous suspension that was further solidified by freeze-drying *** derivatives of different substitution groups and molecular weights,including HPMC,HPC,and MC,were evaluated as the primary stabilizer of probucol *** stabilizers system composed of a primary stabilizer(cellulose derivative,***),a nonionic surfactant(Pluronic R F68),and an anionic surfactant(SDS)was employed to obtain probucol nanosuspension of finer particle size and enhanced dissolution in aqueous *** probucol nanosuspension with good physical stability showed no obvious change of particle size even after storing over 7 d at 4°C or 25°*** solidified probucol nanosuspension with trehalose as the cryoprotectant showed the highest dissolution rate(>60%at 2 h)compared to other *** in vivo pharmacokinetic evaluation indicated about 15-folds higher AUC value of the probucol nanosuspension compared to that of coarse probucol suspension after oral administration to *** probucol nanosuspension prepared by wet-milling and ternary stabilizers system may find wide applications for improving the dissolution and oral absorption of water-insoluble drugs.

Surface electromyography(sEMG)-based gesture recognition is a key technology in the field of human–computer ***,existing gesture recognition methods face challenges in effectively integrating discriminative temporal feature representations from sEMG *** this paper,we propose a deep learning framework named TFN-FICFM comprises a Temporal Fusion Network(TFN)and Fuzzy Integral-Based Classifier Fusion method(FICFM)to improve the accuracy and robustness of gesture ***,we design a TFN module,which utilizes an attention-based recurrent multi-scale convolutional module to acquire multi-level temporal feature representations and achieves deep fusion of temporal features through a feature pyramid ***,the deep-fused temporal features are utilized to generate multiple sets of gesture category prediction confidences through a feedback ***,we employ FICFM to perform fuzzy fusion on prediction confidences,resulting in the ultimate *** study conducts extensive comparisons and ablation studies using the publicly available datasets Ninapro DB2 and *** demonstrate that the TFN-FICFM model outperforms state-of-the-art methods in classification *** research can serve as a benchmark for sEMG-based gesture recognition and related deep learning modeling.
