Inflammation responsive tofacitinib loaded albumin nanomedicine for targeted synergistic therapy in ulcerative colitis

Patients with ulcerative colitis(UC)often loss responses over long term usage of conventional ***,a pan-Janus kinases(JAK)inhibitor is approved for moderate to severe UC treatment,while dose-limiting systemic side effects including infections,cancers and lymphoma limit its popularity of clinical *** study sought to construct an anti-mucosal vascular addressin cell-adhesion molecule-1(anti-MAdCAM-1)antibody modified reactive oxygen species(ROS)responsive human serum albumin-based nanomedicine denoted as THM,to improve the therapeutic efficacy of tofacitinib for UC *** has the drug releasing properties in response to ROS *** vitro studies show that THM selectively adhered to the endothelial cells and had obvious anti-inflammatory effect on ***,the nanomedicine can inhibit the phenotypic switching of M1 macrophages and promote M2 polarization to produce anti-inflammatory medicators during wound *** addition,in vivo fluorescence imaging verified that THM exhibited enhanced preferential accumulation and extended retention in inflamed ***,THM significantly reduced the production of proinflammatory cytokines in the colon and suppressed the homing of T cells to the gut in dextran sodium sulfate induced experimental *** work elucidates that the inflamed colon-targeted delivery of tofacitinib by nanomedicine is promising for UC treatment and sheds light on addressing the unmet medical need.

With the continuous improvement of the performance and the increasing variety of optical mapping and remote sensing satellites,they have become an important support for obtaining global accurate surveying and mapping remote sensing *** present,optical mapping and remote sensing satellites already have sub-meter spatial resolution capabilities,but there is a serious lag problem in mapping and remote sensing information *** is urgent to develop intelligent mapping and remote sensing satellites to promote the transformation and upgrading to real-time intelligent ***,based on the three imaging systems of the optical mapping and remote sensing satellites and their realization methods and application characteristics,this paper analyzes the applicable system of the intelligent mapping and remote sensing ***,according to the application requirements of real-time,intelligence,and popularization,puts forward the design concept of integrated intelligent remote sensing satellite integrating communication,navigation,and remote sensing and focuses on the service mode and integrated function composition of intelligent remote sensing *** expounds on the performance and characteristics of the Luojia-301 satellite,a new generation of intelligent surveying and mapping remote sensing scientific test *** finally summarizes and prospects the development and mission of intelligent mapping remote sensing ***-301 satellite integrates remote sensing and communication *** explores an efficient and intelligent service mode of mapping and remote sensing information from data acquisition to the application terminal and provides a real service verification platform for on-orbit processing and real-time transmission of remote sensing data based on space-ground internet,which is of great significance to the construction of China’s spatial information network.

Developing bifunctional nanoplatforms integrating advanced biological imaging with high-performance therapeutic functions is interesting for nanoscience and ***,thin-layered FePS3 nanosheets modified with amine polyethylene glycol(FePS3-PEG)are synthesized by ultrasonicated exfoliation of bulk Fe PS3 single crystals under Ar atmosphere and in situ chemical functionalization with amine *** prepared Fe PS3-PEG nanosheets give a photothermal conversion efficiency of 22.1%under 808-nm laser and evident T1relaxation(r1=2.69(mmol L^(-1))^(-1)S^(-1))in external magnetic *** experimental results in vitro and in vivo demonstrate that Fe PS3-PEG nanosheets have good biocompatibility and exhibit excellent photothermal therapy(PTT)effect under clear T1-weighted magnetic resonance imaging(MRI).The characteristics of Fe PS3-PEGnanosheets enable them to be a promising nanomedicine for T1-weighted MRI-guided near-infrared PTT of cancers.

In vivo fluorescence imaging in the second near-infrared window(NIR-II)has been considered as a promising technique for visualizing ***,the definition of the NIR-II region and the mechanism accounting for the excellent performance still need to be ***,we simulate the photon propagation in the NIR region(to 2340 nm),confirm the positive contribution of moderate light absorption by water in intravital imaging and perfect the NIR-II window as 900–1880 nm,where 1400–1500 and 1700–1880 nm are defined as NIR-IIx and NIR-IIc regions,***,2080–2340 nm is newly proposed as the third near-infrared(NIR-III)window,which is believed to provide the best imaging *** wide-field fluorescence microscopy in the brain is performed around the NIRIIx region,with excellent optical sectioning strength and the largest imaging depth of intravital NIR-II fluorescence microscopy to *** also propose 1400 nm long-pass detection in off-peak NIR-II imaging whose performance exceeds that of NIR-IIb imaging,using bright fluorophores with short emission wavelength.

Owing to the complicated physiological microenvironment,nanosystems designed with the possibility of noninvasive real-time monitoring the therapeutic progression in the second near-infrared channel（NIR-Ⅱ,1000-1700 nm） could substantially improve the current cancer ***,we designed a novel NIR-Ⅱ theranostic nanoprobe PSY by incorporating organoplatinum（Ⅱ） metallacycles P1 and organic NIR-Ⅱ molecule dye SY1030 into FDA-approved polymer Pluronic *** in vitro and in vivo studies suggested that PSY is capable of being internalized into glioma U87 *** addition,it shows excellent photostability and minimal background to real-time monitor the process of therapy in the NIR-Ⅱ ***,in U87 MG xenografts and orthotopic breast tumor,PSY demonstrated significantly improved anticancer efficacy as compared to clinically approved Pt（Ⅱ）-based anticancer drug,*** engineered nano-cocktail PSY offers a simple strategy of delivering organoplatinum（Ⅱ） macrocycle P1 and NIR-Ⅱ fluorophore SY1030 as a cocktail of diagnosis and therapy and highlights its promising capacity for future cancer treatment.

The construction of multifunctional probes with small molecules has broad prospects in biomedical ***,the complex synthesis steps of probes and the dose and laser trigger ratio of difficult-to-match probes greatly limit the development of this ***,reasonably an intrinsic NIR-Ⅱ multifunctional DD-A-DD scaffold molecular dye（SYL） was designed and then incorporated into DSPE mPEG5000 to facilitate the construction of biocompatible NIR-Ⅱ fluorescence and photoacoustic（PA）.In vivo studies confirmed that SYL NP exhibited bright NIR-Ⅱ fluorescence and PA signals in the tumor area with good signal-to-noise ratio（S/B）.At the same time,SYL NPs effectively inhibited tumor growth under laser irradiation for photothermal therapy without significant side *** promising results highlight that SYL NPs are potential cancer diagnostics（NIR-Ⅱ region） and therapeutic platforms.

High resolution imaging is achieved using increasingly larger apertures and successively shorter *** aperture synthesis is an important high-resolution imaging technology used in *** long baseline amplitude interferometry is susceptible to uncontrollable phase fluctuations,and the technical difficulty increases rapidly as the wavelength *** intensity interferometry inspired by HBT experiment is essentially insensitive to phase fluctuations,but suffers from a narrow spectral bandwidth which results in a lack of effective *** this study,we propose optical synthetic aperture imaging based on spatial intensity *** not only realizes diffraction-limited optical aperture synthesis in a single shot,but also enables imaging with a wide spectral bandwidth,which greatly improves the optical energy efficiency of intensity *** this method is insensitive to the optical path difference between the *** and experiments present optical aperture synthesis diffraction-limited imaging through spatial intensity interferometry in a 100 nm spectral width of visible light,whose maximum optical path difference between the sub-apertures reaches 69λ.This technique is expected to provide a solution for optical aperture synthesis over kilometer-long baselines at optical wavelengths.
