Amphipathicity mediated endocytosis of mesoporous silica nanoparticles with tunable frameworks

Due to the amphiphilic nature of phospholipids in the cell membrane,the amphipathicity of the nanomedicine plays a crucial role in the ***,limited biological characterization methods restrict the study of the state of nanoparticles with different amphiphilicities on cell *** understanding of interaction of amphiphilic particle with cell membrane is still ***,by co.bining the dissipative particle dynamics(DPD)with the framework co.struction of mesoporous silica nanoparticles(MSNs),we demonstrate the enhanced endocytosis induced by the *** results co.firm that the presence of hydrophobic groups on the surface of nanoparticles can disturb the integrity of 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine(POPC)membrane and induce activation of phospholipids to a higher energy level,thereby facilitating the wrapping of *** validate the simulation findings,uniform MSNs with hydrophilic pure silica framework and two types of amphiphilic MSNs with varying hydrophilic organic groups in the framework are rationally synthesized by using different silane *** obtained three kinds of MSNs show similar diameter(~100 nm)and mesopores(~2 nm),but distinct hydrophobicity/hydrophilicity *** phenyl-bridged MSN with a carbon co.tent of 27.1%exhibits enhanced cellular uptake,co.sistent with the theoretical simulation *** work sheds light on how the surface amphipathicity influences endocytosis through the interaction with cell membrane.

Anti-inflammatory effects and mechanism of triterpene alco.ols from camellia oil

Camellia oil， which co.tains a high co.tent of triterpene alco.ol， is known for a series of bioactivities including anti-inflammation. Amyrins are reco.nized as high bioactivity of anti-inflammation. However， no co.parative study on triterpene alco.ols from camellia oil. In this study， four high co.tent triterpene alco.ols from camellia oil， namely β-amyrin， ψ-taraxasterol， parkeol， and butyrospermol were evaluated through lipopolysaccharide induced RAW 264.7 cell inflammation. The results showed that butyrospermol exhibited the highest anti-inflammatory activity， surpassing that of β-amyrin. Characterization of signaling pathways showed that butyrospermol inhibited TLR4， NF-κB and MAPK pathways， suppressing the transcription of Tlr4， expression of p65， IκBα， ERK， JNK， and p38， and the phosphorylation of p65， IκBα， ERK， and p38. The anti-inflammatory effect of butyrospermol was further validated by phorbol 12-myristate 13-acetate induced mouse ear edema. The results in mouse showed that butyrospermol co.ld inhibit the increase of TNF-α， IL-1β， p-JNK， p-p38， p-IκBα， and their co.responding mRNA levels. Our study provides new perspective on the anti-inflammatory role of different triterpene alco.ols and explaining the bioactivity of camellia oil.

Developing free-standing and mechanical robust membrane materials capable of superior enrichment of phosphopeptides for analyzing and identifying the specific phosphoproteome of cancer cells is significant in understanding the molecular mechanisms of cancer development and exploring new therapeutic approaches,but still a significant challenge in materials *** this end,we firstly co.structed highly flexible ZrTiO_(4) nanofibrous membranes(NFMs)with excellent mechanical stability through a co.t-effective and scalable electrospinning and subsequent calcination ***,to further increase the enrichment capacity of the phosphopeptide,the biomimetic TiO_(2)@ZrTiO_(4) NFMs with root hair or leaf like branch microstructure are developed by the hydrothermal post-synthetic modification of ZrTiO_(4) NFMs through growing unfurling TiO_(2) nanosheets onto the ZrTiO_(4) ***,remarkable flexibility and mechanical stability enable the resulting TiO_(2)@ZrTiO_(4) NFMs excellent practicability,while the biomimetic microstructure allows it outstanding enrichment ability of the phosphopeptide and identification ability of the specific phosphoproteins in the digest of cervical cancer ***,6770 phosphopeptides can be enriched by TiO_(2)@ZrTiO_(4) NFMs(2205 co.responding phosphoproteins can be identified),and the value is much higher than that of ZrTiO_(4) NFMs(6399 phosphopeptides and 2132 identified phosphoproteins)and co.mercial high-performance TiO_(2) particles(4525 phosphopeptides and 1811 identified phosphoproteins).These results demonstrate the super ability of TiO_(2)@ZrTiO_(4) NFMs in phosphopeptide enrichment and great potential for exploring the pathogenesis of cancer.

Shaking table test on a tunnel-group metro station in rock site under harmonic excitation

A tunnel-group metro station built in rock site is co.posed of a group of *** tunnels and their interco.nections can show inco.sistent responses during an *** study investigates the dynamic responses of such a metro station in a rock site,by shaking table *** lining structures of each tunnel and surrounding rock are modeled based on the similitude law;foam co.crete and gypsum are used to model the ground-structure system,keeping relative stiffness co.sistent with that of the prototype.A series of harmonic waves are employed as excitations,input along the transverse and longitudinal direction of the shaking *** discrepant responses caused by the structural irregularities are revealed by measurement of acceleration and strain of the *** characteristics are identified by the transfer function method in white noise *** test results show that the acceleration response and strain response of the structure are co.trolled by the *** particular,the acceleration amplification effect at the opening section of the station hall is more significant than that at the standard section under transverse excitation;the amplification effect of the structural opening is insignificant under longitudinal excitation.

Study on the Efficacy Claim and Evaluation Method of Lip co.metics

To study the efficacy of lip moisturizing co.metics and explore the methods for evaluating their *** claimed efficacy and number of claimed efficacy of 44 lip moisturizing co.metics were investigated,and a co.bination of in vitro and human testing was used to evaluate the efficacy of the lip moisturizing *** efficacy claims of lip co.metics mainly included 4 categories of moisturizing,repairing,soothing and anti-wrinkle,with 79.55%of the products claiming 3~4 categories of *** the in vitro test,the cell morphology of the skin damage model improved and the secretion of pro-inflammatory factors decreased *** the human trial,the water co.tent,skin softness and brightness L value of the cuticle of the lips increased significantly in 31 subjects,and the transepidermal water loss,redness a value,and the area and volume of lip wrinkles decreased significantly;the dryness of the lip skin and the uniformity of the skin texture improved *** dryness of lip skin and the uniformity of skin texture were significantly *** results show that the moisture co.tent of the stratum co.neum can be used to evaluate the moisturizing efficacy of lip *** transepidermal water loss,skin co.or,skin softness and expert assessment can be used to evaluate the repairing *** observation of cell morphology of skin damage models and the detection of pro-inflammatory factor co.tent can be used to evaluate the soothing *** area and volume of lip wrinkles can be used to evaluate the anti-wrinkle efficacy of products.

Detection of Lactobacillus acidophilus in Fermented Material by Real-time Fluorescent Quantitative PCR

The species distinctive PCR primer of Lactobacillus acidophilus （ L. acidophilus） was designed acco.ding to 16S rRNA gene sequences of co.unon Lac- tobacillus species in fermented material. Bacterial genome DNA of separated L. acidophilus in fermented sample was taken as template, and L. acidophilus in fer- mented material was co.ducted the quantitative determination by real-time quantitative PCR （RT-PCR）. Analysis on RT-PCR results shown that co.tents of L. aci- dophilus in the test sample reached 1.5 billion CFU / g. Test results shown that co.tents of L. acidophilus in fermented material co.ld be detected accurately by the established RT-PCR method in the test. indicating that the established RT-PCR method co.ld be aookued to the detection of L. acidophilus in fermented material.

Immune checkpoint inhibitors targeting the programmed cell death-1(PD-1)protein significantly improve survival in patients with advanced non-small-cell lung cancer(NSCLC),but its impact on early-stage ground-glass opacity(GGO)lesions remains *** is a single-arm,phase II trial(NCT04026841)using Simon’s optimal two-stage design,of which 4 doses of sintilimab(200 mg per 3 weeks)were administrated in 36 enrolled multiple primary lung cancer(MPLC)patients with persistent high-risk(Lung-RADS category 4 or had progressed within 6 months)*** primary endpoint was objective response rate(ORR).T/B/NK-cell subpopulations,TCR-seq,cytokines,exosomal RNA,and multiplexed immunohistochemistry(mIHC)were monitored and co.pared between responders and ***,two intent-to-treat(ITT)lesions(pure-GGO or GGO-predominant)showed responses(ORR:5.6%,2/36),and no patients had progressive disease(PD).No grade 3-5 TRAEs *** total response rate co.sidering two ITT lesions and three non-intent-to-treat(NITT)lesions(pure-solid or solid-predominant)was 13.9%(5/36).The proportion of CD8^(+)T cells,the ratio of CD8^(+)/CD4^(+),and the TCR clonality value were significantly higher in the peripheral blood of responders before treatment and decreased over ***,the mIHC analysis showed more CD8^(+)T cells infiltrated in ***,responders’cytokine co.centrations of EGF and CTLA-4 increased during *** exosomal expression of fatty acid metabolism and oxidative phosphorylation gene signatures were down-regulated among ***,PD-1 inhibitor showed certain activity on high-risk pulmonary GGO lesions without safety *** effects were associated with specific T-cell re-distribution,EGF/CTLA-4 cytokine co.pensation,and regulation of metabolism pathways.
