Dear editor,The left atrial appendage(LAA)is the most common site for thrombus formation in patients with atrial fibrillation(AF).[1]We present the case of a patient with AF who underwent LAA closure(LAAC)with Watchman(Atritech,Boston Scientific,USA),in whom extracranial systemic embolic events(SEEs)with left atrial thrombus were observed after 4 years,and discuss relevant literature to improve the understanding of SEEs.

Background:Pancreatic cancer(PC)is a highly deadly malignancy with few effective therapies.We aimed to unmask the role that long non-coding RNA small nucleolar RNA host gene 6(SNHG6)plays in PC cells by targeting far upstream element binding protein 1(FUBP1)via microRNA-26a-5p(miR-26a-5p).Methods::SNHG6 expression was predicted by bioinformatics,followed by verification via reverse transcription quantitative polymerase chain reaction.Then,the interactions among SNHG6,miR-26a-5p,and FUBP1 were detected through online software analysis,dual luciferase reporter assay and RNA pull-down.After that,cells were treated with different small interfering RNAs and/or mimic to determine the interactions among SNHG6,miR-26a-5p,and FUBP1 and their roles in PC cells.Finally,the role of SNHG6 in tumor growth in vivo was evaluated by measuring the growth and weight of transplanted tumors in nude mice.A t-test,one-way and two-way analysis of variance were used for data analysis.Results::Compared with that in normal tissues,SNHG6 was highly expressed in PC tissues(1.00±0.05 vs.1.56±0.06,t=16.03,P<0.001).Compared with that in human pancreatic duct epithelial cells(HPDE6-C7),SNHG6 showed the highest expression in PANC-1 cells(1.00±0.06 vs.3.87±0.13,t=34.72,P<0.001)and the lowest expression in human pancreatic cancer cells(MIAPaCa-2)(1.00±0.06 vs.1.41±0.07,t=7.70,P=0.0015).Compared with the levels in the si-negative control group,SNHG6(0.97±0.05 vs.0.21±0.06,t=16.85,P<0.001),N-cadherin(0.74±0.05 vs.0.41±0.04,t=8.93,P<0.001),Vimentin(0.55±0.04 vs.0.25±0.03,t=10.39,P<0.001),andβ-catenin(0.62±0.05 vs.0.32±0.03,t=8.91,P<0.001)were decreased,while E-cadherin(0.65±0.06 vs.1.36±0.07,t=13.34,P<0.001)was increased after SNHG6 knockdown or miR-26a-5p overexpression,accompanied by inhibited cell proliferation,migration,and invasion.SNHG6 overexpression exerted the opposite effects.SNHG6 upregulated FUBP1 expression by sponging miR-26a-5p.Silencing SNHG6 blocked the growth of PC in vivo.Conclusion::Silencing SNHG6 might ameliorate PC through inhib

Motivated by two norm equations used to characterize the Friedrichs angle,this paper studies C^(*)-isomorphisms associated with two projections by introducing the matched triple and the semi-harmonious pair of projections.A triple(P,Q,H)is said to be matched if is a Hilbert C^(*)-module,P and Q are projections on H such that their infimum P∧Q exists as an element of L(H),where L(H)denotes the set of all adjointable operators on H.The C^(*)-sub algebras of L(H)generated by elements in{P-P∧Q,Q-P∧Q,I}and{P,Q,P∧Q,I}are denoted by i(P,Q,H)and o(P,Q,H),respectively.It is proved that each faithful representation(π,X)of o(P,Q,H)can induce a faithful representation(π,X)of i(P,Q,H)such that π~(P−P∧Q)=π(P)−π(P)∧π(Q),π~(Q−P∧Q)=π(Q)−π(P)∧π(Q)..When(P,Q)is semi-harmonious,that is,R(P+Q) and R(2I−P−Q) are both orthogonally complemented in H,it is shown that i(P,Q,H)and i(I-Q,I-P,H)are unitarily equivalent via a unitary operator in L(H).A counterexample is constructed,which shows that the same may be not true when(P,Q)fails to be semi-harmonious.Likewise,a counterexample is constructed such that(P,Q)is semi-harmonious,whereas(P,I-Q)is not semi-harmonious.Some additional examples indicating new phenomena of adjointable operators acting on Hilbert C^(*)-modules are also provided.

The impact of different iron metabolism processes(DIMP)on ovarian cancer remains unclear.In this study,we employed various gene chips and databases to investigate the role of DIMP in the initiation and development of ovarian cancer.cBioPortal was used to determine mutations in DIMP-associated genes in ovarian cancer.Kaplan-Meier plotter was used to examine the influence of DIMP on the prognosis of ovarian cancer.By analyzing 1669 serous ovarian cancer cases,we identified a range of mutations in iron metabolism genes,notably in those coding for the transferrin receptor(19%),melanotransferrin(19%),and ceruloplasmin(10%)in the iron import process,and glucose-6-phosphate isomerase(9%),hepcidin antimicrobial peptide(9%),metal regulatory transcription factor 1(8%),and bone morphogenetic protein 6(8%)in the iron regulation process.Compared to the unaltered group,the group with gene alterations exhibited a higher tumor mutation burden count(43 vs.54)and more advanced histologic grade(78.19%vs.87.90%).Compared to the normal ovarian counterparts,a reduction in expression was observed in 9 out of the 14 genes involved in iron utilization and 4 out of the 5 genes involved in iron export in ovarian cancer;in contrast,an increase in expression was observed in 2 out of the 3 genes involved in iron storage in ovarian cancer.Furthermore,in cisplatin-resistant cells compared to cisplatin-sensitive ones,the expression of all genes in iron storage and 13 out of 14 genes in iron import was decreased,while that of 8 out of the 10 genes in iron utilization was increased.In addition,survival curve analysis indicated that a higher expression in the majority of genes in the iron import process(12/21),or a reduced expression in most genes in the iron export process(4/5)correlated with poor progression-free survival.Additionally,TGF-βcould regulate the expression of most iron metabolism-associated genes;particularly,expression of genes involved in the iron storage process(2/2)was inhibited after TGF-β1 or TGF-β2 treatment.In conclusion,DIMP

The electroencephalogram(EEG)rhythm and functional near-infrared spectroscopy(fNIRS)activation levels have not been compared between a healthy control group(HCG)and methamphetamine user group(MUG)with different addiction histories.This study used 64-electrode EEG and fNIRS to conduct an experiment that analyzed the resting and craving states.The EEG and fNIRS data of 56 participants were collected,including 14 healthy participants,14 methamphetamine users with an addiction history of 0.5–5 years,14 users with an addiction history of 5–10 years,and 14 users with an addiction history of 10–15 years.Isolated effective coherence(iCoh)within the brain network was used to process the EEG data.Statistical analysis was performed to compare differences in iCoh among the delta,theta,alpha,beta,and gamma bands and explore oxyhemoglobin activation levels in the ventrolateral prefrontal cortex,dorsolateral prefrontal cortex,orbitofrontal cortex,and frontopolar prefrontal cortex(FPC)of the control group.Finally,the Kmeans,Gaussian mixed model(GMM),linear discriminant analysis(LDA),support vector machine(SVM),Bayes,and convolutional neural networks(CNN)algorithms were used to classify methamphetamine users based on drug and neutral images.A 3-class accuracy was achieved.Changes in EEG and fNIRS activation levels of HCG and MUG with varied addiction histories were demonstrated.

BACKGROUND Tissue resident memory T(TRM)cells have been reported to play a significant role in the pathogenesis and relapse of chronic eczema.AIM To compare the efficacy and safety of the intralesional injection of 5-fluorouracil(5-FU)and triamcinolone(TA)with those associated with TA alone for the treatment of chronic eczema.METHODS A total of 168 patients were randomized to 5-FU+TA or TA groups and received a one-time intralesional injection of 5-FU+TA or TA only.Biopsies were collected before and 2 wk after treatment for evaluation of histopathological changes.All patients were followed up monthly for up to 1 year.RESULTS No serious adverse event was observed in either group.Although the mean atopic dermatitis severity index scores and effective rates were comparable between the two groups after 2 wk of treatment,the relapse rate was significantly lower in the 5-FU+TA group than in the TA group.Histological examination showed significantly fewer CD8^(+)and CD103^(+)T cells but not CD4^(+)T cells in the 5-FU+TA group.CONCLUSION One-time intralesional injection of 5-FU+TA is effective and safe for chronic eczema treatment and can further reduce the retention of T_(RM) cells in the lesional skin and the relapse rate of chronic eczema.

Parkinson's disease(PD)is a common neuro-degenerative disorder caused by the loss of dopamine neu-rons in the substantia nigra and the formation of Lewy bod-ies,which are mainly composed of alpha-synuclein fibrils.Alpha-synuclein plays a vital role in the neuroinflamma-tion mediated by the nucleotide-binding oligomerization domain-,leucine-rich repeat-,and pyrin domain-containing protein 3(NLRP3)inflammasome in PD.A better under-standing of the NLRP3 inflammasome-mediated neuroin-flammation and the related mitochondrial impairment during PD progression may facilitate the development of promis-ing therapies for PD.This review focuses on the molecular mechanisms underlying NLRP3 inflammasome activation,comprising priming and protein complex assembly,as well as the role of mitochondrial impairment and its subsequent inflammatory effects on the progression of neurodegenera-tion in PD.In addition,the therapeutic strategies targeting the NLRP3 inflammasome for PD treatment are discussed,including the inhibitors of NLRP3 inflammatory pathways,mitochondria-focused treatments,microRNAs,and other therapeutic compounds.

OBJECTIVE:To investigate the efficacy of the extract from Yiyuan Yiliu Tang(益元抑瘤汤,YYYLT)on human lung adenocarcinoma cells A549 and human hepatoma cells Bel7402.METHODS:The cancer cell lines were treated with various concentrations(0,100,200,300,and400μg/m L)of the crude water extract of YYYLT and then cell viability,toxicity,cytokine secretion,and cell cycle/apoptosis were determined by MTT assay,LDH assay,and flow cytometry,respectively.RESULTS:The extract from YYYLT significantly suppressed the proliferation of the cancer cell lines and the release of interleukin-2 and tumor necrosis factor-αin a dose-dependent manner.The extract also promoted apoptosis,caused cell cycle arrest at G0/G1 phase,and increased the expression of caspase-3,B-cell lymphoma-2(Bcl-2),and Bcl-2-associated X proteins.CONCLUSION:The extract from YYYLT might be a potential treatment for human lung and liver cancers.

The influence of nanoparticle shape on bilateral exocytosis from Caco-2cells

Nanoparticles are easily to be taken up by cells but hard to be transported across epithelia, which plays an important role in absorption and toxicity. This paper aims to elucidate the effect of nanoparticle shape on bilateral exocytosis of Caco-2 cells. The fluorescent resonance energy transfer(FRET) probes(DiO and Dil)were utilized to label nanospheres and nanorods for determining and tracking nanoparticles. The results showed that more nanorods were internalized into Caco-2 cells than nanospheres. But the apical exocytosis rate of nanospheres from Caco-2 cells was faster than that of nanorods significantly. In addition, only less than 2% of intact nanoparticles were transported across monolayers for both nanoparticles, but the exocytosis behaviors between them was absolutely different. Compared to nanospheres, nanorods preferred basolateral exocytosis to apical exocytosis. Therefore, the shape is a critical parameter in cellular uptake and transport, even intracellular fate, which should be primarily considered in design of oral nanoparticles.
