Objective:To investigate the analgesic substances in the aerial part of Urtica fissa(Urticae Fissae Herba),commonly used for rheumatoid and rheumatism arthritis.Methods:The analgesic constituents were isolated with the active guidance of hot plate and acetic acid writhing models,and identified by comprehensive spectroscopic analysis.Results:Thirteen alkaloids(1–13),two lignans(14,15),and three amides(16–18)were isolated from the active fractions.Among them,compound 1 was a new alkaloid,and compound 6 was a new natural product.The activity evaluation in vivo indicated that various pyrrole alkaloids(1,3,6,and 12)possessed significant analgesic activities,they could significantly inhibit the mice pain response induced by acetic acid and hot plate at the dosage of 2 mg/kg BW.Conclusion:The study revealed that the pyrrole alkaloids played important roles in the analgesic activities of Urticae Fissae Herba.

Background Aripiprazole(ARI)is often prescribed alone or in combination with other second-generation antipsychotics(SGAs)to treat patients with schizophrenia.However,this may increase the potential clinical significance of drug-drug interactions.Therapeutic drug monitoring(TDM)is an important and fundamental tool both when administering ARI alone and in combination with other SGAs to monitor ARI pharmacokinetics,adjust the dosage and thereby achieve more effective and safer treatment.Aims This study retrospectively investigated the effects of four SGA comedications(clozapine,risperidone,quetiapine(QTP)and olanzapine)and other potential factors(sex,age and ARI dose)on the serum concentrations of ARI and dehydroaripiprazole(DARI)in Chinese patients with schizophrenia using TDM data.Methods High-performance liquid chromatography was used to test the serum concentrations of ARI,DARI and ARI+DARI.In addition,steady-state dose-adjusted serum concentrations(ie,concentration-to dose ratios,C:D ratios)of ARI,DARI and ARI+DARI;sex;age;ARI dose and SGA comedication dose between 299 inpatients with schizophrenia who received ARI or SGA comedication were all collected and analysed.Spearman’s correlation and multiple linear regression analysis were used to evaluate bivariate associations between ARI dose and serum ARI and DARI concentrations and describe the effect of independent variables on serum ARI and DARI concentrations,respectively.Results There were significant differences in the C:D ratios of ARI(χ^(2)=−3.21,p=0.001)and ARI+DARI(χ^(2)=−2.50,p=0.01)between the ARI and SGA groups,as well as in the C:D ratios of ARI(χ^(2)=−3.59,p<0.001)and ARI+DARI(χ^(2)=−3.10,p=0.002)between the female patients in the two groups.Of the four SGAs,only QTP had significant effects on the C:D ratios of ARI(Z=−4.12,p<0.001)and ARI+DARI(Z=−3.62,p<0.001)when compared with the ARI group in the whole sample and on the C:D ratios of ARI,DARI and ARI+DARI(Z=−3.96,p<0.001;Z=−2.22,p=0.03;Z=−3.75,p<0.001,respectively)in women when compared with their

Considerable evidence has revealed that essential oils and their main constituents possess anti-depressant and anxiolytic properties.In the current study,we report the effect ofβ-citronellol,the main component of rose essential oil,on depressive-like and anxiety-like behaviors in chronic restraint stress(CRS)mice.We found that chronic inhalation ofβ-citronellol for 14 days could increase locomotor activity in the open field test,decrease the percentage of immobility duration in the forced swimming test,and increase open arms exploration in elevated plus-maze test in CRS mice.Western blot experiment shows that chronicβ-citronellol inhalation rescues parvalbumin(PV)expression loss in the prefrontal cortex(PFC)of CRS mice.Correlation analysis reveals a strong relationship between the PV expression in PFC and the percentage of sucrose preference of the mice.These findings indicate the relationships between the PV gene expression of PFC and the effects ofβ-citronellol inhalation.

Effects of regulating intestinal microbiota on anxiety symptoms: A systematic review

Background Anxiety symptoms are common in mental diseases and a variety of physical disorders, especially in disorders related to stress. More and more basic studies have indicated that gut microbiota can regulate brain function through the gut-brain axis, and dysbiosis of intestinal microbiota was related to anxiety. However, there is no specific evidence to support treatment of anxiety by regulating intestinal microbiota. Aims To find evidence sup porting improvement of anxiety symptoms by regulation of intestinal microbiota. Methods This systematic review of randomised controlled trials was searched based on the following databases: PubMed, EMBASE, the Cochrane Library, OVID, Web of Knowledge, China National Knowledge Infrastructure (CNKI), Wanfang Data, VIP databases and SinoMed. The retrieval time dated back to 25 July 2018. Then we screened research literatures based on established inclusion and exclusion criteria. Quality evaluation for each included study was done using the Cochrane risk of bias and the Jadad scale. Results A total of 3334 articles were retrieved and 21 studies were included which contained 1503 subjects. In the 21 studies, 15 chose probiotics as interventions to regulate intestinal microbiota and six chose non-probiotic ways such as adjusting daily diets. Probiotic supplements in seven studies contained only one kind of probiotic, two studies used a product that contained two kinds of probiotics and the supplements used in the other five studies included at least three kinds of probiotics. In the studies that used treatment as usual plus interventions regulating intestinal flora (IRIF) as interventions (five studies), only non-probiotic ways were effective (two studies), which means 40% of studies were effective;in the studies that used IRIF alone (16 studies, 11 studies used probiotic ways and 5 studies used non-probiotic ways), 56% of studies could improve anxiety symptoms, and 80% of studies that conducted the non-probiotic interventions were effective, while 45% of studies that used

Evolving Models and Tools for Microglial Studies in the Central Nervous System

Microglia play multiple roles in such processes as brain development,homeostasis,and pathology.Due to their diverse mechanisms of functions,the complex sub-classifications,and the large differences between different species,especially compared with humans,very different or even opposite conclusions can be drawn from studies with different research models.The choice of appropriate research models and the associated tools are thus key ingredients of studies on microglia.Mice are the most commonly used animal models.In this review,we summarize in vitro and in vivo models of mouse and human-derived microglial research models,including microglial cell lines,primary microglia,induced microglia-like cells,transgenic mice,human-mouse chimeric models,and microglial replacement models.We also summarize recent developments in novel single-cell and in vivo imaging technologies.We hope our review can serve as an efficient reference for the future study of microglia.

In recent years,research on meditation as an important alternative therapy has developed rapidly and been widely applied in clinical medicine.Mechanism studies of meditation have also developed progressively,showing that meditation has great impact on brain structure and function,and epigenetic and telomere regulation.In line with this,the application of meditation has gradually been expanded to mental illness,most often applied for major depressive disorders and substance-related and addictive disorders.The focus of this paper is to illustrate the biological mechanisms of meditation and its application in mental disorders.

Current clinical management of major mental disorders,such as autism spectrum disorder,depression and schizophrenia,is less than optimal.Recent scientific advances have indicated that deficits in oxidative and inflammation systems are lessthanoptimal.Recentscientific advances have indicated extensively involved in the pathogenesis of these disorders. These findings have led to expanded considerations for treatment.Sulforaphane(SFN)is a dietary phytochemical extracted from cruciferous vegetables.It is an effective activator of the transcription factor nuclear erythroid-2 like factor-2,which can upregulate multiple antioxidants and protect neurons against various oxidative damages.On the other hand,it can also significantly reduce inflammatory response to pathological states and decrease the damage caused by the immune response via the nuclear factor-kB pathway and other pathways.In this review,we introduce the biological mechanisms of SFN and the pilot evidence from its clinical trials of major mental disorders,hoping to promote an increase in psychiatric clinical studies of SFN.

Major depressive disorder(MDD)is one of the most common and serious mental diseases.Its overall 12-month prevalence rate is〜6%,while the lifetime risk increases about twofold(15%-18%),which means almost one in five people will experience at least one depressive episode during their lifetime[1].MDD is characterized by sad emotion accompanied by anhedonia,lack of energy or appetite,sleep disturbance,and low self-evaluation.In severe cases,suicidal ideation/attemp^ehavior or self-injurious behavior may occur,which seriously affects patients'daily life and social function.In 2010,MDD was listed as the second greatest disease burden in the world,and is expected to rank first by 2030[2].

背景:元认知训练(metacognitive training,MCT)是一组全新的针对精神分裂症的心理治疗方法,但是到目前为止还没有确凿的证据证实其疗效。目的:采用meta分析评估MCT治疗精神分裂症的有效性。方法:使用电子检索和手工检索的方法筛查文献,从而识别出有关MCT治疗精神分裂症疗效的随机对照试验(randomized controlled trials,RCT),这些RCT都满足预先规定的纳入标准。本研究采用Cochrane风险评估偏倚工具评估各种偏倚的风险性,并使用Cochrane Review Manager 5.3版和R 3.1.1版进行数据合成。结果:54项不重复报告中的10项试验被纳入,但评估结果方法上的差异限制了可能被纳入meta分析的研究数量。综合分析了四项在试验结束时使用阳性和阴性症状量表(Positive and Negative Syndrome Scale,PANSS)的研究,发现MCT组比对照组有一个较小的降低,但具有统计学意义。但综合四项在试验结束时使用精神病症状评定量表(psychotic Symptom Rating Scales,PSYRATS)的错觉分量表的研究,发现两组间无显著差异。将那些无法汇集的各研究的其它结果进行定性的综合分析,显示出一些有利于MCT的趋势,但多数发现两组之间是没有差异的。结论:纳入的RCT试验数量有限、各试验结果评价的方法和时间的多样性、以及方法学上的问题使我们无法得出一个关于使用MCT治疗精神分裂症的有效性的结论。我们需要更多的使用标准化的结果评估方法、使用意向性治疗(intention-to-treat ITT)分析、以及在干预后对参与者进行有规律的定期随访的随机试验,从而确定MCT是否应该成为一个值得推荐的精神分裂症辅助治疗。
