Metal-organic frameworks （MOFs）,that have emerged as promising electrocatalysts in oxygen evolution reaction （OER）,usually undergo structural reconstruction to form metal hydroxides as active ***,it remains a great challenge to design and synthesize ideal MOF-based pre-catalysts for properly cognizing and utilizing the reconstruction process toward efficient OER ***,the well-defined MOF-808 is post-modified by 2-hydroxyphosphonoacetic acid （HPAA） to form a micro-adsorber that enables to capture of metal ions ***3+and Ni2+ions are synergistically anchored into the microadsorber,developing a Fe/Ni-containing pre-catalyst that can be dynamically electrooxidized to be an active and stable dual-site electrocatalyst denoted by R-MOF-HPAA@FeNi with abundant metal *** a result,R-MOF-HPAA@FeNi exhibits an excellent OER performance with a low overpotential of 272 mV and Tafel slope of 35.9 m V dec-1,as well as long-term stability over 300 h at 10 m A *** and theoretical calculations suggest the resulting catalyst mainly follows a lattice oxygen mechanism （LOM） of dual-metal-site mechanism （DMSM）.Owing to the chelating role of HPAA,the post-synthesized MOF-HPAA behaves as a trap to uniformly and firmly adhere Fe3+and Ni2+ions,leading to a functionalized MOF-based pre-catalyst which can be structurally evolved into atomic level-distributed OER electrocatalysts.

Laterality is a crucial physiological process intricately linked to the cilium-centrosome complex during embryo *** in the process can result in severe organ ***-coil domain containing 141(CCDC141)has been previously known as a centrosome-related gene,but its role in left-right(LR)asymmetry has not been *** this study,we utilize the zebrafish model and human exome analysis to elucidate the function of ccdc141 in laterality *** knockdown of ccdc141 in zebrafish disrupts early LR signaling pathways,cilia function,and Kupffer's vesicle *** ccdc141-knockdown embryos exhibiting aberrant LR patterns,ccdc141-null mutants show no apparent abnormality,suggesting a genetic compensation response *** parallel,we observe a marked reduction inα-tubulin acetylation levels in the ccdc141 *** treatment with histone deacetylase(HDAC)inhibitors,particularly the HDAC6 inhibitor,rescues the ccdc141 crispant ***,exome analysis of 70 patients with laterality defects reveals an increased burden of CCDC141 mutations,with in-vivo studies verifying the pathogenicity of the patient mutation *** findings highlight the critical role of ccdc141 in ciliogenesis and demonstrate that CCDC141 mutations lead to abnormal LR patterns,identifying it as a candidate gene for laterality defects.

Effects of fluid balance on prognosis of acute respiratory distress syndrome patients secondary to sepsis

BACKGROUND:Fluid management is crucial to acute respiratory distress syndrome(ARDS)secondary to ***,choices of fluid resuscitation strategies and fluid input volumes remain a thorny *** study aimed to elucidate the relationship between fluid balance and prognosis of ARDS patients secondary to ***:Our study included 322 sepsis patients from Ruijin Hospital between 2014 and 2018,and 84 patients were diagnosed as ARDS within 72 hours after onset of sepsis according to Berlin ARDS Defi ***:Among the 322 sepsis patients,84(26.1%)were complicated with ARDS within 72 *** patients had a lower oxygenation index(PaO2/FiO2166.4±71.0 vs.255.0±91.2,P<0.05),longer duration of mechanical ventilation(11[6-24]days vs.0[0-0]days,P<0.05)than those without *** patients with ARDS showed daily positive net fl uid balance during seven days compared with those without ARDS who showed daily negative net fluid balance since the second day with signifi cant statistical *** the 84 sepsis patients with ARDS,58(69.0%)*** daily fl uid input volumes were much lower in survivors than in non-survivors(43.2±16.7 mL/kg vs.51.0±25.2 mL/kg,P<0.05)while output volumes were much higher in survivors(45.2±19.8 mL/kg vs.40.2±22.7 mL/kg,P<0.05).Using binary logistic regression analysis,we found that the mean daily fl uid balance was independently associated with mortality of sepsis patients complicating with ARDS(P<0.05).CONCLUSIONS:Early negative fluid balance is independently associated with a better prognosis of sepsis patients complicated with ARDS.

Bactericidal/permeability-increasing protein originates in both the testis and the epididymis and localizes in mouse spermatozoa

Bactericidal/permeability-increasing protein （BPI） is an endogenous antibiotic protein with activity against gram-negative bacteria. In the present study, we examined the expression of BPI in postnatal mouse testes and epididymides as well as the subcellular localization within epididymal spermatozoa. Our results showed that, BPI mRNA was expressed in testis and epididymis independently. Throughout the epididymis, the BPI protein level gradually decreased in the epididymal epithelium in a spatial manner, specialized within the cytoplasm of clear cells in the cauda part. We detected BPI proteins in intact acrosome, implying its testicular origin; on the other hand, after the acrosome reaction, BPI proteins were observed dispersed across the entire sperm head, especially enriched at the equatorial segment. Our findings suggested a dual origin of the BPI that generated both in the testis and epididymis, and associated with mouse spermatozoa. BPI protein might be involved in the dynamics modification of the sperm plasma membrane and also the fertilization process.

Macrophages-mediated atherosclerosis(AS)is an inflammatory disease and the most common cause of *** the progress of basic and clinical research,anti-cytokine therapy has garnered considerable attention of the research community for the regulation of the inflammatory microenvironment for AS *** of their promising potential,primary clinical trials have revealed that anti-cytokine drugs exhibit poor selectivity and thus affect other parts of the immune system,especially during long-term *** circumvent these limitations,herein we exploited mesoporous silica nanoparticles(MSNs)with a pore size of 15.5 nm as carriers for the anti-interleukin-1β(anti-IL-1β)delivery to be the anti-cytokine *** vitro mechanistic studies indicated that the MSNs@anti-IL-1βcan regulate the macrophage-related inflammatory microenvironment,promote the viability of vascular endothelial cells(vECs),and reduce proliferation and phenotypic switching of vascular smooth muscle cells(vSMCs).In vivo evaluation further revealed that the MSNs@anti-IL-1βwere preferentially accumulated in macrophages,impeding the AS progress by maintaining the endothelium integrity and inhibiting the vSMCs ***,MSNs@antiIL-1βinduced neovascularization and improved hindlimb ischemia *** together,these MSNs affording the sustained release of anti-cytokine agents may have broad implications for the clinical management of the AS,including the reduction of the AS progression and alleviation of the ischemia.

This work presents an adaptive tracking guidance method for robotic fishes. The scheme enables robots to suppress external interference and eliminate motion jitter. An adaptive integral surge line-of-sight guidance rule is designed to eliminate dynamics interference and sideslip issues. Limited-time yaw and surge speed observers are reported to fit disturbance variables in the model. The approximation values can compensate for the system's control input and improve the robots' tracking ***, this work develops a terminal sliding mode controller and third-order differential processor to determine the rotational torque and reduce the robots' run jitter. Then, Lyapunov's theory proves the uniform ultimate boundedness of the proposed method. Simulation and physical experiments confirm that the technology improves the tracking error convergence speed and stability of robotic fishes.

Enhanced endosomal escape of dendrigraft poly-L-lysine polymers for the efficient gene therapy of breast cancer

Dendrimer,such as dendrigraft poly-L-lysine(DGL)polymers,with high surface charge density,well-defined structure,and narrow poly-dispersity is often employed as a gene vector,but its transfection efficiency is still partially inhibited due to poor endosomal escape ***,we used a surface modification strategy to enhance the endosomal escape ability of DGL polymers,and thus improved its gene transfection efficiency.A library of phenylboronic acid(PBA)modified DGL polymers(PBA-DGLs)was designed to screen efficient small interfering RNA(siRNA)*** lead candidate screened from the library shown a capability of inducing nearly 90% gene silencing in MDA-MB-231 *** study of the transfection mechanism revealed that PBA modification not only improves siRNA cellular uptake,but,more importantly,endows DGL polymers the ability of endosomal *** of the top candidates from polyplexes was further shielded with hyaluronic acid to construct targeted nanoparticles,and the yielding nanoparticles significantly suppressed the tumor growth in a breast cancer model by effective siRNA *** research provides a general and effective strategy to enhance the endosomal escape and transfection efficiency of dendrimer.

Localized surface plasmon resonance improves transdermal photodynamic therapy of hypertrophic scars

Photodynamic therapy(PDT)is an emerging therapeutic strategy for hypertrophic scars(HS),which is heavily dependent on reactive oxygen species(ROS)***,the unsatisfactory delivery and excitation of 5-aminolevulinic acid(ALA,a commercial photosensitizer in dermatology)result in an insufficient ROS generation,and thus limit the clinical application of PDT treating HS(HS-PDT).Consequently,sophisticated transdermal co-delivery nanoethosomes(named A/A-ES)with ALA and Au nanotriangles(AuNTs)in cores are prepared via an in-situ seed-mediated growth method,and then applied to improve HS-PDT through localized surface plasmon resonance(LSPR)-enhanced ROS generation.A/A-ES display a satisfactory performance in co-delivery in HS tissue with sufficient protoporphyrin IX production and LSPR effect in cytoplasm,which is beneficial for ALA excitation as well as ROS *** vitrolvivo studies reveal that A/A-ES significantly improve HS-PDT in promoting to fibroblast apoptosis and collagen remodeling through LSPR-enhanced ROS ***,this study provides a feasible strategy that integrates transdermal delivery and LSPR to enable the beneficial effects of HS-PDT through boosting the delivery and excitation of ALA.
