Herpes simplex virus typeⅠ induces an autophagic response and accelerates the fragmentation of apolipoproteins E protein

OBJECTIVE How infection of Herpes simplex virus typeⅠ(HSV-1) induces enhancement of *** The wild type HSV-1 strain Kos 1.1 was propagated in Vero cells and ***-N-SH cells seeded in DMEM/F12 were exposed to HSV-1 with 6 h or 12 h and multiplicity of infection(MOI) for 10 or 40 in each *** infectious titers of the HSV-1 samples were determined by plaque *** staining to test the number of autophagosome within the cell after infection with time and moi was indicated in each *** the molecular level,Western blotting and immunofluorescence analyses were done to study the expression of the proteins related to the cell *** mRNA transcribed from the gene related to autophagy was quantified by reverse transcription followed by real-time *** intranasal infection of different transgenic mice,immunoflurorence studies were done to detected the expression of Aβ42 and proteins related to autophagy from the brain *** water maze experiment was performed to test the change of spatial learning and memory between different transgenic *** SK-N-SH cell showed time-and moi-dependent increase of MDC positive staining after HSV-1 *** blotting analysis showed that LC3-Ⅱ was less in mock-infected cells but it was detected after 12 h from 10 to 40 moi HSV-1 infected *** level increased in a viral concentration-dependent *** agreement with the Western blotting results,direct fluorescence microscopy revealed that the signals of LC3 were consistent with their localization on autophagic compartments.P62,another protein related to autophagoysome formation,also increased with MOI.15 ku fragment of intracellular apolipoproteins E(APOE) protein increased after infection,but at the mRNA level it remained the *** expression of APP showed less decrease but intracellular Aβ42 increased significantly compared with the mock *** the brain,after intranasal infection for 7 d,autophagy re

电针后溪穴治疗急性腰扭伤临床随机对照试验Meta分析(英文)

目的:评价电针后溪穴治疗急性腰扭伤的疗效和安全性。方法:收集电针后溪穴治疗急性腰扭伤临床随机对照试验(R andomized controlled trial,RCT)。计算机检索P ub Med(1966-2014),EMBASE(1980-2014),Cochrane Library(2014年第1期),重庆维普数据库(1989-2014),万方数据库(1998-2014),中国生物医学文献数据库(1978-2014)和中国期刊全文数据库(1979-2014)。同时采用手工检索上海交通大学图书馆收录期刊收集相关的数据。所有收集的数据首先由两名研究者分别对每个纳入试验进行偏倚风险和质量评估,再使用R ev Man 5.2.0软件进行Meta分析。结果:共纳入6个研究,合计1288例患者。M eta分析结果显示电针后溪穴组在总有效率上优于尼美舒利药物组[RR=1.33;95%CI(1.19-1.49)]和莫比可药物组[RR=1.08;95%CI(1.03-1.14)],但与针刺腰痛点组[RR=1.09;95%CI(1.00-1.19)]和双氯芬酸钠组[RR=1.08;95%CI(0.96-1.21)]疗效比较差异无统计学意义。而在治愈率方面,电针后溪穴组优于莫比可组[RR=1.67;95%CI(1.45-1.92)]和尼美舒利组[RR=1.37;95%CI(1.15-1.62)],与针刺腰痛点组[RR=1.35;95%CI(0.99-1.84)]和双氯芬酸钠药物组[(RR=1.19;95%CI(0.88-1.61)]疗效差异无统计学意义。结论:截止至检索结束,几乎没有收集到评价电针后溪穴治疗急性腰扭伤临床疗效的大样本高质量的R CT文献,且大部分为中文文献。忽略其在临床试验中存在的固有的缺点,电针后溪穴仍然作为一个有效且具有安全性的辅助治疗方法用于治疗急性腰扭伤,但仍需更多的大样本R CT进行验证其在治疗急性腰扭伤的疗效。

Characteristics of Chinese population development from 1949 to 2018, and lessons learned

China’s population increased from 540 million in 1949 to 1.4 billion in ***-graphic transition provides a theoretical framework to understand China’s popula-tion changes,and we can divide the history of China’s population development into three periods:the period of mortality decline-driven demographic transition(prior to 1970),the period of fertility decline-driven demographic transition(1970-1999),and post-demographic transition period(after 2000).This paper provides a histori-cal overview of China’s population development,and concludes by identifying the main characteristics of this development and the main lessons learned from the pro-cess of population development.

Visual functional changes during acute elevation of intraocular pressure

Glaucoma is closely related to elevation of intraocular pressure （IOP）. Many studies have done on the effect of chronic elevation of lOP on the retina and optic nerve, but less attention was paid to the effect of acute elevated lOP. Here we briefly review experimental studies on functional changes of the visual system from the retina to the visual cortex under acute elevated lOP condition, which is similar to that of acute primary angle-closure glaucoma.

The efficient induction and long-term persistence of pathogen-specific memory CD8 T cells are pivotal to rapidly curb the *** studies indicated that long-noncoding RNAs expression is highly cell-and stage-specific during T cell development and differentiation,suggesting their potential roles in T cell ***,the key lncRNAs playing crucial roles in memory CD8 T cell establishment remain to be *** CD8 T cell subsets profiling of lncRNAs,this study found a key lncRNA-Snhgl with the conserved naivehl-effectorlo-memoryh,expression pattern in CD8 T cells of both mice and human,that can promote memory formation while impeding effector CD8 in acute viral ***,Snhgl was found interacting with the conserved vesicle trafficking protein Vps13D to promote IL-7Ra membrane location *** the deep mechanism probing,the results show Snhgl-Vps13D regulated IL-7 signaling with its dual effects in memory CD8 generation,which not just because of the sustaining role of STAT5-BCL-2 axis for memory survival,but more through the STAT3-TCF1-Blimp1 axis for transcriptional launch program of memory ***,we performed further study with finding a similar high-low-high expression pattern of human SNHG1A/PS13D/IL7R/TCF7 in CD8 T cell subsets from PBMC samples of the convalescent COVID-19 *** central role of Snhgl-Vps13D-IL-7R-TCF1 axis in memory CD8 establishment makes it a potential target for improving the vaccination effects to control the ongoing pandemic.

背景:元认知训练(metacognitive training,MCT)是一组全新的针对精神分裂症的心理治疗方法,但是到目前为止还没有确凿的证据证实其疗效。目的:采用meta分析评估MCT治疗精神分裂症的有效性。方法:使用电子检索和手工检索的方法筛查文献,从而识别出有关MCT治疗精神分裂症疗效的随机对照试验(randomized controlled trials,RCT),这些RCT都满足预先规定的纳入标准。本研究采用Cochrane风险评估偏倚工具评估各种偏倚的风险性,并使用Cochrane Review Manager 5.3版和R 3.1.1版进行数据合成。结果:54项不重复报告中的10项试验被纳入,但评估结果方法上的差异限制了可能被纳入meta分析的研究数量。综合分析了四项在试验结束时使用阳性和阴性症状量表(Positive and Negative Syndrome Scale,PANSS)的研究,发现MCT组比对照组有一个较小的降低,但具有统计学意义。但综合四项在试验结束时使用精神病症状评定量表(Psychotic Symptom Rating Scales,PSYRATS)的错觉分量表的研究,发现两组间无显著差异。将那些无法汇集的各研究的其它结果进行定性的综合分析,显示出一些有利于MCT的趋势,但多数发现两组之间是没有差异的。结论:纳入的RCT试验数量有限、各试验结果评价的方法和时间的多样性、以及方法学上的问题使我们无法得出一个关于使用MCT治疗精神分裂症的有效性的结论。我们需要更多的使用标准化的结果评估方法、使用意向性治疗(intention-to-treat ITT)分析、以及在干预后对参与者进行有规律的定期随访的随机试验,从而确定MCT是否应该成为一个值得推荐的精神分裂症辅助治疗。
