Global Profiles of Acetylated Proteins in Brains of Scrapie Agents 139A- and ME7-Infected Mice Collected at Mid-Early, Mid-Late, and Terminal Stages

Objective To describe the global profiles of acetylated proteins in the brains of scrapie agents 139Aand ME7-infected mice collected at mid-early,mid-late,and terminal stages.Methods The acetylated proteins from the cortex regions of scrapie agent(139A-and ME7)-infected mice collected at mid-early(80 days postinfection,dpi),mid-late(120 dpi),and terminal(180 dpi) stages were extracted,and the global profiles of brain acetylated proteins were assayed with proteomic mass spectrometry.The proteins in the infected mice showing 1.5-fold higher or lower levels than that of agematched normal controls were considered as differentially expressed acetylated peptides(DEAPs).Results A total of 118,42,and 51 DEAPs were found in the brains of 139A-80,139A-120,and 139A-180dpi mice,respectively.Meanwhile,390,227,and 75 DEAPs were detected in the brains of ME7-80,ME7-120,and ME7-180 dpi mice,respectively.The overwhelming majority of DEAPs in the mid-early stage were down-regulated,and more portions of DEAPs in the mid-late and late stages were up-regulated.Approximately 22.1%(328/1,485) of acetylated peptides mapped to 74 different proteins were mitochondrial associated.Kyoto Encyclopedia of Genes and Genomes pathway analysis identified 39(80dpi),13(120 dpi),and 10(180 dpi) significantly changed pathways in 139A-infected mice.Meanwhile,55,25,and 18 significantly changed pathways were observed in the 80,120,and 180 dpi samples of139A-and ME7-infected mice(P < 0.05),respectively.Six pathways were commonly involved in all tested samples.Moreover,many steps in the citrate cycle(tricarboxylic acid cycle) were affected,represented by down-regulated acetylation for relevant enzymes in the mid-early stage and upregulated acetylation in the mid-late and late stages.Conclusion Our data here illustrated the changes in the global profiles for brain acetylated proteins during prion infection,showing remarkably inhibited acetylation in the early stage and relatively enhanced acetylation in the late stage.

The study describes some of the major findings of changes in intracellular components and biological pathways in the brain during prion infection and hypothesizes some important physiological and pathological approaches mainly based on our studies.Omics techniques analysis of messenger RNA(mRNA)and proteins were carried out in the study.Meanwhile,Western blot,immunohistochemistry,and immunofluorescence were used for protein analysis in different signaling pathways.Statistical analyses were used to describe the protein differences in signaling pathways of infected and normal samples.This report reviewed and summarized our studies on the aberrant changes in intracellular components and biological functions in the brains of prion disease(PrD).Omics analyses proposed extensive abnormal alterations of brain mRNAs transcriptions,protein expressions,and post-translation modifications.The molecular disturbances for microtubule instability and depolymerization,the dysregulations of different signals related with neuron loss and synaptic plasticity,the abnormalities of mitochondrial and endoplasmic reticulum stress,and disturbance of intracellular reactive oxygen species homeostasis during prion infection were precisely analyzed and reviewed.Aberrant disturbances of numerous biological molecules and signals in brain tissues were found during prion infection.

Characteristics of Different Types of Prion diseases—China’s Surveillance

This report briefly described the establishment and implementation of national surveillance for human prion disease(PrD)in China.Reported cases came from Chinese surveillance network for PrD.Immunohistochemistry,Western blot,enzyme-linked immunosorbent assay(ELISA),Polymerase Chain Reaction(PCR),and real-time quaking-induced conversion(RT-QuIC)tests were used for the samples of brain,cerebrospinal fluid(CSF),and blood.Diagnosis standard for the PrDs is based on the National Commission of Health(WS/T 562-2017).The study summarized major epidemiological,clinical and laboratory features of more than 2,100 diagnosed different types of Chinese PrD cases.Sporadic Creutzfeldt-Jacob disease(sCJD)is the predominant type of PrD(88.7%).19 different genotypes of genetic PrDs(gPrDs)were identified,accounting for about 11.3%of all PrDs,revealing ethno-relationships.No iatrogenic CJD(iCJD)and variant CJD(vCJD)was identified.The characteristics of different types of sCJD in China showed similar features as those reported globally,but gPrDs showed an obvious ethnorelationship.

Unlike infectious diseases caused by conventional microbes,there are no detectable specific humoral or cellular immunoresponses to prion infection.However,extensive and active gliosis is observable in affected brain regions along with significant deposits of scrapielike prion protein(PrP^(Sc)).Here,we summarize our studies of vibrant activation of host non-specific immunocomponents and autophagy in the microenvironment of prion infected brains.Activation of the brain’s innate immunity and autophagy upon prion infection reflect non-specific host defense systems attempt to dispose of accumulated prions.Vibrant elevation of neuroinflammation leads to neuron injury.

Prion disease(PrD)or transmissible spongiform encephalopathy(TSE)is a group of transmissible and fatal neurodegenerative diseases affecting humans and many animal species.Clinically,PrD,either in humans or in animals,has been documented for more than hundreds of years,e.g.,scrapie in the early 18th century and human Creutzfeldt-Jakob disease(CJD)in the early 20th century(1–2).However,the hypothesis and conception of“prions”have been gradually accepted only since the 1980s.As an unconventional infectious agent without nucleic acids,the principle of the“prion”is unsolved conformational changes from host normal membrane protein PrPc to abnormal pathogenic scrapie-like prion protein(PrP^(Sc))((3).The hypothesis of the prion concept mostly explains the pathogenesis of PrD;however,there are still many gaps that need to be filled.More importantly,“prion theory”opens a completely new window in biology,possibly highlighting a new type of life.

Coxsackievirus A10(CVA10)is one of the major causative agents of hand,foot and mouth disease(HFMD).To investigate the epidemiological characteristics as well as genetic features of CVA10 currently circulating in shanghai,China,we collected a total of 9,952 sporadic HFMD cases from January 2016 to December 2020.In the past five years,CVA10 was the fourth prevalent causatives associated with HFMD in shanghai and the overall positive rate was 2.78%.The annual distribution experienced significant fluctuations over the past five years.In addition to entire VP1 sequencing,complete genome sequencing and recombination analysis of CVA10 isolates in shanghai were further performed.A total of 64 near complete genomes and 11 entire VP1 sequences in this study combined with reference sequences publicly available were integrated into phylogenetic analysis.The CVA10sequences in this study mainly belonged to genogroup C and presented 91%-100%nucleotide identity with other Chinese isolates based on VP1 region.For the first time,our study reported the appearance of CVA10 genogroup D in Chinese mainland,which had led to large-scale outbreaks in Europe previously.The recombination analysis showed the recombination break point located between 5,100 nt and 6,700 nt,which suggesting intertypic recombination with CVA16 genogroup D.To conclusion,CVA10 genogroup C was the predominant genogroup in shanghai during 2016-2020.CVA10 recombinant genogroup D was firstly reported in circulating in Chinese mainland.Continuous surveillance is needed to better understand the evolution relationships and transmission pathways of CVA10 to help to guide disease control and prevention.

Summary What is already known about this topic?An outbreak of coronavirus disease 2019(COVID-19)of Omicron BA.2 emerged in Jilin City since March 3,2022,which involved in 27,036 cases by April 12.The vaccination program with inactivated COVID-19 vaccines has been implemented since the beginning of 2021.What is added by this report?The incidences of moderate,severe,and critical cases in the whole population of the group of 0+1 dose were 1.82-,9.49-,and 3.85-fold higher than those in the group of 2 doses,and 5.03-,44.47-,and∞-fold higher than those received 3 doses vaccination.For the population≥60 years,the incidences of moderate,severe,and critical cases in the group of 0+1 dose were 29.92,9.62,and 4.27 per 100,000,showing 4.13-,43.72-,and 4.85-fold higher than 2 doses,as well as 13.28-,22.37-,and∞-fold higher than 3 doses.What are the implications for public health practice?The incidences of each type of COVID-19 in the population who were fully vaccinated or booster vaccinated in Jilin City were significantly lower than those who were unvaccinated and/or partially vaccinated.Booster vaccination with homologous inactivated vaccines induces stronger protectiveness for COVID-19 caused by variant of concern(VOC)Omicron.

Since early December 2019,the novel coronavirus has caused an outbreak of pneumonia that has claimed over 2000 lives,with more than 77,000 confirmed cases of infection by February 23,2020 in China.[1]The World Health Organization(WHO)recently named the disease caused by this new coronavirus as"coronavirus disease 2019(COVID-19)".The International Committee on Taxonomy of Viruses(ICTV)has named the new virus"severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)".[2]However,questions have been raised by different academic and professional bodies regarding whether the nomenclature is appropriate.
