Determination of Veterinary Drug Residues in Animal-derived Foods by Liquid Chromatography-Mass Spectrometry

[Objectives]This study was co.ducted to establish a rapid and effective method for simultaneous extraction of 54 kinds of veterinary drug residues in animal-derived food, including sulfonamides, quinolones, tetracyclines, malachite greens, penicillins, nitroimidazoles, tranquilizers and macrolides, by HPLC-MS. [Methods] The samples were extracted with 80% acetonitrile water(co.taining 0.1% formic acid), co.bined with QuEChERS extraction technology and C18 and PSA purification, analyzed by high performance liquid chromatography-mass spectrometry, and quantified by external standard method. The target substances were analyzed on ZORBAX Eclipse C18 chromatographic co.umn using 0.2% formic acid water and 0.2% methanol as mobile phases. The gradient elution mode was used for chromatographic separation and multiple reaction detection. [Results] In the linear range of 0.5-50.0 ng/ml, the linear relationship of the 54 kinds of veterinary drug residues was good, with co.relation co.fficients(r~2) greater than 0.995, and the detection limits ranged from 0.30 to 1.00 μg/kg. The results showed that the reco.ery ranged from 75.4% to 118.2% when different matrixes were added for reco.ery. [co.clusions] This method is simple, efficient, accurate, stable, and highly operable. It is applicable to simultaneous batch screening of veterinary drug residues in animal-derived food, and has high practical application value.

[Objectives]This study was co.ducted to establish an ultra-high performance liquid chromatography-tandem mass spectrometry for the rapid extraction of sodium pentachlorophenoxide from animal-derived food.[Methods]The samples were extracted with an acetonitrile water solution(8∶2),0.1 mol/L hydrochloric acid and a purification extraction bag with *** was performed to obtain supernatants,which were added to purification tubes co.taining PSA and C_(18) for purification,and then filtered with membranes for *** test solution was separated by a ZORBAX Eclipse plus C_(18) co.umn with acetonitrile and 5 mmol/L ammonium acetate as mobile phases,and determined with electrospray ionization and multiple reaction monitoring.[Results]The method had good linearity in the co.centration range of 1.0-50 ng/ml,and the co.relation co.fficient was *** limit of detection was 0.25μg/kg and the limit of quantification was 0.75μg/*** reco.ery was between 87.4%and 112.5%,and the RSD%was between 0.5%and 10.0%.[co.clusions]The method has simple operation and high sensitivity,and is suitable for trace detection of sodium pentachlorophenoxide in large quantities of animal-derived food.

Numbers of vertebrae is an important eco.omic trait associated with body size and meat productivity in ***,the genetic basis of vertebrae number in donkey remains to be well *** aim of this study was to identify candidate genes affecting the number of thoracic(TVn)and the number of lumbar vertebrae(LVn)in Dezhou donkey.A genome-wide association study was co.ducted using whole genome sequence data imputed from low-co.erage genome *** TVn,we identified 38 genome-wide significant and 64 suggestive SNPs,which relate to 7 genes(NLGN1,DCC,SLC26A7,TOX,WNT7A,LOC123286078,and LOC123280142).For LVn,we identified 9 genome-wide significant and 38 suggestive SNPs,which relate to 8 genes(GABBR2,FBXO4,LOC123277146,LOC123277359,BMP7,B3GAT1,EML2,and LRP5).The genes involve in the Wnt and TGF-βsignaling pathways and may play an important role in embryonic development or bone formation and co.ld be good candidate genes for TVn and LVn.

The objective of this study was to co.struct the reco.binant eukaryotic expression plasmid of ORF2 gene harboring enhanced green fluorescent protein （EGFP） report gene. ORF2 gene of porcine circo.irus type 2 cloned by PCR was ligated to the expression vector pEGFP-N1, which co.tains enhanced green fluorescent protein （EGFP） report gene, the reco.binant eukaryotic expression plasmid pEGFP-N1-ORF2 was co.structed successfully and was transfected into pre- pared duck myocardial cells （DMCs） by lipofectin. Acco.ding to the result, the fluorescence expression was directly detected with fluorescence microsco.e, and the expression of ORF2 were analyzed by RT-PCR and indirect immunofluorescence assay （IFA） respectively. About 48 h after transfection, green fluorescent can be observed on transfected cells ： T-PCR and IFA were positive. This indicated that ORF2 gene of PCV2 was expressed efficiently in transfected duck myocardial cells.

Euco.mia ulmoides, also called hardy rubber tree, is an eco.omically important tree; however, the lack of its genome sequence restricts the fundamental biological research and applied studies of this plant species. Here, we present a high-quality assembly of its ~l.2-Gb genome （scaffold N50 = 1.88 Mb） with at least 26 723 predicted genes for E. ulmoides, the first sequenced genome of the order Garryales, which was obtained using an integrated strategy co.bining Illumina sequencing, PacBio sequencing, and BioNano mapping. As a sister taxon to lamiids and campanulids, E. ulmoides underwent an ancient genome triplication shared by co.e eudico.s but no further whole-genome duplication in the last ~125 million years. E. ulmoides exhibits high expression levels and/or gene number expansion for multiple genes involved in stress responses and the biosynthesis of seco.dary metabolites, which may acco.nt for its co.siderable environmental adaptability. In co.trast to the rubber tree （Hevea brasiliensis）, which produces cis-polyisoprene, E. ulmoides has evolved to synthe- size long-chain trans-polyisoprene via farnesyl diphosphate synthases （FPSs）. Moreover, FPS and rub- ber elongation factor/small rubber particle protein gene families were expanded independently from the H. brasiliensis lineage. These results provide new insights into the biology of E. ulmoides and the origin of polyisoprene biosynthesis.

‘Lvyou No. 1＇,bred in Huasheng Agriculture Limited Liability co.pany of shandong Province,is a new cucumber F1 hybrid developed by crossing two inbred lines HS267014 and HS6201. It is an early-maturing variety with wide adaptability,high yield,and good co.mercial breeding. The plant has strong growth vigor and high resistance to white powder mildew. The first fruit sets on the 4 ^th-5^ th section,and the fruiting ratio of cucumber is 60%-80%. The cylindrical fruit is about 30-35 cm in length,3. 2-3. 5 cm in diameter with light green skin and glossy appearance. This plant has demonstrated adaptability in early-spring and late-autumn planting as well as spring and autumn tunnel planting.

Artificial intelligence-assisted automatic and index-based microbial single-cell sorting system for One-Cell-One-Tube

Identification,sorting,and sequencing of individual cells directly from in situ samples have great potential for in-depth analysis of the structure and function of *** this work,based on an artificial intelligence(AI)-assisted object detection model for cell phenotype screening and a cross-interface co.tact method for single-cell exporting,we developed an automatic and index-based system called EasySort AUTO,where individual microbial cells are sorted and then packaged in a microdroplet and automatically exported in a precisely indexed,“One-Cell-One-Tube”*** target cell is automatically identified based on an AI-assisted object detection model and then mobilized via an optical tweezer for ***,a crossinterface co.tact microfluidic printing method that we developed enables the automated transfer of cells from the chip to the tube,which leads to co.pling with subsequent single-cell culture or *** efficiency of the system for single-cell printing is>93%.The throughput of the system for single-cell printing is~120 cells/***,>80%of single cells of both yeast and Escherichia co.i are culturable,suggesting the superior preservation of cell viability during ***,AI-assisted object detection supports automated sorting of target cells with high accuracy from mixed yeast samples,which was validated by downstream single-cell proliferation *** automation,index maintenance,and vitality preservation of EasySort AUTO suggest its excellent application potential for single-cell sorting.
