Cloning and expression analysis of the FvNCED3 gene and its promoter from ash(Fraxinus velutina)

The 9-cis-epoxycarotenoid dioxygenase(NCED)gene is rate-limiting in abscisic acid(ABA) *** this study, an NCED gene, designated FvNCED3(KY008746), was cloned from velvet ash(Fraxinus velutina Torr.) with a RACE method. The full length c DNA of FvNCED3 encodes a 573-amino acid *** analysis showed that the FvNCED3 protein was highly homologous to other NCED proteins. The expression patterns of FvNCED3 in different ash organs were analyzed by real-time PCR which revealed that FvNCED3 expression levels were highest in leaves and lowest in roots. The gene expression patterns of FvNCED3 under abiotic stress indicated that its expression increased under drought, salt and ABA stress and decreased due to high and low temperatures. There were no obvious changes under ultraviolet light. The 1094-bp upstream sequence 5' flank regulation region of the FvNCED3 gene was also cloned from ash using the Genome Walking method. To assess the activity of the FvNCED3 promoter, a p FvNCED3 p::GUS plant expression vector was constructed for tobacco transformation. GUS expression of the FvNCED3 GUS enzyme activity was detected in almost all transgenic tobacco tissues, especially in the young leaves,stigma, anther, ovule and ovary. After treating the transgenic tobacco with NaCl and placing it under drought stress, GUS staining of tobacco leaves increased compared with that under normal growth conditions. This result indicates that gene expression driven by the FvNCED3 promoter can be induced by salt and drought stress.

Multiple natural and human factors in estuarine wetlands result in complicated land surface characteristics with distinct spatial and temporal heterogeneities,thereby contributing to the difficulty in identifying spatiotemporal variations and influencing factors of plant diversity.A unique estuarine wetland gradient system(UEWGS)consisting of soil,vegetation,heat,distance,landscape,and anthropogenic gradients was established based on the ecological features of estuarine wetland through remote sensing and field investigation *** resolved the complicated land surface characteristics,covered all aspects of factors influencing plant diversity,and possessed distinct spatiotemporal *** yellow river delta,the largest estuarine wetland in the northern China,was selected as the study area to demonstrate UEWGS in four seasons in 2017.A total of 123 species were recorded with considerable seasonal *** australis,Suaeda salsa,and Tamarix chinensis were the dominant species,and crop species also played important *** single effect,all aspects of gradients exerted significant influences,yet only vegetation gradient possessed significant influences in all *** comprehensive effect,soil,vegetation,heat,and distance gradients showed significant gross *** content in soil gradient and net primary productivity in vegetation gradient possessed significant net influences in all seasons and can be considered as the main driving factor and indicator,respectively,of plant *** results validated the significance of UEWGS in revealing the plant diversity spatiotemporal characteristics and influencing factors,and UEWGS possessed universal applicability in the spatiotemporal analysis of plant diversity in estuarine areas.

Soil inorganic carbon(SIC),including mainly carbonate,is a key component of terrestrial soil C *** microorganisms can assimilate carbonate as the main or unique C source,how microorganisms convert SIC to soil organic carbon(SOC)remains unclear.A systematic field survey(n=94)was performed to evaluate the shift in soil C components(i.e.,SIC,SOC,and microbial residues)along a natural salinity gradient(ranging from 0.5‰to 19‰),and further to explore how microbial necromass as an indicator converting SIC into SOC in the yellow river *** observed that SIC levels linearly decreased with increasing salinity,ranging from~12 g kg^(-1)(salinity6‰).Additionally,the concentrations of SOC and microbial residues exponentially decreased from salinity6‰,with the decline of 39%and 70%,*** residues and SOC was tightly related to the variations in *** structural equation model showed the causality on explanation of SOC variations with SIC through microbial residues,which can contribute 89% of the variance in SOC storage combined with *** together,these two statistical analyses can support that microbial residues can serve as an indicator of SIC transition to *** study highlights the regulation of microbial residues in SIC cycling,enhancing the role of SIC playing in C biogeochemical cycles and enriching organic C reservoirs in coastal saline soils.

To test the patterns of the root morphology and architecture indexes of Tamarix chinensis in response to water and salt changes in the two media of the groundwater and soil,three-year-old *** seedlings were chosen as the research *** with four salinity levels was created,and three groundwater level(GL)were applied for each salinity treatment to measure the root growth and architecture *** the fresh water and brackish water treatments,the topological index(TI)of the *** roots was close to 0.5,and the root architecture was close to a dichotomous branching *** the saline water and saltwater treatments,the TI of the *** roots was large and close to 1.0,and the root architecture was close to a herringbone-like branching *** different GLs and salinities,the total root length was significantly greater than the internal link length,the external link length was greater than the internal link length,and the root system showed an outward expansion *** treatment with fresh water and a GL of 1.5 m was the most suitable for *** root growth,while the root growth of *** was the worst in the treatment with saline water and a GL of 0.3 *** can adapt to the changes in soil water and salt by regulating the growth and morphological characteristics of the root *** can adapt to high-salt environments by reducing its root branching and to water deficiencies by expanding the distribution and absorption area of the root system.

Sargassum horneri is a common brown macro-alga that is found in the inter-tidal ecosystems of China. To investigate the current status of seaweed resources and provide basic data for its sustainable development, ISSR (inter simple sequence repeat) and SRAP (sequence related amplified polymorphism) markers were used to analyze the population genetics among nine natural populations of S. horneri. The nine studied populations were distributed over 2 000 km from northeast to south China. The percentage of polymorphic loci P% (ISSR, 99.44%; SRAP, 100.00%), Nei's genetic diversity H (ISSR, 0.107-0.199; SRAP, 0.100-0.153), and Shannon's information index I (ISSR, 0.157-0.291; SRAP, 0.148-0.219) indicated a fair amount of genetic variability among the nine populations. Moreover, the high degree of gene differentiation G st (ISSR, 0.654; SRAP, 0.718) and low gene flow N m (ISSR, 0.265; SRAP, 0.196) implied that there was significant among-population differentiation, possibly as a result of habitat fragmentation. The matrices of genetic distances and fixation indices (F st ) among the populations correlated well with their geographical distribution (Mantel test R=0.541 5, 0.541 8; P=0.005 0, 0.002 0 and R=0.728 6, 0.641 2; P=0.001 0, 0.001 0, respectively); the Rongcheng population in the shandong peninsula was the only exception. Overall, the genetic differentiation agreed with the geographic isolation. The fair amount of genetic diversity that was revealed in the S. horneri populations in China indicated that the seaweed resources had not been seriously affected by external factors.

Increasing use of pyrethroid insecticides has resulted in concerns regarding potential effects on human health and ecosystems. Cypermethrin and its metabolite 3-phenoxybenzoic acid(PBA) have exerted adverse biological impacts on the environment;therefore,it is critically important to develop different methods to enhance their degradation. In this study,incubation experiments were conducted using samples of an Aquic Inceptisol supplied with nitrogen(N) in the form of NH4NO3 at different levels to investigate the effect of nitrogen on the degradation of cypermethrin and PBA in soil. The results indicated that appropriate N application can promote the degradation of cypermethrin and PBA in soil. The maximum degradation rates were 80.0% for cypermethrin after 14 days of incubation in the treatment with N at a rate of 122.1 kg ha-1 and 41.0% for PBA after 60 days of incubation in the treatment with N at a rate of 182.7 kg ha-1. The corresponding rates in the treatments without nitrogen were 62.7% for cypermethrin and 27.8% for PBA. However,oversupplying N significantly reduced degradation of these compounds. Enhancement of degradation could be explained by the stimulation of microbial activity after the addition of N. In particular,dehydrogenase activities in the soil generally increased with the addition of N,except in the soil where N was applied at the highest level. The lower degradation rate measured in the treatment with an oversupply of N may be attributed to the microbial metabolism shifts induced by high N.

NiMZn/C@melamine sponge-derived carbon(MSDC)composites(M=Co,Fe,and Mn)were prepared by a vacuum pump-ing solution method followed by carbonization.A large number of carbon nanotubes(CNTs)homogeneously attached to the surfaces of the three-dimensional cross-linked of the sponge-derived carbon in the NiCoZn/C@MSDC composite,and CNTs were detected in the NiFeZn/C@MSDC and NiMnZn/C@MSDC ***_(3)ZnC_(0.7),Ni_(3)Fe,and MnO in-situ formed in the NiFeZn/C@MSDC and NiMnZn/C@MSDC *** CNTs in the NiCoZn/C@MSDC composite efficiently modulated its complex ***,the composite exhibited the best performance among the composites,with the minimum reflection loss(RL_(min))of-33.1 dB at 18 GHz and thickness of 1.4 *** bandwidth for RL of≤-10 dB was up to 5.04 GHz at the thickness of 1.7 mm and loading of 25wt%.The op-timized impedance matching,enhanced interfacial and dipole polarization,remarkable conduction loss,and multiple reflections and scat-tering of the incident microwaves improved the microwave absorption *** effects of Co,Ni,and Fe on the phase and mor-phology provided an alternative way for developing highly efficient and broadband microwave absorbers.

Soil moisture is a major limiting factor for plant growth on shell ridge islands in the yellow river delta. However, it is difficult to carry out situ experiment to study dominant plant photosynthesis physiological on the shell ridge islands under extreme soil water stress. To evaluate the adaptability of plants to light and moisture variations under extreme soil moisture conditions present on these islands, we measured photosynthetic gas exchange process, chlorophyll fluorescence, and stem sap flow variables for 3-year-old trees of Tamarix chinensis Lour, a restoration species on these islands, subjected to three types of soil water levels： waterlogging stress （WS）, alternating dry-wet （WD）, and severe drought stress （SS） to inform decisions on its planting and management on shell ridge islands. Gas exchange, chlorophyll fluorescence, and stem sap flow in T. chinensis were then measured. Net photosynthetic rate （PN）, transpiration rate （E）, and water use efficiency （WUE） were similar under WS and alternating dry-wet conditions, but their mean E and WUE differed significantly （P 〈 0.05）. Under SS, the PN, E and WErE of T. chinensis leaves varied slightly, and mean PN, E and WUE were all low. Apparent quantum efficiency （AQY）, light compensation point （LCP）, light saturation point （LSP）, and maximum net photosynthetic rate （PNmax） of leaves were not significantly different （P 〉 0.05） under WS and dry-wet conditions; however, under extreme drought stress, compared with the dry-wet conditions, LCP was higher, Lsp was lower, and AQy and PNmax were both at the lowest level. Therefore, drought stress weakened light adaptability of leaves, and the efficiency of light transformation was poorer. （3） Maximum photochemical efficiency （Fv/Fm） and the actual photochemical efficiency （ΦPSII） were similar under waterlogged stress and dry-wet conditions, indicating a similar healthy photosynthetic apparatus and photosynthetic reaction cen- ter activity, respectively.
