Objective To establish a sensitive,simple and rapid detection method for African swine fever virus(ASFV)B646L *** A recombinase-aided amplification-lateral flow dipstick(RAA-LFD)assay was developed in this ***-aided amplification(RAA)is used to amplify template DNA,and lateral flow dipstick(LFD)is used to interpret the results after the amplification is *** lower limits of detection and specificity of the RAA assay were verified using recombinant plasmid and pathogenic nucleic *** addition,30 clinical samples were tested to evaluate the performance of the RAA *** The RAA-LFD assay was completed within 15 min at 37°C,including 10 min for nucleic acid amplification and 5 minutes for LFD reading *** detection limit of this assay was found to be 200 copies per *** there was no cross-reactivity with other swine *** A highly sensitive,specific,and simple RAA-LFD method was developed for the rapid detection of the ASFV.

An online survey conducted March 18–19,2020 on the official China CDC WeChat account platform was used to evaluate the effect of public education about masks usage during the new coronavirus disease 2019(COVID-19)*** nationals older than 18 were eligible for the *** survey collected 5,761 questionnaires from the 31 provinces,municipalities,and autonomous regions of China's Mainland.99.7%and 97.2%of the respondents answered correctly that respiratory droplets and direct contact were the main transmission routes.73.3%of the respondents considered COVID-19 to be‘serious’or‘very serious’.

H9N2 avian influenza virus(AIV) has widely circulated in poultry worldwide and sporadic infections in humans and mammals. During our surveillance of chicken from 2019 to 2021 in shandong Province, China, we isolated 11 H9N2AIVs. Phylogenetic analyses showed that the eight gene segments of the 11 isolates were closely related to several sublineages of Eurasian lineage: BJ/94-like clades(HA and NA genes), G1-like clades(PB2 and M genes), and SH/F/98-like clades(PB1, PA, NP and NS genes). The isolates showed mutation sites that preferentially bind to humanlike receptors(HA) and mammalian fitness sites(PB2, PB1 and PA), as well as mutations in antigen and drug resistance sites. Moreover, studies with mice revealed four isolates with varying levels of pathogenicity. The average antibody titer of the H9N2 AIVs was 8.60 log2. Based on our results, the epidemiological surveillance of H9N2 AIVs should be strengthened.

Objective To study the polymorphism in P66 and its human B-cell epitopes of Borrelia burgdorferi strains in *** Polymerase chain reaction(PCR)and sequencing were used to obtain the P66 sequences of59 Chinese *** the sequences were analyzed by MEGA 5.10 software and compared with the human B-cell epitope sequences from the Immune Epitope Database(IEDB)based on the reference strain of each *** Results showed that genetic and amino acid diversity presented in the 66 kD protein of all 59 Chinese strains,especially in Borrelia garinii(B.g)and Borrelia afzelii(B.a)strains.B.g strains were divided into three subclusters and two scattered strains JC1-7 and JC2-2 according to the amino acid sequences of *** P66 sequences of 15 Xinjiang strains represented by XI91-12 in the B.g subcluster1,changed from CAA to TAA codon at 508 aa position,resulting in early *** A and C were inserted at sequence position 1523 bp of strains FP1,LB20,LB21,and SZ21 in the B.a genotype,which resulted to early termination at position 511 aa.G base was inserted at 438 bp of LIP94-11 strain,which led to early termination at position 172 *** In P66 of 59 Chinese strains,polymorphisms were widely *** importantly,the P66 amino acid sequences of B.g strains had a certain regional *** of the characteristics of Xinjiang B.g isolates might be the variation at the 508 aa location in 15 Xinjiang B.g strains,which may be related to the strains’pathogenicity in this area.

Adipogenesis is the healthy expansion of white adipose tissue (WAT),serving as a compensatory response to maintain metabolic homeostasis in the presence of excess energy in the ***,the identification of novel regulatory molecules in adipogenesis,specifically membrane receptors such as G protein-coupled receptors (GPCRs),holds significant clinical *** receptors can serve as viable targets for pharmaceuticals,offering potential for restoring metabolic homeostasis in individuals with *** utilized trajectory inference methods to analyze three distinct single-nucleus sequencing (sNuc-seq) datasets of adipose tissue and systematically identified GPCRs with the potential to regulate *** verification in primary adipose progenitor cells (APCs) of mice,we discovered that ADGRD1 promoted the differentiation of APCs,while GPR39 inhibits this *** the obese mouse model induced by a high-fat diet(HFD),both gain-of-function and loss-of-function studies validated that ADGRD1 promoted adipogenesis,thereby improving metabolic homeostasis,while GPR39 inhibited adipogenesis,leading to metabolic ***,through the analysis of 2,400 ChIP-seq data and 1,204 bulk RNA-seq data,we found that the transcription factors (TFs) MEF2D and TCF12 regulated the expression of ADGRD1and GPR39,*** study revealed the regulatory role of GPCRs in adipogenesis,providing novel targets for clinical intervention of metabolic dysfunction in obese patients.

Introduction:Salmonella is a key intestinal pathogen of foodborne disease,and the plasmids in Salmonella are related to many biological characteristics,including virulence and drug resistance.A large number of plasmid contigs have been sequenced in bacterial draft genomes,however,these are often difficult to distinguish from chromosomal ***:In this study,three different customized Kraken databases were used to build three different Kraken *** genome benchmark datasets and simulated draft genome benchmark datasets were ***-fold cross-validation was used to evaluate the performance of the three different Kraken classifiers by two benchmark ***:The predictive performance of the classifier based on all National Center for Biotechnology Information plasmids and Salmonella complete genomes was *** optimal Kraken classifier was performed with Salmonella isolated in *** plasmid carrying rate of Salmonella in China is 91.01%,and it was found that the Kraken classifier could find more plasmid contigs and antibiotic resistance genes(ARGs)than results derived from a plasmid replicon-based method(PlasmidFinder).Moreover,it was found that in the strains carrying ARGs,plasmids carried more ARGs[three,95%confidence interval(CI):1–14]than chromosomes(one,95%CI:1–7).Discussion:We found building a high-quality customized database as a Kraken classifier to be ideal for the prediction of Salmonella plasmid sequences from bacterial draft *** the future,the Kraken classifier established in this study will play a significant role in ARG monitoring.

Epstein-Barr virus (EBV) infection is closely associated with nasopharyngeal carcinoma (NPC)and considered one of the major risk factors[1]. A limited number of viral proteins, such as latent membrane proteins (LMP1, LMP2) and EB nuclear antigen1 (EBNA1), are expressed in NPC cells[2].Recognition epitopes for CD8+and CD4+T cells were included in the LMP2 antigen and EBNA1 C-terminal region (amino acid No. 380-641), respectively[3,4].Both CD4+and CD8+memory T-cell responses were efficiently reactivated by EBNA1 and LMP2[5;6].
